Pinnatoxins (PnTXs) are emerging neurotoxins that were discovered about 30 years ago. They are solely produced by the marine dinoflagellate Vulcanodinium rugosum, and may be transferred into the food chain, as they have been found in various marine invertebrates, including bivalves. No human intoxication has been reported to date although acute toxicity was induced by PnTxs in rodents. LD50 values have been estimated for the different PnTXs through the oral route. At sublethal doses, all symptoms are reversible, and no neurological sequelae are visible. These symptoms are consistent with impai

This commentary is based upon the author's lecture given as the 2010 recipient of the award named in honor of Drs. Joseph V. The commentary provides an overview of how advances utilizing animal and human models of dependence and withdrawal came to inform public health policy and more recently, tobacco product regulation. The commentary also recounts how efforts by the to …

The nicotinic acetylcholine receptor (nAChR) antagonist alpha-bungarotoxin (alpha-Btx) binds to two different classes of high affinity binding sites from the Drosophila central nervous system. We have used the bivalent reagent 1-ethyl-3-[3-(dimethylamino)propyl]carbodiimide (EDAC) to cross-link 125I-alpha-Btx (M(r) = 8 kDa) to Drosophila head membranes. Upon sodium dodecylsulphate polyacrylamide gel electrophoresis (SDS-PAGE), one major adduct of M(r) approximately 50 kDa was identified, suggesting that a 42 kDa polypeptide binds the toxin. Adduct formation was inhibited by other cholinergic l

We have studied antibodies to the acetylcholine receptor (AChR) that inhibit alpha-bungarotoxin binding in 22 mothers with myasthenia gravis (MG) and in their 23 newborns. Of the 13 showing neonatal MG, seven of the mothers had detectable direct blocking antibodies, all 13 had decamethonium-dependent (DC) blocking, and nine had high titres of precipitating antibodies (greater than 40 nM). In those with symptom-free newborns, the corresponding figures were 2/10, 8/10 and 2/10; the mean titres of DC blocking and of precipitating antibodies were 5- and 3-fold lower than in the mothers of affected

BACKGROUND: Posterior cervical fusion (PCF) with lateral mass screws is a favorable treatment option to revise a symptomatic pseudarthrosis … e following risk factors: age, sex, number of levels revised, body mass index (BMI), and history of nicotine use.
RESULTS: The average age of patients at the time of PCF revision was 55 ± 11 years and 63% were female. The average BMI was 29 ± 6 kg/m2 and 19% reported a history of nicotine use. Postoperative follow-up visits were available with a median of …

BACKGROUND: Most smokers are ambivalent about quitting-they want to quit someday, but not now. Interventions are needed that can engage ambi … nce-based, best-practice smoking cessation advice and resources, including the ability to earn free nicotine patches. OBJECTIVES: To investigate the relationship between purchasing loose cigarettes and adolescent smoking habits in Indonesia.
DESIGN AND SETTING: This study employed a mixed-methods sequential explanatory design. We analysed the secondary data from a national survey, the 2019 Global Youth Tobacco Survey, using multivariable logistic regression models to examine the association between loose cigarette purchase and smoking frequency and intensity and nicotine dependence. Based on the quantitative findings, we arranged focus group discussions (FGDs) in junior and senior high schools located in Sou…

Pesticides are important resources in the control of pests and pathogens of plants and animals. Unfortunately, there are concerns around their broad impacts on non-target organisms and the environment. In this study we optimize a platform biased for the nicotinic acetylcholine receptor EAT-2 , a physiological regulator of feeding in C. elegans . We show that by screening for chemicals that inhibit C. elegans pharyngeal pumping in lev-1 ( x427 ) , we can identify lead compounds that modulate EAT-2 function. This provides a motivation for further studies to investigate the role of EAT-2 in paras

Nicotinamide, also known as niacinamide, is the amide form of vitamin B3. It is a precursor of essential coenzymes for numerous reactions in the body including adenosine triphosphate (ATP) production. Nicotinic acid, also known as niacin, is converted into nicotinamide in the body. The use of topical nicotinamide in the treatment of acne vulgaris; melasma; atopic dermatitis; rosacea; and oral nicotinamide in preventing nonmelanoma skin cancer is discussed. The possible side effects and consequences of excessive nicotinamide exposure are reviewed, including suggestions nicotinamide might have a

Myasthenia gravis is a chronic autoimmune disease characterized by a reduction of postsynaptic nicotinic acetylcholine receptors at the neuromuscular junction. Most myasthenia gravis patients require thymectomy. Intravenous (IV) anesthetics may be superior to inhalation agents in these patients. The Laryngeal Mask Airway (LMA), when compared with the endotracheal tube, causes less airway resistance, which in turn may lead to a decreased bronchoconstrictive reflex, less atelectasis, and fewer pulmonary infections. We report 5 patients with myasthenia gravis, who underwent transsternal thymectom

We studied a young man with persistent elevation of transaminases and indirect bilirubin following acute viral hepatitis. Liver biopsy showed chronic persistent hepatitis, and clinical evaluation was otherwise negative for significant liver disease. The indirect-reacting serum bilirubin rose after both caloric restriction and intravenous nicotinic acid, responses believed by some to be characteristic of Gilbert's syndrome. Fasting and postprandial serum bile acid conjugates were not elevated. These results suggest that "Gilbert's-like" aberrations in bilirubin metabolism may be part of the spe

BACKGROUND: Cholinergic neuromuscular transmission is central to gastrointestinal (GI) motility and is traditionally attributed to calcium-dependent, vesicular acetylcholine (ACh) release from enteric neurons. However, non-quantal, calcium-independent mechanisms-possibly involving transporter-mediated ACh efflux-may also contribute to cholinergic signaling.
AIM: To investigate both classical and alternative mechanisms of ACh release in the colon, focusing on the potential role of non-vesicular, transporter-dependent pathways in modulating smooth muscle contractility.
METHODS: Experiments wer

β-Nicotinamide mononucleotide (NMN) is an important intermediate in nicotinamide adenine dinucleotide (NAD+) metabolism and has attracted increasing interest as a bioactive compound and biomanufacturing product. In this study, Pseudomonas putida KT2440 was engineered for NMN production using a "block-enhance-transport" strategy. Deletion of nicB impaired nicotinic acid degradation and resulted in the accumulation of 66.2 μM nicotinic acid in cell extracts, whereas additional deletion of the putative NMN-consuming genes pncC and ushA did not lead to detectable NMN accumulation. Coexpression of
