Chloroquine differentially modulates coronary vasodilation in control and diabetic mice.

BACKGROUND AND PURPOSE: Chloroquine is a traditional medicine to treat malaria. There is increasing evidence that chloroquine not only induces phagocytosis but regulates vascular tone. Few reports investigating the effect of chloroquine on vascular responsiveness of coronary arteries have been made. In this study, we examined how chloroquine affected endothelium-dependent relaxation in coronary arteries under normal and diabetic conditions. EXPERIMENTAL APPROACH: We isolated coronary arteries from mice and examined endothelium-dependent relaxation (EDR). Human coronary endothelial cells and m

Activation of nuclear factor kappaB by nitric oxide in rat striatal neurones: differential inhibition of the p50 and p65 subunits by dexamethasone.

Nitric oxide (NO), an intercellular messenger in the brain, has been implicated in both neuronal plasticity and neurotoxicity. It has been suggested that NO can activate the DNA binding activity of nuclear factor kappaB (NF-kappaB) family proteins in some cell types while having an inhibitory effect in others. In this study we have investigated the effect of acute NO in primary neuronal cultures of rat striatum using immunohistochemistry. Exposure of neurones to the NO-mimetic S-nitroso-n-acetylpenicillamine (SNAP; 200 microM) and to bacterial lipopolysaccharide (LPS; 10 microg/ml) for 30 min

Protective effects of melatonin on mouse ovarian tissue against oxidative stress and ferroptosis may involve the MT1 receptor.

Ovarian cryopreservation is an important method for fertility preservation, with oxidative stress and ferroptosis being key contributors to cryopreservation-induced ovarian damage. Previous studies have shown that melatonin (MLT) protects vitrified-warmed mouse ovarian tissues (OTs), but the underlying mechanism remains unclear. This study investigated whether MLT inhibits oxidative stress and ferroptosis via MT1/MT2 receptors. The cryopreservation/thawing medium was supplemented with 0 mM MLT, 0.1 mM MLT, 0.1 mM MLT + Luzindole (a non-selective MT1/MT2 antagonist), or 0.1 mM MLT + 4P-PDOT (a

Phenotype consequences of myophosphorylase dysfunction: insights from the McArdle mouse model.

KEY POINTS: This is the first study to analyse the effect of muscle glycogen phosphorylase depletion in metabolically different muscle types. In McArdle mice, muscle glycogen phosphorylase is absent in both oxidative and glycolytic muscles. In McArdle mice, the glycogen debranching enzyme (catabolic) is increased in oxidative muscles, whereas the glycogen branching enzyme (anabolic) is increased in glycolytic muscles. In McArdle mice, total glycogen synthase is decreased in both oxidative and glycolytic muscles, whereas the phosphorylated inactive form of the enzyme is increased in both oxidat

Vascular oxidant stress early after balloon injury: evidence for increased NAD(P)H oxidoreductase activity.

Available evidence for oxidative stress after angioplasty is indirect or ambiguous. We sought to characterize the pattern, time course, and possible sources of free radical generation early after arterial balloon injury. Ex vivo injury performed in arterial rings in buffer with lucigenin yielded a massive oxygen-dependent peak of luminescence that decayed exponentially and was proportional to the degree of injury. Signals for injured vs. control arteries were 207. 1 +/- 17.9 (n = 13) vs 4.1 +/- 0.7 (n = 22) cpm x 10(3)/mg/min (p <. 001). Data obtained with 0.25 mmol/l lucigenin were validated

Redirecting radical phosphonylation via energy-transfer-enabled diradical rearrangement.

Radical phosphonylation has traditionally relied on single-electron transfer mechanisms. In contrast, strategies using energy-transfer pathways to enable scaffold rearrangement are rare. Here, we report an energy transfer catalysis strategy that diverts reactivity toward a previously unexplored pathway: a direct photochemical diradical rearrangement between homoallylic alcohols and phosphorus(III) reagents. Mechanistic studies, supported by computational calculation, provide definitive support for an energy-transfer-mediated diradical mechanism. This operationally simple and mild process, driv

Optimised Gynura divaricata polysaccharide extract ameliorated type 2 diabetes in mice by activating the PI3K/Akt/GLUT-4 signalling pathway.

Type 2 diabetes mellitus (T2DM) is a long-term metabolic disorder, which is characterised by insulin resistance and persistent hyperglycaemia. Natural polysaccharides exhibit promising hypoglycaemic potential; however, optimising their extraction and mechanistic understanding of polysaccharides from Gynura divaricata (GDP) remain limited. Here, GDP underwent hot-water extraction followed by ethanol precipitation. A response surface methodology known as Box-Behnken design was utilised to refine the extraction process (extraction time, 120 min; temperature, 90 °C; solid-to-liquid ratio, 1:20 [g/

Early retinal synaptic dysfunction and proteomic remodeling precede neurodegeneration in a Parkinson's disease model.

Parkinson's disease (PD) affects motor and non-motor systems; however, retinal changes and their molecular basis are not well understood. Using a transgenic mouse model overexpressing A53T-mutant human α-synuclein, we examined retinal function, structure, and proteomics at 6- and 16 months. Early retinal dysfunction was detected by a reduction in scotopic oscillatory potential amplitudes on electroretinography. Optical coherence tomography showed early thinning of the retinal nerve fiber layer/ganglion cell layer, and photoreceptor layer, accompanied by thickening of the inner plexiform layer.

Preparation and characterization of modified sunflower meal protein-quercetin nanoparticles: Shelf-life study of seasoned lamb kebabs.

This study developed a quercetin (Que) nanoencapsulation system (PU-SMP-Que) using phosphorylated and ultrasonicated sunflower meal protein (PU-SMP) as the carrier. At a protein-polyphenol mass ratio of 15:1, the spherical PU-SMP-Que nanoparticles exhibited an average diameter of 175.3 nm, a polydispersity index of 0.351, and a high encapsulation efficiency of 98.09 %. Spectroscopic and X-ray diffraction analyses confirmed the incorporation of Que into amorphous PU-SMP matrices through hydrogen bonding, electrostatic, hydrophobic, and van der Waals interactions. Nanoencapsulation significantly

Design and Synthesis of Novel Indole Ethylamine Derivatives as a Lipid Metabolism Regulator Targeting PPARα/CPT1 in AML12 Cells.

Peroxisome proliferator-activated receptor alpha (PPARα) and carnitine palmitoyltransferase 1 (CPT1) are important targets of lipid metabolism regulation for nonalcoholic fatty liver disease (NAFLD) therapy. In the present study, a set of novel indole ethylamine derivatives (4, 5, 8, 9) were designed and synthesized. The target product (compound 9) can effectively activate PPARα and CPT1a. Consistently, in vitro assays demonstrated its impact on the lipid accumulation of oleic acid (OA)-induced AML12 cells. Compared with AML12 cells treated only with OA, supplementation with 5, 10, and 20 μM o

Preanalytical considerations in parathyroid hormone measurement.

Automated immunoassays used to evaluate parathyroid function are vulnerable to different types of interference, which can affect clinical practices. This review provides a detailed overview of the six main types of interference known to affect the measurement of parathyroid hormone (PTH): heterophilic antibodies, biotin, PTH fragments, oxidized PTH (oxPTH), phosphorylated PTH, and some preanalytical factors. Because the prevalence of some of these conditions has been reported to approach 11.7%, and the frequency of testing for parathyroid function is important, the scale of the problem might b

Ginsenoside Rb3 attenuates skin flap ischemia-reperfusion damage by inhibiting STING-IRF3 signaling.

We investigate the protective effect of ginsenoside Rb3 on skin flap microvasculature following ischemia-reperfusion (I/R) injury and its regulatory mechanism. We used a rat model of I/R injury with the right iliolumbar artery and oxidative stress model of human dermal microvascular endothelial cells. The effects of Rb3 on skin flap tissue and endothelial cell survival, STING-IRF3 pathway activation, and endothelial cell adhesion were measured. Following reperfusion, the survival rate of rat perforator flaps in the Rb3-treated group gradually increased with increasing Rb3 concentration. The tr

Gallium Nanodroplets are Anti-Inflammatory without Interfering with Iron Homeostasis.

Gallium (Ga) compounds, as the source of Ga ions (Ga3+), have been historically used as anti-inflammatories. Currently, the widely accepted mechanisms of the anti-inflammatory effects for Ga3+ are rationalized on the basis of their similarities to ferric ions (Fe3+), which permits Ga3+ to bind with Fe-binding proteins and subsequently disturbs the Fe homeostasis in the immune cells. Here in contrast to the classic views, our study presents the mechanisms of Ga as anti-inflammatory by delivering Ga nanodroplets (GNDs) into lipopolysaccharide-induced macrophages and exploring the processes. Whey fermented by Enterococcus faecalis M157 exhibits antiinflammatory and antibiofilm activities against oral pathogenic bacteria.

The aim of the present study was to investigate the antiinflammatory and antibiofilm effects of whey fermented by Enterococcus faecalis M157 (M157-W) against oral pathogenic bacteria. The M157-W significantly inhibited IL-1β, IL-6, and nitric oxide induced by the lipopolysaccharide of Porphyromonas gingivalis in RAW 264.7 cells. The M157-W also inhibited the production of IL-1β and IL-8 in human periodontal ligament cells. Treatment with M157-W suppressed the phosphorylation of mitogen-activated protein kinases as well as the activation of nuclear factor-κB in RAW 264.7 cells stimulated by P.

Protective effects of phenolic acid extract from ginseng on vascular endothelial cell injury induced by palmitate via activation of PI3K/Akt/eNOS pathway.

Elevated free fatty acids may impair insulin-mediated signaling to eNOS that contributes to the pathophysiology of endothelial dysfunction. Previous studies have indicated the protective effect of ginseng and the regulatory potential of phenolic acid components from other plants on endothelial function. Therefore, this study investigated the protective effects of phenolic acid extract from ginseng (PG2) on endothelial cells against palmitate-induced damage. We found that PG2 increases cell viability, inhibits the palmitate-induced intracellular accumulation of lipids, and the overexpression of

Crucial players in Alzheimer's disease and diabetes mellitus: Friends or foes?

Alzheimer's disease (AD) and diabetes mellitus, especially type 2 (T2DM), are very common and widespread diseases in contemporary societies, and their incidence is steadily on the increase. T2DM is a multiple metabolic disorder, with several mechanisms including hyperglycaemia, insulin resistance, insulin receptor and insulin growth factor disturbances, glucose toxicity, formation of advanced glycation end products (AGEs) and the activity of their receptors. AD is the most common form of dementia, characterized by the accumulation of extracellular beta amyloid peptide aggregates and intracellu

Long term Westernized diet leads to region-specific changes in brain signaling mechanisms.

Western diets, high in fat and energy, are associated with cognitive deficits in humans and animal models, but the underlying mechanisms are not fully elucidated. This includes whether diet-induced dyslipidemia per se negatively impacts brain signaling. Here we investigate the effects of dyslipidemia induced by two high fat diets with or without high sucrose on hippocampal and frontal cortical oxidative stress, brain-derived neurotrophic factor (BDNF) and down-stream markers of synaptic plasticity, as well as alterations in monoaminergic neurotransmitter levels. A high fat diet was associated
