Parasitic infections control practices and antiparasitic drug usage by veterinarians in the Azores archipelago, Portugal.

In the Azores archipelago, data regarding the current parasite control strategies adopted in domestic animals within the local veterinary community is inexistent till the moment. The present study aimed to evaluate the knowledge, approaches, and practices of local veterinarians of Azores islands, regarding parasitic infections control and antiparasitic drugs usage in veterinary practice. The research was conducted between June 2024 and April 2025 with 107 veterinarians from eight different islands of the Azores archipelago. The study surveyed a diverse group of veterinarians working in various

Changing parasite landscapes in captive primates: methodological advances and findings from the Wrocław Zoo.

Long-term parasitological studies of non-human primates (NHPs) kept in zoological gardens offer valuable insights into host-parasite relationships in controlled settings. Despite regular veterinary supervision and preventative measures, many surveys have shown that captive primates often harbour diverse intestinal parasites, including species of zoonotic importance. This work summarises the historical and current parasitological research conducted on primates at Wrocław Zoological Garden, Poland, and highlights changes in diagnostic techniques, parasite diversity, and infection patterns over t

A novel anti-Toxoplasma peptide suppresses parasite invasion and rescues host autophagic defenses.

Toxoplasma gondii, a ubiquitous intracellular protozoan parasite, poses life-threatening risks to immunocompromised hosts. Current first-line treatments for toxoplasmosis are limited by significant toxicity and high post-treatment recurrence rates. In this study, we employed phage display technology to identify peptides targeting TgMIC6 and disrupting its immune evasion function. Among these, the C8 peptide exhibited dual efficacy: it not only inhibited T. gondii invasion in vitro but also restored host autophagy, countering the parasite's immune escape mechanisms. Furthermore, in vivo studies

An Integrated Model of Biphasic Apoptosis in Avian Coccidiosis: Molecular Networks and Host-Parasite Interplay.

Coccidiosis is one of the most serious parasitic diseases in poultry, with Eimeria-induced apoptosis of IECs recognized as a key pathogenic mechanism. This review systematically delineates the molecular mechanisms governing this apoptotic process. The invasion process of Eimeria app. is mediated by the AMA1-RON2 moving junction complex and secreted effector proteins. An integrated model of apoptotic regulation is proposed. This model comprises the mitochondrial, death receptor, and endoplasmic reticulum stress pathways, which are coordinated by signaling hubs, such as PI3K/Akt, NF-κB, and JNK/

Interplay of obesity and parasitic infection: current evidence of immunogenesis, tumorigenesis and leptin receptor involvement.

BACKGROUND: Parasitic infections possess comparable risk factors to obesity. In addition, obesity impairs innate and adaptive immunity subsequently increasing vulnerability to infectious diseases. AIM: The study investigated the leptin/leptin receptors, obesity-parasites mutual relationship and the effect of parasitic infections on immune metabolism, microbiota, and tumorigenesis in the context of obesity. METHODOLOGY: To implement the current review, articles were gathered using the Egyptian Knowledge Bank (EKB), Web of Science, PubMed, and Google Scholar. CONCLUSION: Leptin enhances anti-

A new blood parasite of the accentor birds: description, molecular characterization, phylogenetic relationships and distribution.

Haemoproteus bobricklefsi sp. nov. (Haemosporida, Haemoproteidae) was found in the dunnock Prunella modularis and represents the first blood parasite described in accentor birds of the Prunellidae. The description is based on the morphology of blood stages and includes information about a barcoding segment of the mitochondrial cytochrome b gene (lineage hDUNNO01) and the full mitochondrial genome, which can be used for identification and diagnosis of this infection. The new parasite can be readily distinguished from described species of haemoproteids parasitizing passeriform birds due to marke

Role for gene conversion in the evolution of cell-surface antigens of the malaria parasite Plasmodium falciparum.

While the malaria parasite Plasmodium falciparum has low average genome-wide diversity levels, likely due to its recent introduction from a gorilla-infecting ancestor (approximately 10,000 to 50,000 years ago), some genes display extremely high diversity levels. In particular, certain proteins expressed on the surface of human red blood cell-infecting merozoites (merozoite surface proteins (MSPs)) possess exactly 2 deeply diverged lineages that have seemingly not recombined. While of considerable interest, the evolutionary origin of this phenomenon remains unknown. In this study, we analysed t

Positive-unlabeled learning identifies vaccine candidate antigens in the malaria parasite Plasmodium falciparum.

Malaria vaccine development is hampered by extensive antigenic variation and complex life stages of Plasmodium species. Vaccine development has focused on a small number of antigens, many of which were identified without utilizing systematic genome-level approaches. In this study, we implement a machine learning-based reverse vaccinology approach to predict potential new malaria vaccine candidate antigens. We assemble and analyze P. falciparum proteomic, structural, functional, immunological, genomic, and transcriptomic data, and use positive-unlabeled learning to predict potential antigens ba

Application of machine learning in a rodent malaria model for rapid, accurate, and consistent parasite counts.

Rodent malaria models serve as important preclinical antimalarial and vaccine testing tools. Evaluating treatment outcomes in these models often requires manually counting parasite-infected red blood cells (iRBCs), a time-consuming process, which can be inconsistent between individuals and labs. We have developed an easy-to-use machine learning (ML)-based software, Malaria Screener R, to expedite and standardize such studies by automating the counting of Plasmodium iRBCs in rodents. This software can process Giemsa-stained blood smear images captured by any camera-equipped microscope. It featu

Calcium-binding protein TgpCaBP regulates calcium storage of the zoonotic parasite Toxoplasma gondii.

Toxoplasma gondii, the causative parasite of toxoplasmosis, is an apicomplexan parasite that infects warm-blooded mammals. The ability of the calcium-binding proteins (CBPs) to transport large amounts of Ca2+ appears to be critical for the biological activity of T. gondii. However, the functions of some members of the CBP family have not yet been deciphered. Here, we characterized a putative CBP of T. gondii, TgpCaBP (TGME49_229480), which is composed of four EF-hand motifs with Ca2+-binding capability. TgpCaBP was localized in the cytosol and ER of T. gondii, and parasites lacking the TgpCaBP

Cap-independent translation directs stress-induced differentiation of the protozoan parasite Toxoplasma gondii.

Translational control mechanisms modulate microbial latency of eukaryotic pathogens, enabling them to evade immunity and drug treatments. The protozoan parasite Toxoplasma gondii persists in hosts by differentiating from proliferative tachyzoites to latent bradyzoites, which are housed inside tissue cysts. Transcriptional changes facilitating bradyzoite conversion are mediated by a Myb domain transcription factor called BFD1, whose mRNA is present in tachyzoites but not translated into protein until stress is applied to induce differentiation. We addressed the mechanisms by which translational

The cyclophilin D (CypD) of Toxoplasma gondii is involved in the parasite's response to oxidative stress damage.

The mitochondrial permeability transition pore (mPTP) significantly impacts mitochondrial responses to cell death signals through its structural opening. Cyclophilin D (CypD) serves as a key regulator of the mPTP and plays a pivotal role in governing mitochondrial responses to cell death. In this study, we have demonstrated that Toxoplasma expresses a homolog of cyclophilin D, named TgCypD, which is localized in the mitochondria. Depletion of TgCypD resulted in a modest inhibition of tachyzoite invasion and proliferation, with no notable effect on mitochondrial morphology. However, TgCypD defi

Cutaneous Rosai-Dorfman disease with MAP2K1 mutation, initially mimicking an infection with parasitized histiocytes.

Rarely, Rosai-Dorfman disease (RDD) manifests exclusively in the skin, typically as nodules on the trunk and extremities. Recognition of characteristic histopathologic features enables diagnosis of RDD. A 55-year-old female presented with a 7-year history of cutaneous nodules involving the trunk and extremities. A prior skin biopsy specimen at a different institution had demonstrated a dense dermal lymphohistiocytic infiltrate with histiocytes containing GMS+ forms, favored to represent cryptococcal organisms, with a differential diagnosis including other infections with parasitized histiocyte

Iron transport pathways in the human malaria parasite Plasmodium falciparum revealed by RNA-sequencing.

Host iron deficiency is protective against severe malaria as the human malaria parasite Plasmodium falciparum depends on bioavailable iron from its host to proliferate. The essential pathways of iron acquisition, storage, export, and detoxification in the parasite differ from those in humans, as orthologs of the mammalian transferrin receptor, ferritin, or ferroportin, and a functional heme oxygenase are absent in P. falciparum. Thus, the proteins involved in these processes may be excellent targets for therapeutic development, yet remain largely unknown. Here, we show that parasites cultured

Locomotor activity and physiological responses of parasite-infected Gammarus fossarum exposed to the herbicide metazachlor.

Herbicides are among the most commonly found contaminants in freshwater ecosystems. Standard tests are frequently employed to assess their ecotoxicological impacts, but sublethal endpoints in non-target species are often not considered. In addition, ecotoxicological investigations rarely take into account that many species from field populations are naturally infected with parasites. To overcome these gaps, our study aimed to investigate how environmentally relevant concentrations of the herbicide metazachlor affect the locomotor activity and selected physiological responses of Gammarus fossar

Intracranial Parasitic Fetus in a Living Infant: A Case Study with Surgical Intervention and Prognosis Analysis.

BACKGROUND Fetus in fetu (FIF), or parasitic fetus, is a rare malformation that typically occurs in the retroperitoneum, but can be found in other unusual locations, such as the skull, sacrum, and mouth. The presence of a spine is necessary for diagnosis. CASE REPORT Intracranial FIFs were retrospectively studied. Abnormalities were detected in the fetal head during a 33-week prenatal examination; however, MRI could not provide more information, due to space occupation. A baby girl was born via cesarean delivery at 37 weeks, with a large head circumference. She had delays in motor skills and s

Mosquito salivary apyrase regulates blood meal hemostasis and facilitates malaria parasite transmission.

The evolution of hematophagy involves a series of adaptations that allow blood-feeding insects to access and consume blood efficiently while managing and circumventing the host's hemostatic and immune responses. Mosquito, and other insects, utilize salivary proteins to regulate these responses at the bite site during and after blood feeding. We investigated the function of Anopheles gambiae salivary apyrase (AgApyrase) in regulating hemostasis in the mosquito blood meal and in Plasmodium transmission. Our results demonstrate that salivary apyrase, a known inhibitor of platelet aggregation, int
