Role of mGluR7 in Alzheimer's disease: pathophysiological insights and therapeutic approaches.

Alzheimer's disease (AD) is a progressive neurodegenerative disease characterised by oxidative stress, mitochondrial dysfunction, synaptic impairment, and neuronal loss. The progression of AD depends on two main pathologic features, amyloid-beta accumulation, and tau pathology, whereas the disruption of glutamatergic neurotransmission plays an essential role in disease progression. Glutamate, the brain's primary excitatory neurotransmitter, acts on ionotropic and metabotropic glutamate receptors (mGluRs). Metabotropic glutamate receptor 7 (mGluR7) is a pre-synaptic type III mGluR receptor play

A multifunctional hydrogel system with synergistic effects against Staphylococcus aureus infections and inflammation in perioperative cataract surgery.

Advancing therapeutic strategies with high bioavailability, simplified regimens, and reduced bacterial resistance is crucial for perioperative cataract care. Herein, multifunctional hydrogel eye drops (designated as FQT) were synthesized through a simple one-pot synthesis, employing thermosensitive F127 as the structural scaffold and incorporating quaternized chitosan (QCS) and tannic acid (TA). QCS and TA not only serve as active agents conferring synergistic non-antibiotic antibacterial and anti-inflammatory effects, but also cooperatively interact with F127 through hydrogen bonding, ionic i

Milk-derived peptide fibril-iron nanoparticles prepared by a coordination-driven in-situ reduction approach with high bioavailability and sustained iron (II) protection.

Iron deficiency anemia is a worldwide public health problem that urgently needs to be addressed. Current iron supplementation is restricted … th fibrils at pH 2.0 and 60 °C. The fibrils reduce 71 % of iron (III) ions in-situ without external reducing agents. This process produced iron NPs that were coordinated with -COO-, -NH2, and -SH groups on fibrils. The fibril‑iron NPs remain intact and protect iron NPs under simulated gastric digestion. However, they degrade to peptide‑iron NPs after being expose …

Harnessing Azelaic Acid for Acute Myeloid Leukemia Treatment: A Novel Approach to Overcoming Chemoresistance and Improving Outcomes.

Azelaic acid (AZA), an aliphatic dicarboxylic acid (HOOC-(CH2)7-COOH), is widely used in dermatology. It functions as an inhibitor of tyrosinase, mitochondrial respiratory chain enzymes, and DNA synthesis, while also scavenging free radicals and reducing reactive oxygen species (ROS) production by neutrophils. AZA has demonstrated anti-proliferative and cytotoxic effects on various cancer cells. However, its therapeutic potential in acute myeloid leukemia (AML) remains largely unexplored. AML is a complex hematologic malignancy characterized by the clonal transformation of hematopoietic precur

Nanogel delivered benzimidazole-triazole hybrids as potent multitarget inhibitors of topoisomerase and tubulin in triple-negative breast cancer.

Breast cancer remains a major therapeutic challenge due to poor selectivity, multidrug resistance, and systemic toxicity of conventional chemotherapeutics. Here, we report the design, synthesis, and nanogel-based delivery of benzimidazole-triazole hybrids (HSRA1,2, and 3) for targeted anticancer therapy. Structurally, HSRA1 is a bis-N/S-triazole amide bearing fluorinated aromatic substituents, HSRA2 incorporates a methyl-substituted nucleoside-like triazole, while HSRA3 is a minimal S-triazole amide scaffold. Chitosan/polyvinyl pyrrolidone (Cs/PVP) nanogels encapsulated these compounds efficie

Inonotus obliquus polysaccharides improve hyperlipidemia with respect to lipid synthesis and intestinal flora modulation.

Chronic high-fat diet can lead to fatty liver metabolism, hyperlipidemia disorders, and other diseases. Polysaccharides are natural lipid-lowering agents. Nevertheless, the lipid-lowering mechanism of Inonotus obliquus polysaccharides (IOP) needs to be clarified. This study extracted polysaccharides from Inonotus obliquus, and their physicochemical properties and hypolipidemic activity were investigated. Structural characterization by scanning electron microscopy and Fourier transform infrared spectroscopy indicated that IOP displayed functional groups and morphological features typical of pol

An updated review on systemic therapy for brain metastases in non-small cell lung cancer.

Brain metastases (BrM) are a frequent and devastating complication of non-small cell lung cancer (NSCLC), affecting up to 40% of patients during their disease course. Historically, treatment relied primarily on surgery and radiation, as systemic therapies were thought to have limited activity in the central nervous system (CNS). However, over the past two decades, the development of more targeted therapies has transformed the treatment landscape. Many of these newer agents have improved CNS penetration, which may offer patients the possibility of deferring or reducing the need for local therap

Exploring the potential of selective FKBP51 inhibitors on melanoma: an investigation of their in vitro and in vivo effects.

FKBP51 is a marker of melanocyte malignancy, correlating with vertical growth phase and lesion thickness. It promotes the typical features of epithelial to mesenchymal transition and sustains apoptosis resistance. The present study aimed to assess in vitro and in vivo the efficacy against melanoma of selective small molecules targeting FKBP51, called SAFits. Our findings reveal differing outcomes for SAFits in vitro compared to in vivo. SAFit increased the doxorubicin and dacarbazine cytotoxicity of cultured melanoma cells and was effective in impairing NF-κB activity and related pro-survival

N-Acetyldopamine Dimer from Cicadidae Periostracum Is Enantioselectively Neuroprotective via Antioxidant Property.

N-Acetyldopamine oligomers are typically biosynthesized as racemic mixtures, yet enantiomers can differ markedly in pharmacological efficacy and safety. In this study, we isolated a pair of enantiomers of an N-acetyldopamine dimer (compound 1) from Cicadidae Periostracum, a traditional medicinal substance, and characterized their structures using mass spectrometry and 1D/2D NMR spectroscopy. The absolute configurations of the enantiomers-1a (2S,3R,1''R) and 1b (2R,3S,1''S)-were determined for the first time through a combination of electronic circular dichroism and Mosher's esterification anal

Proteomic Learning of Gamma-Aminobutyric Acid (GABA) Receptor-Mediated Anesthesia.

Anesthetics are crucial in surgical procedures and therapeutic interventions, but they come with side effects and varying levels of effectiveness, calling for novel anesthetic agents that offer more precise and controllable effects. Targeting Gamma-aminobutyric acid (GABA) receptors, the primary inhibitory receptors in the central nervous system, could enhance their inhibitory action, potentially reducing side effects while improving the potency of anesthetics. In this study, we introduce a proteomic learning of GABA receptor-mediated anesthesia based on 24 GABA receptor subtypes by considerin
