Establishing synergistic role of acacia honey and vitamin C: A promising strategy for faster wound tissue regeneration.

Being established as an important natural component for wound healing, acacia honey (AH) in combination with ascorbic acid (AA) was evaluated for its tissue regenerative properties to establish a synergistic effect. A thermosensitive hydrogel formulation was developed and characterized for viscosity, mucoadhesion, moisture retention, pH, and morphology, where the parameters were found to be favourable for topical application and longer retention. The porosity and swelling of the hydrogel matrix allow for prolonged and sustained release of AH and AA. The release patterns of AH and AA from the g

Integrated haemostasis-immunomodulation-regeneration design: Shape-memory PGM sponges for noncompressible haemorrhage control and diabetic wound healing.

To achieve rapid haemostasis in noncompressible wounds, haemostatic sponges that combine high liquid absorption with rapid shape recovery are urgently required. However, conventional sponges struggle to balance high porosity with rapid shape restoration. In this study, we developed a water-triggered shape-memory haemostatic sponge loaded with MaBo powder (WSH-M). Oxidised pectin (OG) was crosslinked with a porcine acellular dermal matrix (PADM) via Schiff base reactions, yielding a highly porous (∼ 85%) three-dimensional interpenetrating network with rapid water-activated shape recovery. The P

Dynamic dual regulation of amphiregulin in liver pathophysiology: balancing regeneration and disease progression via the EGFR axis.

Liver diseases, ranging from acute injury to chronic fibrosis and hepatocellular carcinoma (HCC), represent a major global health challenge with limited therapeutic options. The liver's inherent regenerative capacity can be disrupted during chronic disease, resulting in poor outcomes. However, this process is complex, as key regulatory molecules can exert opposite effects. A notable example is Amphiregulin (AREG), an epidermal growth factor receptor (EGFR) ligand whose role changes from promoting protective regeneration in acute injury to driving harmful processes in chronic settings. This rev

Mechanically Preconditioned Biomimetic Gradient Scaffold for Tendon-to-Bone Regeneration in a Rabbit Rotator Cuff Tear Model.

BACKGROUND: Healing of the tendon-bone interface (TBI) after rotator cuff injury is limited, often leading to scar-mediated repair. Decellularized tendon fibrocartilage-bone composite (dTFBC) and bone marrow mesenchymal stem cell sheets (BMSCS) improve repair in preclinical models, but full scaffold encapsulation with mechanical preconditioning remains unexplored. PURPOSE: To evaluate TBI regeneration using mechanically preconditioned, BMSCS-encapsulated dTFBC scaffolds in a rabbit model. STUDY DESIGN: Controlled laboratory study. METHODS: A total of 48 male New Zealand White rabbits were r

An oxygen-glucose co-releasing platform fostering dental pulp regeneration by driving metabolic recovery of stem cells.

In dental pulp regeneration, the ischemic microenvironment within the root canal severely compromises the survival and function of transplanted human dental pulp stem cells (hDPSCs). Here, we developed a PDMS-based core-shell oxygen-glucose delivery platform (P-C@P-G) that continuously released oxygen and glucose for up to 40 and 29 days, respectively, while minimizing peroxide-associated cytotoxicity through diffusion-controlled regulation without the need for exogenous enzymes. Under oxygen-glucose deprivation (OGD, 0.1% O2, glucose-free conditions) conditions, P-C@P-G improved cellular meta

Fisetin promotes liver regeneration by inducing β-catenin-mediated inhibition of p53 and suppression of TGF-β.

Partial hepatectomy (PHx) is frequently done for the repair of traumatic liver injury, tumor resection, and donation. The regenerative ability of the residual liver impacts mortality and morbidity. Fisetin is an antioxidant flavonoid that was found to target β-catenin, inducing regeneration in cardiac, nervous, bony tissues, and skin. This study assesses the impact of fisetin on liver regeneration following major PHx. Twenty-four rats were assigned to four groups: sham, fisetin control, 70% PHx, and PHx + Fisetin (10 mg/kg/day) for four days prior to surgery, and two days after surgery. Animal

Naringenin modulate peroxisome proliferator-activated receptorα (PPARα) to ameliorate hepatic ferroptosis by restoring GPX4 during liver regeneration.

Liver regeneration is essential for recovery after major resection or acute injury, yet early regenerative failure remains common. Ferroptosis, an iron-dependent form of lipid peroxidation-driven cell death, impairs hepatocyte survival and may constrain regeneration. Naringenin (NAR), a citrus flavonoid with hepatoprotective activity, has been reported to modulate oxidative stress; however, whether it supports liver regeneration through ferroptosis modulation and which upstream targets are involved remains unclear. Here, hepatocyte models (AML12, Huh7, HepG2) and a murine 70% partial hepatecto

Glutathione peroxidase 3 preserves hepatocyte mitochondrial quality control to enhance macrophage pro-regenerative phenotype during liver regeneration.

BACKGROUND: Precise regulation of mitochondrial function is critical for liver regeneration. However, the underlying regulatory mechanism remains elusive. Here, we aimed to investigate the role of hepatocellular glutathione peroxidase 3 (GPX3) in liver regeneration. METHODS: In a 70% partial hepatectomy (PH) mouse model, immunostaining and single-cell RNA sequencing revealed significant enrichment but down-regulation of mitochondrial oxidative phosphorylation pathways post-PH, along with up-regulated hypoxia-inducible factor 1a (HIF-1a) and GPX3 in hepatocytes. Single-cell analysis confirmed

A reparative neutrophil subpopulation accelerates spinal cord regeneration in zebrafish by controlling macrophage inflammation via Il-4.

In mammals, a dysregulated immune response is detrimental to spinal cord repair. In zebrafish, which are capable of spinal cord regeneration, the immune response promotes regeneration. Neutrophils are the first immune cells to arrive at a spinal cord injury site, but their role in successful regeneration is not fully understood. Here we show that ablating neutrophils, including a subpopulation that expresses the cytokine il4, increases expression of il1b (coding for Il-1β) mainly in macrophages/microglia and delays anatomical and functional recovery after a spinal cord injury in larval zebrafi

Macrophages orchestrate tail regeneration in lizards Scincella tsinlingensis through spatiotemporal control of immunity, oxidative stress, and myogenesis.

BACKGROUND: Macrophages facilitate blastema and wound epithelium formation by clearing debris and remodeling the immune and extracellular matrix. The M1-to-M2 transition is well documented in amphibians, but its occurrence and timing in diverse lizards remain unclear. This study examined macrophage function during tail regeneration in Scincella tsinlingensis. METHODS: A tail amputation model was established in S. tsinlingensis, and macrophages were depleted using clodronate liposomes (L-Clodronate). Transcriptomic analysis was performed during the wound epithelialization phase, followed by qR

Bioactive Ion-Doped Monetite/Chitosan Thermosensitive Hydrogels with Dual Osteogenic and Pro-Angiogenic Stimuli for Bone Regeneration.

The development of multifunctional scaffolds capable of orchestrating the complex cascade of bone regeneration, cell recruitment, vascularization, and extracellular matrix deposition remains a significant challenge in regenerative medicine. This study introduces a dual-strategy biomaterial system. On one hand, pure monetite (CaP), cobalt-doped monetite (CoCaP), and magnesium-doped monetite (MgCaP) powders were synthesized via coprecipitation to deliver specific ionic cues. On the other hand, these bioactive powders were incorporated into a thermosensitive chitosan/β-glycerophosphate (β-GP) hyd

Dual-Role Phosphorus Species Enable Regeneration of Spent Graphite via Coupled Bulk Doping and Surface Interphase Engineering.

With the rapid expansion of electric mobility and large-scale energy storage systems, the high-value regeneration of graphite anodes from retired lithium-ion batteries has attracted increasing attention. The degradation mechanism of graphite anodes involves multiple factors, including bulk structural damage and interfacial deterioration during cycling. However, state-of-the-art regeneration approaches have been restricted to addressing either structural degradation or interfacial instability in isolation, precluding the attainment of desirable electrochemical performance. To circumvent this fu

Exosome-Laden 3D Composite Mesh Attenuates Stress Concentration and Promotes Collagen Regeneration toward Pelvic Organ Prolapse Repair.

Pelvic organ prolapse is a prevalent condition that impairs the quality of life in middle-aged and elderly women. Traditional polypropylene (PP) surgical meshes often lead to long-term complications due to mechanical mismatch and biological inertness, manifesting as stress concentration and chronic inflammation. To concurrently address these dual challenges, this study designed and fabricated a multifunctional composite mesh. The mesh incorporates a clinical-grade PP base for mechanical support, integrated with a three-dimensional printed polycaprolactone (PCL) biomimetic network modified by q

Attenuation of epicardial activation and myofibroblast abundance via the Fbln2-Nupr1b axis stimulates cardiac regeneration in zebrafish.

After injury, the adult human heart fails to regenerate and forms a persistent fibrotic scar. By contrast, fibrosis is transient in the injured zebrafish heart, facilitating cell recruitment and providing regenerative cues. The mechanisms that restrain excessive fibrosis while enabling regeneration remain poorly understood. Here we show that fibulin-2 (Fbln2) regulates specific populations of activated epicardial cells to balance the response to cardiac injury. Using genetic tools for Fbln2 dosage, we find that attenuation of epicardial activation stimulates regenerative programs. Mechanistica

Stem cell-derived extracellular vesicles as immunomodulators: a novel paradigm for post-myocardial infarction repair and regeneration.

Myocardial infarction (MI) initiates a rapid and highly coordinated immune response that is essential for the clearance of necrotic tissue and activation of reparative processes. However, prolonged or dysregulated post-MI inflammation can exacerbate myocardial injury, promote adverse cardiac remodeling, and ultimately contribute to heart failure. Although current therapeutic strategies improve survival and symptom management, they remain limited in their ability to restore lost cardiomyocytes or effectively modulate the post-infarction immune microenvironment. In this context, stem cell-derive

Bone-targeted calcification-photothermal nanoplatform for synergistic tumor ablation and bone regeneration in breast cancer bone metastasis.

Breast cancer bone metastasis is a common and devastating complication among patients with advanced breast cancer, marked by aggressive tumor growth and extensive osteolytic destruction. Effective treatment requires simultaneous elimination of tumor cells and restoration of bone tissue. To address this challenge, we developed a bone-targeting nanomaterial, CPPA NPs, composed of a polydopamine-coated calcium peroxide core embedded with palladium nanoparticles, and functionalized with phytic acid for bone affinity. This design integrates calcification and photothermal activity to achieve synergi
