Increased low-molecular-weight mucins in muco-obstructive airway disease limit Staphylococcus aureus growth.

Muco-obstructive airway diseases result in an increase in mucus accumulation and a decrease in mucus clearance. MUC5B is the most abundant secreted mucin in the human airways, and MUC5B mucin strands dimerize to create the mucus mesh network in the healthy respiratory tract. In muco-obstructive airway diseases like cystic fibrosis (CF), immune cells and bacteria release enzymes that degrade MUC5B into smaller fragments that become entangled and compacted, contributing to pathogenesis. We utilized synthetic cystic fibrosis sputum media (SCFM) to examine how mucin polymers can impact Staphylococ

Case Report: Moxibustion-induced burns leading to disseminated methicillin-resistant Staphylococcus aureus infection in a patient with type 2 diabetes mellitus.

PURPOSE: To assess methicillin-resistant Staphylococcus aureus (MRSA) bloodstream dissemination risk from moxibustion burns in type 2 diabetes and raise awareness of iatrogenic skin injuries in high-risk populations. BACKGROUND: Staphylococcus aureus (S. aureus) causes bloodstream and metastatic infections, with a mortality rate up to 30%. In diabetic patients, minor skin injuries can lead to fatal systemic infections due to immune dysfunction and neuropathy. Moxibustion is widely used for chronic pain, but its thermal safety in high-risk groups is under-evaluated. Multiorgan MRSA disseminati

Intra-house circulation and persistence of methicillin-resistant Staphylococcus pseudintermedius in Brazil: Evidence of transmission between a dog and its family.

We investigated a case of postoperative osteomyelitis in a dog caused by methicillin-resistant Staphylococcus pseudintermedius (MRSP) at a veterinary teaching hospital in Brazil, and screened the cohabiting dog, five household members and the attending surgeon for MRSP and other methicillin-resistant staphylococci (MRS) during infection (2021) and two years later (2023). Nasal and skin swabs were cultured on oxacillin-supplemented media. Isolates were identified as MRSP by PCR and characterized by PFGE and whole-genome sequencing, including MLST, SCCmec typing, resistance gene profiling and co

Daptomycin-Induced Eosinophilic Pneumonia in a Patient Treated for MRSA Foot Infection: A Case Report.

Daptomycin is widely used for serious methicillin-resistant Staphylococcus aureus (MRSA) infections, but it can cause eosinophilic pneumonia, an uncommon and potentially severe adverse drug reaction. We report probable daptomycin-induced eosinophilic pneumonia (DIEP) in a 77-year-old woman receiving intravenous daptomycin with rifampin for a postoperative MRSA foot infection with concern for orthopedic-device involvement and osteomyelitis. Daptomycin had been initiated 10 days before admission, when she developed acute hypoxemic respiratory failure with bilateral ground-glass and consolidative

Imaging Staphylococcus aureus biofilms in vivo reveals neutrophil exclusion by PNAG exopolysaccharide.

Staphylococcus aureus forms biofilms on biotic and abiotic surfaces, enabling persistent infections that evade immune clearance. Although many S. aureus strains can produce the biofilm-associated exopolysaccharide, poly-β-1,6-N-acetyl-D-glucosamine (PNAG), they often form protein-dominated biofilm matrices in vitro, leaving a role for PNAG unclear. Using intravital imaging in a foreign-body infection model, we found that PNAG in biofilms hinders neutrophil access and delays bacterial clearance. Neutrophil elastase was crucial for eventual biofilm clearance. In vivo PNAG labeling revealed that

Factors Affecting the Initial Serum Concentration of Teicoplanin in Pediatric Patients: A Single-Center Retrospective Analysis.

BACKGROUND: The glycopeptide antibiotic teicoplanin (TEIC) is used to treat infections caused by methicillin-resistant Staphylococcus aureus and other gram-positive bacteria. Because higher target trough concentrations are now recommended, higher loading dose (LD) regimens have been proposed. However, the factors affecting trough concentration (Ctrough) levels and the safety of high-dose regimens remain unclear. This study aimed to investigate the factors that should be considered in the LD regimen of TEIC in pediatric patients and assess the safety of a higher LD. METHODS: We enrolled pediat

The toxic effects of α-hemolysin and β-hemolysin produced by Staphylococcus aureus.

Staphylococcus aureus (S. aureus) is a Gram-positive bacterium widely distributed in nature, which can cause skin and soft tissue infections, sepsis, pneumonia and other invasive diseases. In recent years, the emergence and spread of drug-resistant strains such as Methicillin-resistant Staphylococcus aureus (MRSA) and Vancomycin-resistant Staphylococcus aureus (VRSA) have greatly increased the difficulty of clinical treatment, and there is currently no effective vaccine for the treatment of S. aureus. The pathogenicity of S. aureus depends on a complex and diverse set of virulence factor syste

A review of the randomized clinical trial results from the Staphylococcus aureus network adaptive platform (SNAP) meticillin-susceptible (MSSA) and penicillin-susceptible (PSSA) domains and CloCeBa.

Current guidelines for the management of meticillin-susceptible Staphyloccocus aureus bloodstream infection/bacteraemia (SAB) recommend intravenous (flu)cloxacillin as the first-line treatment. This is based on decades of clinical practice. The high acute kidney injury rates seen with (flu)cloxacillin in the recently published SNAP trial, which is the largest randomized clinical trial ever in S. aureus bacteraemia (SAB), shows us the need to regularly test and question our usual clinical practice. Acute kidney injury is common in SAB when high doses of (flu)cloxacillin are used. The contributi

Interventions for the eradication of meticillin-resistant Staphylococcus aureus (MRSA) in people with cystic fibrosis.

RATIONALE: Cystic fibrosis (CF) is a genetic disorder characterised by recurrent and persistent pulmonary infections from resistant organisms, resulting in deterioration of lung function and early mortality. Meticillin-resistant Staphylococcus aureus (MRSA) is an important infection in hospital patients and harmful to people with CF (pwCF). Chronic pulmonary MRSA infection may confer a worse clinical outcome on pwCF and result in a faster decline in lung function. Robust evidence clearly guiding MRSA eradication in CF is urgently needed. This is an updated review. OBJECTIVES: To evaluate the

Multi-Resistant Staphylococcus aureus Growth Inhibition Using an Innovative High Voltage Nanosecond Pulser: In Vitro Experimental Results.

Multi-Resistant Bacteria (MRB) is a threatening biomedical problem, whose solution is of paramount importance. Due to the antibiotics resistance there is an emerging need for novel treatment strategies and protocolls. As bacteria tolerance in modern chemotherapeytic agents expands, the introduction of alternative methods is fundamental. The use of High voltage Electric Pulses, through a process known as Irreversible Electroporation (IRE), is an effective alternative bacterial control method. This paper describes a new prototype high voltage nanosecond pulser and validates its effectiveness in

Staphylococcus biofilm dynamics and antibiotic resistance: insights into biofilm stages, zeta potential dynamics, and antibiotic susceptibility.

Staphylococcus spp. infections often involve biofilms, but standard antibiotic minimum inhibitory concentration (MIC) testing used to determine treatment evaluates planktonic bacterial growth only and does not account for biofilm presence, strength, or growth stage. To aid in determining a cost-effective method to solve this issue, we built upon in vitro methods initially published by Stepanovic et al. used to determine weak and strong biofilm formations. First, we determined 115 unique S. aureus isolate biofilms at 2, 4, 6, 8, 16, and 24 h to classify the hourly stages of biofilm development

Emergence of NOX1/DUOX NADPH oxidases is a key feature of functional neutrophil reprogramming at the gut and lung barrier.

Functional neutrophil diversity is recognized as a driver of development, progression and resolution of disease. Recruited neutrophils are imprinted by biochemical, biophysical and mechanical stimuli of the encountered microenvironment, altering their genetic and phenotypic program. The functional implications of this reprogramming are of critical importance for devising strategies to modify neutrophil behavior. Oxidant production affects neutrophil responses, shapes the microenvironment and often determines disease outcome in inflammation, infection and cancer. Here we report neutrophil diver

Drug Repurposing: An Emerging Strategy in Staphylococcus aureus Infections.

Infections are a growing global threat to public health, and the rapid spread of drug-resistant pathogens is a big challenge. The development and approval of novel antimicrobials targeting multi-drug-resistant infections lag significantly behind the rapid emergence of resistance. For these threats and challenges, new strategies are critically needed. Drug repurposing, using approved or investigational drugs for other clinical conditions, has emerged as an alternative approach for rapid identification of effective antibacterial agents. Although drug repurposing is not novel, recent years have w

Geographic divergence of methicillin-resistant Staphylococcus aureus ST5-SCCmecI in the aftermath of a major earthquake and tsunami: impact of a plasmid harboring heavy metal resistance genes.

Methicillin-resistant Staphylococcus aureus (MRSA) is a major public health menace. The global spread of MRSA is characterized by successive waves of epidemic clones dominating specific geographical regions. The acquisition of genes encoding resistance to heavy metals (HMRGs) is thought to be a key feature in the geographic divergence of MRSA. However, the cause-effect relationship between the presence of HMRGs and the divergence of MRSA clones remains to be clarified. In this study, we assessed the role that HMRGs may have played in the evolutionary divergence of the MRSA ST5-SCCmecI lineage

Immune escape of Staphylococcus aureus mediated by osteocyte lacuna-canalicular network leads to persistent and uncured bone infection.

Bone infections, specifically chronic osteomyelitis, are characterized by recurrent episodes. They are considered intractable clinical diseases as they require protracted and difficult-to-cure courses. Staphylococcus aureus (S. aureus) is the most common pathogen responsible for bone infections and has high destruction rates. Previous literature has indicated that during S. aureus osteomyelitis, immune evasion mainly involves three mechanisms: biofilm formation, intracellular infection, and abscess formation. However, recently, it was observed that S. aureus can enter and persist for a long ti

Intracellular survival of Staphylococcus aureus in macrophages during osteomyelitis.

Staphylococcus aureus, traditionally viewed as an extracellular pathogen, is increasingly recognized for its ability to persist intracellularly, particularly within macrophages. This intracellular lifestyle is central to osteomyelitis, a chronic bone infection characterized by persistent inflammation, bone destruction, and impaired repair. Within bone, S. aureus exploits macrophage plasticity by driving a shift from pro-inflammatory, bactericidal M1-like states to anti-inflammatory, tissue-reparative M2-like phenotypes. This polarization suppresses immune clearance and promotes an environment

Butyrate Reducing Bone Mass Loss by Regulating the Expression of m6A Methyltransferase METTL3 in Implant-Associated Staphylococcus aureus Osteomyelitis.

Staphylococcus aureus (S. aureus) has been identified as a hindrance to osteoblast differentiation, thereby contributing significantly to the development of osteomyelitis. Consequently, exploring pharmaceutical interventions targeting osteoblast differentiation mediated by S. aureus may present a novel approach for treating osteomyelitis. It has been reported that N6-methyladenosine (m6A) methylation is highly associated with infection. Moreover, studies continue to validate that short-chain fatty acids play an important role in transcriptional modification and have been considered as a potent

Staphylococcus aureus colonization in the pharynx and nasal cavity: why are some people more susceptible?

Staphylococcus aureus is one of the most important pathogenic bacteria in humans. The nose and pharynx constitute two fundamental ecological niches for this bacterium, supporting the asymptomatic carrier state and acting as sources of infection in susceptible organisms. Colonization dynamics depend on the balance between the bacteria's virulence factors, the host's immune response, and the environment. Colonization is favored by attenuated immune responses, with evidence of partial tolerance and low protective antibody titers. Colonization also appears to depend on the microbiome of the coloni

Case Report: Novel variants in the MYD88 gene (c.104T>C, c.141G>C) in a patient with recurrent abscesses as a cause of type 68 immunodeficiency.

BACKGROUND: Primary immunodeficiencies (PIDs) comprise a heterogeneous group of disorders characterized by defects in the immune system, predisposing patients to recurrent and severe infections. Type 68 immunodeficiency, caused by biallelic pathogenic variants in MYD88, is rare; to date, at least 26 affected individuals have been reported in the literature, several of whom belong to the same families. This condition typically presents in early childhood with recurrent severe bacterial infections (SBIs), often accompanied by an absent or attenuated inflammatory response. CASE PRESENTATION: We

Effect of antibiotics on Kupffer cell immunometabolism relative to intracellular killing of S. aureus using NAD(P)H fluorescence lifetime imaging.

Staphylococcus aureus bacteremia (SAB) remains a significant clinical burden with high mortality rates. Kupffer cells (KCs) are integral to clearing Staphylococcus aureus (SA) from the bloodstream yet also serve as a pathogenic reservoir, driving persistence and treatment failure. KCs utilize their metabolic plasticity to coordinate successful antimicrobial responses, and this process may be therapeutically targeted. We investigated the immunometabolic effects of antistaphylococcal antibiotics (vancomycin, ceftobiprole, daptomycin, and tedizolid) on KCs relative to clearance of intracellular S
