Immunological Evaluation of mRNA-LNP Vaccines Targeting Conserved S2 and NSP3 Domains of SARS-CoV-2.

The evolution of SARS-CoV-2 necessitates next-generation vaccine strategies targeting conserved viral elements to ensure broad and durable protection. In this study, we designed and evaluated novel mRNA-based vaccines encoding conserved domains of SARS-CoV-2, including the spike S2 subunit, NSP3 Macrodomain I (Mac I), and papain-like protease (PLpro). These mRNAs were synthesized, cloned, and encapsulated into lipid nanoparticles (LNPs) with 70%-80% efficiency and favorable physicochemical properties. In vitro characterization showed efficient antigen expression with 60%-80% cell survival. Imm

Outbred mice as an appropriate model for evaluating the immunogenicity of vaccines against porcine circovirus 2.

BACKGROUND: Porcine circovirus type 2 (PCV2) is the causative agent of porcine circovirus-associated diseases, leading to substantial economic losses in global swine production. Although various commercial vaccines exist, an appropriate and translational animal model is essential for evaluating vaccine-induced immune responses. Inbred mouse strains are traditionally preferred due to their genetic uniformity; however, outbred animals may reflect a broader immunological variability than the natural target species. The number of comparative studies assessing outbred mice for PCV2 vaccine immunoge

Large and low-PEG lipid nanoparticles enable efficient dendritic cell targeting for potent mRNA Cancer vaccines.

Efficient delivery of messenger RNA (mRNA) to dendritic cells (DCs) remains a major challenge limiting the efficacy of mRNA cancer vaccines. Here, we report a large-sized lipid nanoparticle (LLNP) platform specifically engineered for enhanced DC targeting. By reducing PEG-lipid content to 0.3% and proportionally increasing the concentrations of structural lipids and mRNA sixfold relative to the Moderna classic formulation, we generated LLNPs with enlarged size and optimized surface properties that favor DC uptake. Following intravenous administration, LLNPs achieved markedly enhanced mRNA expr

Personalised neoantigen vaccines for precision oncology in Southeast Asia: clinical opportunities, infrastructure challenges, and policy perspectives.

Personalised neoantigen vaccines are patient-specific immunotherapies designed to target tumour-restricted antigens arising from genomic and transcriptomic alterations, including single-nucleotide variants, insertions and deletions, frameshifts, gene fusions and aberrant splicing. Early clinical studies have established manufacturing feasibility and immunogenicity, while randomised phase 2b evidence in resected melanoma has provided a signal of clinical activity that requires confirmation in larger trials. Clinical development nevertheless remains concentrated in high-income academic centres,

Next-Generation Nanocarrier Platforms for RNA Vaccines: Advances in Formulation, Stability Engineering, and Translational Manufacturing Challenges.

RNA vaccines have emerged as an attractive platform for treating infectious diseases, cancer immunotherapy, and personalized medicine; however, their clinical success depends on multiple factors, including efficient, stable, and scalable delivery systems. Because RNA molecules are highly sensitive to factors such as enzymatic degradation, oxidation, poor cellular uptake, and limited endosomal escape, nanocarrier platforms play essential roles in protecting RNA cargo and enabling effective intracellular delivery. The biological performance of RNA nanocarriers depends on efficient cellular uptak

Potential association of cluster headache with certain COVID-19 vaccines: An assessment from the pharmacovigilance databases.

OBJECTIVES/BACKGROUND: The main objective of our study was to assess the potential association between coronavirus disease 2019 (COVID-19) vaccination and cluster headache (CH) using a pharmacovigilance approach. A secondary objective was to determine whether COVID-19 vaccines predominantly triggered onset or worsening of the disease. Recent pharmacovigilance studies and case reports have suggested an association between COVID-19 vaccines and cluster headache. In order to evaluate whether COVID-19 vaccines could possibly elicit the onset of cluster headache or be responsible for worsening of t

Enhanced immunogenicity and dose-sparing efficacy of self-amplifying RNA vaccines against seasonal influenza across subtypes.

Recent clinical data on seasonal influenza mRNA vaccines have demonstrated suboptimal efficacy against the influenza B virus (IBV). We employed sequence optimization strategies that successfully enhanced the antigen expression of hemagglutinin (HA) and developed mRNA vaccine candidates targeting the WHO-recommended strains. When administered at a low dose (0.1 μg), both mono-and trivalent influenza A mRNA vaccines induced robust humoral immunity and conferred complete protection against homologous viral challenge in murine models, outperforming the quadrivalent inactivated vaccine (QIV, 2 μg).

The Silent Vehicle: Validating the Biological Signals and Noises of Baculovirus Vectors in Macrophage-Targeted Vaccines.

OBJECTIVE: The Baculovirus Expression Vector System (BEVS) is an established eukaryotic platform for high-yield recombinant protein production and is widely utilized for generating vaccine antigens, including recombinant classical swine fever virus E2 glycoprotein, African swine fever virus structural proteins (p72, p30, and p54), and porcine reproductive and respiratory syndrome virus envelope proteins GP5 and M. These antigens interact with macrophage-associated receptors, including CD163 and CD206, to modulate immune responses through macrophage polarization. However, whether baculovirus ve

mRNA Vaccines Targeting the Conserved SARS-CoV-2 Fusion Machinery Elicit Cross-Variant and Robust Immunity.

BACKGROUND/OBJECTIVES: Despite the gradual waning of global interest in SARS-CoV-2, the continued evolution and emergence of viral variants underscore the need for broadly protective vaccines. Vaccines targeting the highly mutable S1 subunit of the spike protein have shown limited breadth of protection, whereas conserved regions within the S2 fusion machinery remain critical targets for the development of vaccines effective against future variants. This study evaluated two mRNA-lipid nanoparticle (mRNA-LNP) vaccines encoding HR121, a conserved HR1-HR2-HR1 fusion-intermediate immunogen, in comp

Investigating the effects and underlying mechanisms of glycosylation sites on the immunogenicity of COVID-19 vaccines.

Glycosylation plays a pivotal role in modulating the structure and immunogenicity of viral antigens. Three glycosylation sites on the receptor-binding domain (RBD) of SARS-CoV-2, including N331, N343 (N-linked), and T323 (O-linked), are highly conserved and remain unchanged across multiple variant strains. To investigate their functional relevance, a series of site-directed glycosylation-deficient mutants were generated on the basis of the RBD-dimer antigen of the ZF2001 vaccine (developed by ZFSW Biologics), including N-N331-NA, N-N343-NA, O-T323-NA, and an RBD-NA control. CircRNA based bi-antigen vaccines against mpox virus induce potent and durable cross-protection in mice.

The ongoing epidemic of mpox highlights the urgency of developing novel and effective vaccines against mpox virus (MPXV). Circular RNA (circRNA) has emerged as a promising novel vaccine platform due to its high stability, resistance to exonuclease-mediated degradation, and potential to support prolonged antigen expression. Based on our lipid nanoparticle (LNP)-encapsulated circRNA vaccine platform, we developed two novel bi-antigen circRNA vaccines cirEV and cirMV, which respectively encode the combination of MPXV extracellular enveloped virion (EEV) antigens (A35R and B6R) or intracellular ma

Enhanced cellular immunity against varicella-zoster virus by β-sitosterol-substituted lipid nanoparticles in mRNA vaccines.

Herpes zoster (HZ), caused by reactivation of varicella-zoster virus (VZV), imposes a substantial health burden, particularly in immunocompromised individuals. Although current HZ vaccines provide effective protection, the live-attenuated vaccine shows age-dependent and waning efficacy, whereas the recombinant zoster vaccine relies on a complex adjuvant system that may present manufacturing and supply challenges. mRNA vaccines offer a promising alternative for HZ prevention, but their efficacy depends on optimized lipid nanoparticle (LNPs) that protect mRNA and support efficient intracellular
