Considerations on the role of buprenorphine in recovery from heroin addiction from a UK perspective.

The United Kingdom Drug Strategy emphasises recovery as a key focus in the treatment of drug dependence. A framework for recovery is defined … ing of on-top use of illicit drugs, the treatment of comorbidities through the minimisation of drug-drug interactions, and a good safety profile. In addition, the newer abuse-deterrent formulation of buprenorphine combined with the opioid antagonist naloxone is likely to strengthen recovery-orientated systems of care due to its potential to reduce misuse and divers …

Influence of preparation methods on solid state supersaturation of amorphous solid dispersions: a case study with itraconazole and eudragit e100.

PURPOSE: The present study aims to determine the drug / polymer miscibility level as a function of the preparation method for an amorphous solid dispersion model system containing itraconazole and eudragit E100. This value was compared to the theoretical crystalline drug solubility in the amorphous polymer and the miscibility of the amorphous drug in the amorphous polymer. METHODS: The amorphous solid dispersions were prepared via spray drying and film casting in order to evaluate the influence of the solvent drying rate. The experimental miscibility level was estimated using XRPD, MDSC, FT-I

Pharmacokinetic/pharmacodynamic correlation and blood-level testing in imatinib therapy for chronic myeloid leukemia.

Imatinib is the current standard of care in the treatment of chronic myeloid leukemia (CML), inducing durable responses and prolonged progression-free survival. However, plasma exposure to the drug from a given dosing regimen can vary widely among patients. Reasons for this may include incomplete adherence, intrinsic variations in the metabolism of imatinib, and drug-drug interactions. Data from two recent studies have shown a correlation between imatinib trough plasma concentration and clinical response, leading to suggestions that maintaining imatinib blood concentrations above approximately…

Genetic alterations determine chemotherapy resistance in childhood T-ALL: modelling in stage-specific cell lines and correlation with diagnostic patient samples.

Acquired drug resistance eventually leads to treatment failure in T-cell acute lymphoblastic leukaemia (T-ALL). Immunophenotypic and cytogenetic heterogeneities within T-ALL influence susceptibility to cytotoxic therapy, and little is known about the mechanisms of drug resistance at specific stages of T-cell ontogeny. We developed tolerance to therapeutic concentrations of daunorubicin (DNR) and L-asparaginase (L-asp) in Jurkat (CD1a(-), sCD3(+)) and Sup T1 (CD1a(+), sCD3(-)) cell lines, having respective 'mature' and 'cortical' stages of developmental arrest. DNR resistant cells acquired mult

[Therapeutic strategy in epileptic patients].

Choosing an antiepileptic drug is crucial because it will be usually taken for many years by the patient. The development of several new dru … ations seems to be not significantly different. The newer drugs may be better tolerated, have fewer drug interactions, and seem to affect cognitive functions to a lesser extent than old drugs, but data on the new drugs are more limited than for older ones. The choice is then guided by the individual appreciation of the risk-benefit ratio in each patient. There are …

Plasma concentrations of praziquantel during the therapy of neurocysticerosis with praziquantel, in the presence of antiepileptics and dexamethasone.

Plasma praziquantel concentrations were measured in 11 Thai patients with active neurocysticercosis (8 males and 3 females). Praziquantel (B … ailability of the drug observed in this group of patients could be a consequence of pharmacokinetic drug interactions of the concomitant therapy with antiepileptic drugs and dexamethasone. Active metabolite(s), rather than praziquantel itself, may play a significant part in the therapy of neurocysticerosis.

Prioritization of Trypanosoma brucei editosome protein interactions interfaces at residue resolution through proteome-scale network analysis.

BACKGROUND: Trypanosoma brucei is the causative agent for trypanosomiasis in humans and livestock, which presents a growing challenge due to drug resistance. While identifying novel drug targets is vital, the process is delayed due to a lack of functional information on many of the pathogen's proteins. Accordingly, this paper presents a computational framework for prioritizing drug targets within the editosome, a vital molecular machinery responsible for mitochondrial RNA processing in T. brucei. Importantly, this framework may eliminate the need for prior gene or protein characterization, pot

New Approach for Targeting Small-Molecule Candidates for Intrinsically Disordered Proteins.

Intrinsically disordered proteins (IDPs), such as the Alzheimer's-associated tau protein, pose challenges for conventional drug discovery. This study applied the Informational Spectrum Method for Small Molecules (ISM-SM), a computational technique utilizing electron-ion interaction potentials (EIIPs), to identify potential tau modulators. Characteristic interaction frequencies derived from known ligands and conserved mammalian tau sequences were used to screen DrugBank and the COCONUT natural product database. The screening identified approved drugs previously reported to indirectly influence

Identification of medication-microbiome interactions that affect gut infection.

Most people in the USA manage their health by taking at least one prescription drug, and drugs classified as non-antibiotics can adversely affect the gut microbiome and disrupt intestinal homeostasis1,2. Here we identify medications that are associated with an increased risk of gastrointestinal infections across a population cohort of more than one million individuals monitored over 15 years. Notably, the cardiac glycoside digoxin and other drugs identified in this epidemiological study are sufficient to alter the composition of the microbiome and the risk of infection with Salmonella enterica

Electrophoretic Approaches for Investigating Plant-Derived Compound Interactions With DNA and Proteins.

Plant-derived compounds are increasingly recognized as promising therapeutic agents due to their broad biological activities and structural diversity, making their interactions with DNA and proteins central to understanding mechanisms of action and guiding drug discovery. Electrophoretic techniques probe these interactions by tracking changes in electrophoretic mobility, conformation, complex formation, and biopolymer integrity. This review surveys electrophoretic strategies for characterizing plant-derived compound-DNA and -protein interactions, covering fundamental DNA-binding modes (interca

Utilizing drugs, food, and natural product libraries: A computational approach to targeting PGAM1 in clinical cancer therapy.

Phosphoglycerate mutase 1 (PGAM1) is an important glycolytic enzyme that plays a significant role in cancer metabolism. It catalyzes the conversion of 3-phosphoglycerate (3-PG) to 2-phosphoglycerate (2-PG), facilitating energy release during glycolysis. PGAM1 is often upregulated in various cancers, including breast, lung, and prostate cancers, contributing to tumor growth and progression by coordinating glycolysis and anabolic processes which makes it an appealing target for drug development. In this study, we screened drugs, food, and natural compound libraries against PGAM1 by using structu

Identification of JEV capsid interacting domain(s) with host protein(s): Novel target for developing anti-JEV therapeutics.

UNLABELLED: The Japanese encephalitis virus (JEV) is a mosquito-borne flavivirus. There are no approved drugs for the treatment of JEV infection. AIM: The virus-host protein interactions are essential for viral replication and could be novel drug targets. The JEV capsid protein does not have enzymatic functions; therefore, all functions are mediated by protein-protein interactions. We wanted to develop inhibitors to prevent such interactions. MATERIALS AND METHODS: Our study has identified two functional domains of the capsid protein by competitive peptide binding (P2 and P7) that is crucial

Shape Selective Copper-Based Metallo-Tweezer: Bridging Molecular Recognition and Solubility Enhancement of Natural Therapeutics.

Natural bioactive compounds exhibit remarkable therapeutic potential, particularly in combating chronic inflammatory conditions and associated diseases like cancer. However, their clinical application is often constrained by poor aqueous solubility and limited bioavailability, primarily ascribed to their pronounced self-aggregation propensities driven by hydrophobic and hydrogen bonding interactions. To address this challenge, the study introduces a bisimidazole pyridine-based metallo-tweezer (MRE) as a novel strategy to enhance the solubility of three naturally derived therapeutics─camptothec
