CoREST complex inhibition alters RNA splicing to promote neoantigen expression and enhance tumor immunity.

Epigenetic macromolecular enzyme complexes tightly regulate gene expression at the chromatin level and have recently been found to colocalize with RNA splicing machinery during active transcription; however, the precise functional consequences of these interactions are uncertain. Here, we identify unique interactions of the CoREST repressor complex (LSD1-HDAC1-CoREST) with components of the RNA splicing machinery and their functional consequences in tumorigenesis. Using mass spectrometry, in vivo binding assays, and cryo-EM, we find that CoREST complex-splicing factor interactions are direct a

Polysialylation of Glioblastoma Cells Is Regulated by Autophagy Under Nutrient Deprivation.

Glioblastoma (GBM) is a highly aggressive brain tumor marked by invasive growth and therapy resistance. Tumor cells adapt to hostile conditions, such as hypoxia and nutrient deprivation, by activating survival mechanisms including autophagy and metabolic reprogramming. Among GBM-associated changes, hypersialylation, particularly, the aberrant expression of polysialic acid (PSA), has been linked to increased plasticity, motility, and immune evasion. PSA, a long α2,8-linked sialic acid polymer typically attached to the NCAM, is abundant in the embryonic brain and re-expressed in cancers, correla

Assessment of lncRNA biomarkers based on NETs for prognosis and therapeutic response in ovarian cancer.

Ovarian cancer (OC) usually progresses rapidly and is associated with high mortality, while a reliable clinical factor for OC patients to predict prognosis is currently lacking. Recently, the pathogenic role of neutrophils releasing neutrophil extracellular traps (NETs) in various cancers including OC has gradually been recognized. The study objective was to determine whether NETs-related biomarkers can be used to accurately predict the prognosis and guide clinical decision-making in OC. In this study, we utilized univariate and multivariate Cox regression to identify key prognostic features a

The Tumor-Associated Neutrophils-Related Signatures Predict Prognosis and Indicate Immune Landscape in Colorectal Carcinoma.

Background: Tumor-associated neutrophils (TANs), a major immune cell type in the tumor microenvironment (TME), are associated with antitumor or pro-tumor phenotypes. A wealth of evidence evidences demonstrated that the content of TANs has been implicated in the pathogenesis of cancer and may be ideal targets for cancer therapy. In this study, we identified TANs-related genes to construct a prognostic model and evaluated its potential as a biomarker for predicting prognosis and immunotherapeutic response in colorectal carcinoma (CRC). Methods: Kaplan-Meier survival curves were used to assess th

CPSF1 inhibition promotes widespread use of intergenic polyadenylation sites and impairs glycolysis in prostate cancer cells.

Localized prostate cancer can be cured by radiation or surgery, but advanced prostate cancer continues to be a clinical challenge. Altered alternative polyadenylation occurs in numerous cancers and can downregulate tumor-suppressor genes and upregulate oncogenes. We found that the cleavage and polyadenylation specificity factor (CPSF) complex factor CPSF1 is upregulated in patients with advanced prostate cancer, with high CPSF1 expression correlating with worse progression-free survival. Knockdown of CPSF1 selectively inhibited the growth of prostate cancer cells and reduced glycolytic output.

xCT as a potential marker for neuroendocrine cells in high-risk prostate cancer and the relation to AL122023.1-miR-26a/30d/30e axis.

Prostate cancer is the second most common type of cancer in male worldwide. Stromal-epithelial interaction is thought to have a major impact on cancer development and progression. Previous studies have shown that interaction via soluble factors lead to a reduction in the expression of xCT and AL122023.1 in the prostate carcinoma cell line LNCaP after seven days of co-culture with primary stromal p21 cells. In this study, we validated the repression of xCT and AL122023.1 at RNA level using quantitative real-time PCR and at protein level by Western Blotting. Furthermore, xCT is known to be a put

Discovery of lncRNA-Based ProsRISK Score in Serum as Potential Biomarkers for Improved Accuracy of Prostate Cancer Detection.

Circulating lncRNAs have emerged as promising biomarkers for the diagnosis of various cancers. This study aimed to establish an accurate risk prediction model based on serum lncRNAs to facilitate the detection of prostate cancer (PCA). RT-qPCR was used to analyse the levels of candidate lncRNAs, and four lncRNAs (NEAT1, ARLNC1, FOXP4-AS1 and DSCAM-AS1) were identified to be differently expressed in serum from 190 PCA patients, 140 benign controls, and 170 healthy controls. A ProsRISK score based on four lncRNAs and prostate-specific antigen (PSA) was established in the training set. ROC analys

Identification of protein-coding genes associated with metastatic prostate cancer.

Prostate cancer (PCa) is a significant cause of male mortality worldwide. Since metastases are the underlying cause of lethality, identifying markers for metastatic potential would be highly valuable. To address this issue, we set out to identify protein-coding genes with metastasis-specific expression changes in PCa. We employed a previously reported unique cohort consisting of metastases from castration-resistant prostate cancer (mCRPC) and matching primary tumors. Our comprehensive gene expression analyses identified 85 differentially expressed genes (DEGs) associated specifically with mCRP

Regulation of 15-PGDH by ACSM1 in modulating PGE2 signaling and ECM remodeling in prostate cancer.

Prostate cancer remains one of the most common and aggressive cancers worldwide, but the molecular mechanisms underlying its progression are not yet fully understood. The ACSM1 gene is intimately associated with the development of prostate cancer. Here we demonstrated that down-regulation of ACSM1 expression in LNCaP prostate cancer cells significantly inhibits cell proliferation and migration, while overexpression enhances these processes in normal cells. Interestingly, the subcellular localization shift of ACSM1 to cytoplasm coincides with its interaction with 15-PGDH. Overexpression of ACSM

Tumor transcriptome-wide expression classifiers predict treatment sensitivity in advanced prostate cancers.

Advanced prostate cancers respond to hormone therapy but outcomes vary and no predictive tests exist for informed treatment selection. To identify novel biomarker-treatment pairings, we examined associations between biological pathways and 14-year survival outcomes of patients randomized in practice-changing phase 3 trials (testing docetaxel or abiraterone). We included transcriptome-wide expression signatures and immunohistochemistry markers (Ki-67 and PTEN) on prostate tumors from 1,523 patients (832 metastatic). Tumor androgen receptor signaling is associated with longer survival, whereas i

Combined SNPs sequencing and allele specific proteomics capture reveal functional causality underpinning the 2p25 prostate cancer susceptibility locus.

Genome wide association studies (GWASs) have identified numerous risk loci associated with prostate cancer, yet unraveling their functional significance remains elusive. Leveraging our high-throughput SNPs-seq method, we pinpointed rs4519489 within the multi-ancestry GWAS-discovered 2p25 locus as a potential functional SNP due to its significant allelic differences in protein binding. Here, we conduct a comprehensive analysis of rs4519489 and its associated gene, NOL10, employing diverse cohort data and experimental models. Clinical findings reveal a synergistic effect between rs4519489 genoty

A microRNA CRISPR screen reveals microRNA-483-3p as an apoptotic regulator in prostate cancer cells.

The development of traditional protein-targeted cancer therapies is a slow and arduous process, often taking years or even decades. In contrast, RNA-based therapies targeting crucial microRNA (miRNA) offer a faster alternative due to the sequence-specific nature of miRNA inhibitor binding. This, combined with the capacity of individual miRNA to influence multiple cellular pathways, makes these small RNA attractive targets for cancer therapy. While miRNA are known to be dysregulated in prostate cancer (PCa), identifying their individual contributions to disease progression and the identificatio

Deep Transfer Learning Links Benign Glands to Prostate Cancer Progression via Transcriptomics.

The field effect describes the phenomena where environmental exposures, infection, and genetic predisposition result in molecular changes in cells that predispose them to developing cancer. Though this is a well-established concept in pathology, it remains underexplored in the context of high-resolution omics. We utilized the Diagnostic Evidence Gauge of Single Cells (DEGAS) deep transfer learning framework to analyze prostate cancer spatial transcriptomics to identify cells and tissues that are highly associated with cancer progression. DEGAS highlighted morphologically benign glands with red

TRIM29 promotes bladder cancer invasion by regulating the intermediate filament network and focal adhesion.

Bladder cancer is a common malignancy whose lethality is determined by invasive potential. We have previously shown that TRIM29, also known as ATDC, is transcriptionally regulated by TP63 in basal bladder cancers where it promotes invasive progression and metastasis, but the molecular events which promote invasion and metastasis downstream of TRIM29 remained poorly understood. Here we identify stimulation of bladder cancer migration as the specific role of TRIM29 during invasion. We show that TRIM29 physically interacts with K14+ intermediate filaments which, in turn, regulates focal adhesion
