Development of ELISA based on Bacillus anthracis capsule biosynthesis protein CapA for naturally acquired antibodies against anthrax.

Anthrax is a zoonotic disease caused by the gram-positive spore-forming bacterium Bacillus anthracis. Detecting naturally acquired antibodie … protein CapA antigen of B. anthracis, which is non-cross-reactive to vaccine-induced antibodies in horses. Using in silico analyses, we screened coding sequences encoded on pXO2 plasmid, which is absent in the veterinary vaccine strain Sterne 34F2 but present in virulent strains of B. anthracis. Among the 8 selected antigen candidates, capsule biosynthe …

MODIFIED CULTURE METHODOLOGY FOR SPECIFIC DETECTION OF BLASTOCYSTIS HOMINIS IN STOOL SAMPLES.

Blastocystis hominis provides major challenges for laboratory diagnosis due to its polymorphic nature in wet mounts which can result in confusion with other protozoa, yeast or even fat globules. Studies revealed that simple smears were less sensitive than in vitro cultivation using different media for the detection of B. hominis in stool specimens. Cultures of B. hominis are usually enriched by different types of sera to enhance growth and multiplication of the parasite. The aim of this study is to assess the use of two sera types other than horse serum that is commonly used in culture media f

Current joint therapy usage in equine practice: a survey of veterinarians 2009.

REASONS FOR PERFORMING STUDY: Medications are frequently employed to treat intra-articular (IA) problems in the performance horse. Actual usage of the different IA medications in horses is not available. OBJECTIVES: To determine the most common usage of these medications, members of the American Association of Equine Practitioners (AAEP) were surveyed. METHODS: An email link to an online survey was electronically sent to 6305 AAEP members and the responses tabulated and analysed with a logistic regression model. RESULTS: A total of 831 survey responses were submitted and tabulated. Eighty p…

A musculoskeletal model of the equine forelimb for determining surface stresses and strains in the humerus--part I. Mathematical modeling.

Knowledge of the forces that act upon the equine humerus while the horse is standing and the resulting strains experienced by the bone is useful for the prevention and treatment of fractures and for assessing the proximolateral aspect of the bone as a site for obtaining autogenous bone graft material. The first objective was to develop a mathematical model to predict the loads on the proximal half of the humerus created by the surrounding musculature and ground reaction forces while the horse is standing. The second objective was to calculate surface bone stresses and strains at three cross se

Assessing the effects of ex vivo hormonal exposure on oxidative responses in equine leukocytes: A preliminary study.

Breed differences exist between horses and ponies in circulating concentrations of several hormones, notably ACTH and insulin. These hormones regulate stress and metabolic responses, but in other species, they also impact leukocyte oxidant responses. The effects of these hormones on equine leukocytes have not been evaluated to date. If equine leukocytes are similarly regulated, breed differences in increased plasma hormone concentrations or altered sensitivity to them at the leukocyte level could result in breed-related differences in oxidant responses or oxidative status. The objective of thi…

Autologous bone marrow mesenchymal stromal cells for regeneration of injured equine ligaments and tendons: a clinical report.

The use of Mesenchymal Stromal Cells (MSCs) in orthopedic practice has recently and rapidly acquired an important role. Therapies based on the use of MSCs for the treatment of acute injuries as well as chronic inflammatory disorders are gradually becoming clinical routine. These cells have demonstrated intriguing therapeutic potentialities (i.e.: inflammation control, tissue regeneration and pathological scar prevention), that have been taken into consideration for use in both human and veterinary medicine. In particular, horses represent high performance athletes considered models for human p…

Polysaccharide Storage Myopathy.

Type 1 Polysaccharide Storage Myopathy (PSSM1) is an autosomal dominant glycogen storage disorder affecting more than 20 breeds of horses that can present with a variety of signs, including exertional rhabdomyolysis (ER). It is diagnosed by genetic testing or muscle biopsies containing muscle fibers with abnormal amylase-resistant polysaccharide. Type 2 PSSM has recently been subdivided. PSSM2-ER is a glycogen storage disorder identified in Quarter Horses that causes ER and is diagnosed by muscle biopsy as its genetic basis is unknown. Both PSSM1 and PSSM2-ER respond well to a low nonstructura…

Approach to Immune-mediated Ocular Diseases.

Immune-mediated ocular inflammation is a common clinical diagnosis reached for horses with keratitis and uveitis. This diagnosis is made as a diagnosis of exclusion following a thorough effort to rule out an underlying cause for the inflammation, most importantly infectious and neoplastic disease. Practically, response to ophthalmic and systemic anti-inflammatory or immunomodulatory medications is used to support a diagnosis of immune-mediated ocular inflammation; however, such medications are often contraindicated in the face of infection or neoplasia. This article will summarize our current …

Heterobilharzia americana infection in a Grant's zebra (Equus burchelli boehmi).

Heterobilharzia americana is a trematode parasite (family: Schistosomatidae) of mammalian wildlife that occasionally infects domestic dogs and horses in the southeastern United States. This report presents the first case of H. americana infection in a Grant's zebra (Equus burchelli boehmi). The parasite was diagnosed post mortem as an incidental finding by histopathologic identification of H. americana eggs in the liver parenchyma after the zebra succumbed in an accidental traumatic death. Molecular analysis of a portion of the parasite small subunit ribosomal RNA gene demonstrated 100% identi…

Dental sepsis.

Dental sepsis or periapical abscess formation constitutes a large percentage of dental conditions that afflict horses. Dental sepsis occurs when the pulp chamber of the tooth is exposed to the oral cavity or external environment, allowing bacterial localization with resulting infection. Although acute, primary, septic pulpitis in horses is rare, dental sepsis often results from colonization of the pulp chamber with pathogenic bacteria secondary to maleruption or impaction of teeth with secondary alveolar bone lysis, primary fractures of the tooth, mandible, or maxilla, periodontal disease, or …

Equine pulmonary disease: a case control study of 300 referred cases. Part 3: Ancillary diagnostic findings.

The majority of 270 adult horses with chronic pulmonary diseases had excessive volumes of respiratory secretions (RS) present in their trachea, elevated tracheal RS neutrophil ratios and lowered arterial oxygen partial pressures. Some control horses, had inexplicably elevated trachael RS neutrophil ratios. Only the chronic obstructive pulmonary disease (COPD) affected group had significantly elevated bronchoalveolar lavage fluid (BALF) neutrophil ratios. Maximal intrapleural pressure changes (max dPpl) were raised in 48% of COPD cases and rarely with other pulmonary disorders. Arterial pH or c…

Suspected red maple (Acer rubrum) toxicosis with abortion in two Percheron mares.

Horses develop severe and often fatal hemolytic anemia after ingesting dried leaves from red maple (Acer rubrum) trees. Toxicosis appears related to an unknown oxidant present in the dried or wilted leaves. This case report describes 2 horses that aborted and developed fatal hemolytic anemia after consuming wilted leaves from red maple (Acer rubrum). While an absolute diagnosis was not confirmed due to lack of proper antemortem and postmortem examinations, red maple toxicosis appeared a reasonable diagnosis based on clinical signs and laboratory findings. Other differentials include equine inf…

Epidemiology and pathology of a tetrachlorodibenzodioxin poisoning episode.

We describe epidemiological and laboratory investigation of a poisoning outbreak that involved three riding areas and killed fifty-seven horses, five colts, and numerous other animals. The outbreak was traced to the spraying of the areas with salvage oil contaminated with tetrachlorodibenzodioxin, trichlorophenol, and polychlorinated biphenyls. The contamination of the salvage oil resulted from the improper disposal of toxic industrial waste. In the tissues of the horses available for study, the most prominent lesion was a centrilobular fibrosis of the liver that arose in the wall of the centr…

Neurofibromatosis-I Presentating with Multiple Spinal and Intracranial Neurofibromas.

Von Recklinghausen's neurofibromatosis (NF-1) is a phacomatosis characterised by widespread nervous system tumours with cutaneous manifestations and variably associated anomalies. We report here a case, who, in addition to classical features of NF-1 (café-au-lait spots, cutaneous and subcutaneous neurofibromas) demonstrated radiological evidence of both spinal and intracranial neurofibromas and an incidentally discovered horse-shoe kidney. The unique constellation of spinal and intracranial neurofibromas, with associated horse-shoe kidney.

[Development and study of properties of immunoglobulins against Lassa fever].

A horse may serve the producer of immune antiserum to Lassa virus. Specific immunoglobulin with at least 1:512 titer of virus-neutralizing antibodies to Lassa fever was obtained by alcohol sedimentation after Cohn from the blood serum of immunized horses. The preparation does not differ from heterologous commercial immunoglobulins. Preclinical studies of immunoglobulin to Lassa fever demonstrated its safety and a high specific activity. The agent can be injected both alone and in combination with virasole.

Detection of exercising ectopic atrial and ventricular beats using non-linear analysis of clinically normal racehorse electrocardiograms at rest or low-intensity exercise.

Cardiac arrhythmias are common in healthy athletic horses and may lead to poor athletic performance or exercise-associated sudden death. Early detection of high-risk horses is an important goal of cardiovascular diagnostics. We hypothesised that non-linear analysis of electrocardiogram disorderliness can be used to identify horses exhibiting intermittent ectopic atrial and ventricular heart rhythm abnormalities at exercise using brief, artifact-free recordings of normal sinus rhythm electrocardiograms collected at submaximal heart rates. In a convenience prospective cross-sectional study, ambu…

ER-associated degradation pathway protein SEL1L plays an evolutionarily conserved role in platelet adhesion.

SEL1L is a well-known protein in the ER-associated degradation (ERAD) pathway. While it is known to be expressed in platelets, SEL1L has never been shown to play an active role. Here, we present evidence that SEL1L regulates platelet function. We first identified SEL1L through the study of Atypical Equine Thrombasthenia (AET), an autosomal recessive platelet disorder found in thoroughbred horses. A missense variant in SEL1L (c.1810A>G p.Ile604Val) was found in AET-affected horses, which we show is associated with decreased protein expression. SEL1L is intracellular in equine platelets and loca…
