Expression and Purification of Integral Membrane Enzymes for Functional and Structural Studies.

Enzymes that reside in the membrane are responsible for a myriad of vital tasks such as assembly of and changes to the membrane itself, and modification of proteins that reside therein or are secreted through the various cellular compartments. Understanding how these enzymes work at a molecular level is most often dependent on high resolution structural information. While the rates of success for the determination of structures of integral membrane proteins in general, enzymes included, have improved considerably-most notably because of recent advances in single particle cryo-electron microsco

Plasma membrane order maps functional diversity in immune cells.

Cell membranes undergo biophysical remodeling as an adaptation to the surroundings and to perform specific biological functions. However, the extent and relevance of such changes in human immune cells remain unknown, largely because of the lack of single-cell and multidimensional methodologies. Here we apply a cytometry-based method to fill this gap by combining biophysical profiling with simultaneous analysis of immune cell markers. This platform reveals notable cell-type-dependent plasma membrane order heterogeneity in immune cells. By sorting immune cells according to their membrane order a…

Shedding light on the proteolytically released ectodomains of membrane-bound Semaphorins.

Semaphorins (Semas) are secreted and membrane-bound molecules classically characterized as axonal guidance cues in nervous system development. However, it is now widely appreciated that Semas have essential functions in the development of many systems including the cardiovascular, endothelial, and immune systems. Secreted Semas enable functionality distant from the site of release, while membrane-bound Semas mediate localized contact-dependent signaling. Interestingly, functional soluble ectodomains have been discovered in each of the four vertebrate membrane-bound classes which can drive chan

Heat-triggered phospholipid flipping stabilizes plasma membrane fluidity.

Cells must rapidly counteract heat stress-induced hyperfluidization of the plasma membrane to prevent membrane damage1,2, yet how cells achieve such early protection remains unknown. Here we show that in rice (Oryza sativa), the P4-ATPase OsALA5, together with its β-subunit OsALIS2, mediates a heat-responsive flipping of saturated phosphatidylcholines that rapidly stabilizes plasma membrane fluidity. Using leaflet-resolved lipidomics and complementary transport assays, we demonstrate that heat exposure induces a minute-timescale shift in OsALA5 transport activity that leads to selective enrich

Complementary and alternative medicine-associated membranous nephropathy: a systematic review.

BACKGROUND: Complementary and alternative medicine use can lead to the development of membranous nephropathy (MN). The true prevalence and etiologies of complementary and alternative medicine-induced MN remain likely underestimated. The purpose of this review was to report on the incidence of MN induced by various types of complementary and alternative medicine, their clinical presentation, and treatment responses. METHODS: The study was conducted in accordance with the Preferred Reporting Items for Systematic Reviews (PRISMA) guidelines. We performed searches in MEDLINE, Embase, PubMed, the

Extracorporeal Membrane Oxygenation for Severe Burn Injury: A Multidisciplinary Roundtable.

Extracorporeal membrane oxygenation (ECMO) has been used to successfully minimize, replace, or avoid the use of mechanical ventilation in burned and nonburned populations. Further research on ECMO in the burned population is warranted, particularly given the insufficient patient numbers reported in the literature. A panel composed of national leaders in ECMO and/or burn injury discussed the use of ECMO in patients with burn injuries as part of the MedStar Washington Hospital Center Burn Center Multidisciplinary Lecture Series. Panelists examined current methods, institutional practices, and cl

Emerging roles of progesterone receptor membrane components in pregnancy and parturition.

Progesterone receptor membrane components 1 and 2 (PGRMC1 and PGRMC2) are single-pass proteins that function as multi-ligand regulators. They integrate signals from progesterone (P4), heme, and cytochrome P450 enzymes (CYPs). Accumulating evidence implicates PGRMCs in non-genomic progesterone signaling in cell, cancer, and reproductive biology. Heme binding (through their heme binding domain) and cytochrome P450 enzymes (CYPs) binding provide distinct functional roles for PGRMCs in various cells under specific cellular environment. In reproductive tissues, multiple functional roles have been r

New Biochemical Insights into RIT GTPases Regulation and Membrane Interactions.

Both RIT1 and RIT2 are members of the RAS superfamily of small GTPases, which regulate various cellular processes. RIT1 is widely expressed, whereas RIT2 is primarily found in neuronal tissues. Dysregulation of these proteins has been associated with several human diseases, including Noonan syndrome, cancer, Parkinson's disease, autism, and schizophrenia. Although RIT1 and RIT2 are often compared to classical RAS proteins, they exhibit distinct regulatory and biochemical properties. Here, we demonstrate that RIT1 differs from classical RAS in GTPase cycling. Unlike classical RAS proteins, RIT1

Coexistence of ANCA-associated glomerulonephritis and membranous nephropathy: a scoping review.

BACKGROUND: Coexistence of anti-neutrophil cytoplasmic antibody -associated glomerulonephritis (ANCA-GN) and membranous nephropathy (MN) is rare. This scoping review mapped evidence on clinicopathologic features, phenotypic heterogeneity, and reporting gaps relevant to future rigorous phenotype-specific comparative studies. METHODS: Comprehensive searches of PubMed, Web of Science, and CENTRAL from inception to April 16, 2026 identified original studies of patients with coexisting ANCA-GN and MN. Two reviewers independently extracted study and participant characteristics, renal and systemic f

Early Outcomes of Extracorporeal Membrane Oxygenation in Congenital Heart Surgery.

OBJECTIVES: Extracorporeal membrane oxygenation is widely used to treat cardiopulmonary failure after congenital heart surgery. This study evaluated patient characteristics, outcomes, and risk factors associated with perioperative veno-arterial extracorporeal membrane oxygenation use. METHODS: All patients undergoing congenital heart surgery between 2001 and 2024 at our centre were included. Prevalence, outcomes, risk factors for extracorporeal membrane oxygenation support, and predictors of in-hospital mortality following extracorporeal membrane oxygenation were analysed. RESULTS: Among 989

Electrostatic control of membrane disruption and amorphous coaggregation by dynorphin A variants.

Dynorphin A is a highly cationic neuropeptide that exhibits membrane activity beyond its canonical opioid receptor signaling. Here, we investigate how sequence variants of Dynorphin A associated with spinocerebellar ataxia type 23 modulate membrane disruption and aggregation behavior. Using computational electrophysiology simulations combined with liposome leakage and fluorescence-based aggregation assays, we show that Dynorphin A variants interact with lipid bilayers primarily via electrostatic recruitment, followed by mutation-dependent insertion and transient pore formation. Anionic lipids

Membrane properties modulate methane oxidation by particulate methane monooxygenase.

The copper-dependent membrane monooxygenases particulate methane monooxygenase (pMMO) and ammonia monooxygenase (AMO) oxidize methane to methanol and ammonia to hydroxylamine, respectively. These enzymes, which are important targets for biotechnology, reside in intracytoplasmic membranes (ICMs) where they form densely packed hexagonal arrays. While cryoEM structures of pMMO and AMO in ICMs have revealed closely associated lipids, little is known about how specific lipids and membrane morphologies influence activity. Here we show through cryoelectron tomography (cryoET) that three species of me…

Evaluation of Neuromuscular Blocking Agent Utilization in Extracorporeal Membrane Oxygenation.

Neuromuscular blocking agent (NMB) utilization has been infrequently described in patients requiring extracorporeal membrane oxygenation (ECMO). The goal of this analysis was to evaluate NMB utilization at a high-volume ECMO center. This was a retrospective cohort study of patients who required ECMO at the Cleveland Clinic between January 2022 and December 2023. NMB utilization was collected, including the agent, dose, duration (if continuous infusion), and indication. Other clinical outcomes recorded included duration of ECMO support, evidence of neuromuscular dysfunction, and hospital mortal

Diagnostic and prognostic signatures of glomerular membrane dysregulation in immune nephropathies.

BACKGROUND: Immunonephropathy, encompassing disorders such as anti-neutrophil cytoplasmic antibody (ANCA)-associated vasculitis (AAV), focal segmental glomerulosclerosis (FSGS), minimal change disease (MCD), and membranous nephropathy (MN), is characterized by immune-mediated glomerular injury leading to progressive renal dysfunction. Despite advances in clinical characterization, the precise molecular mechanisms underlying glomerular damage remain poorly understood. METHODS: Gene expression profiles from the Gene Expression Omnibus (GEO) database were analyzed to identify plasma membrane hom

Acetylcholinesterase immobilization on a natural thin membrane for stable biocatalyst design.

This study involved the immobilization of acetylcholinesterase (AChE) onto a natural thin membrane extracted from the inner epidermal layer of Allium cepa, with the aim of enhancing the enzyme's stability, activity, and reusability for potential biotechnological applications. The membrane, composed of biocompatible polymers and naturally occurring functional groups, was used as a sustainable support in a combined strategy involving adsorption followed by glutaraldehyde-mediated cross-linking. Immobilization conditions were optimized by evaluating key factors such as enzyme concentration, membr

Emerging biomarkers in IgA nephropathy, membranous nephropathy, and lupus nephritis.

INTRODUCTION: Diabetic kidney disease (DKD) studies have identified circulating proteins linked to progression to kidney function decline. Whether these markers generalize to autoimmune nephritides is unclear. We evaluated DKD-derived biomarkers in IgA nephropathy (IgAN), membranous nephropathy (MN), and lupus nephritis (LN) for associations with renal pathology and longitudinal kidney function decline. METHODS: In a single-center cohort of biopsy-proven IgAN (n = 82), MN (n = 22), and LN (n = 22) with healthy controls (n = 88), serum was obtained immediately before renal biopsy. Seven protei

Contribution of Proteins to Membrane and Cell Wall Structures in Mycobacterium tuberculosis.

The cell wall of Mycobacterium tuberculosis (Mtb) is an exceptionally complex, multilayered structure that is fundamental to the bacterium's survival, pathogenicity, and resistance to both host immune defenses and antibiotics. This cell envelope consists of several distinct components: a surface capsule, an outer membrane, and a peptidoglycan layer that is covalently attached to arabinogalactan. The arabinogalactan is further esterified with mycolic acids, forming the characteristic mycolyl-arabinogalactan-peptidoglycan (mAGP) complex, which is anchored to the underlying plasma membrane. The a
