MRI-based estimation of physiologically consistent head conductivity and its impact on depth-dependent electric field distributions in transcranial magnetic stimulation.

Objective. Accurate estimation of spatially heterogeneous conductivity distributions is essential for reliable electric field modeling in non-invasive brain stimulation. Existing deep learning-based approaches tend to produce conductivity distributions converging toward uniform values, failing to capture physiologically plausible intra-tissue variability or MRI-conductivity relationships. This limitation restricts the ability to resolve depth-dependent electric field characteristics that are relevant for understanding the mechanisms of neural activation. Here, we propose a novel framework for

Multimodal Categorical and Dimensional Approaches to Understanding Threat Conditioning and Its Extinction in Individuals With Anxiety Disorders.

IMPORTANCE: The Research Domain Criteria project of the National Institute of Mental Health aims to guide neuropsychiatry toward precision medicine. Its inception was partly in response to the overlap of clinical manifestations between different DSM-IV diagnoses within a category. For example, anxiety disorders comprise a DSM-IV category that includes diagnoses that differ from each other but are all characterized by dysregulated fear levels. Whether DSM-IV-based and Research Domain Criteria-based analytic approaches provide distinct or similar information with regard to the fear circuitry of

Loss of Mecp2 in substantia nigra dopamine neurons compromises the nigrostriatal pathway.

Mutations in the methyl-CpG-binding protein 2 (MeCP2) result in Rett syndrome (RTT), an X-linked disorder that disrupts neurodevelopment. Girls with RTT exhibit motor deficits similar to those in Parkinson's disease, suggesting defects in the nigrostriatal pathway. This study examined age-dependent changes in dopamine neurons of the substantia nigra (SN) from wild-type, presymptomatic, and symptomatic Mecp2(+/-) mice. Mecp2(+) neurons in the SN in Mecp2(+/-) mice were indistinguishable in morphology, resting conductance, and dopamine current density from neurons in wild-type mice. However, the

Effects of repetitive scanning ultrasound on the intraneuronal dendritic signalling of CA1 pyramidal neurons.

INTRODUCTION: Neural development is regulated by several spatiotemporally changing factors, which are essential for enabling neurons to develop functional networks, during early developmental stages. Additionally, external physical stimuli, like scanning prenatal ultrasound (US) examination, may influence neural development. Aim of our study is to examine potential consequences of diagnostic level ultrasound exposure during the early phase of hippocampal CA1 neural network development by using computational models. METHODS: Using in silico modelling, this study explores and compares multiple

Altered cognitive performance is associated with auditory cortex functional connectivity in patients with unilateral sensorineural hearing loss.

Sensorineural hearing loss (SNHL) has been suggested to be closely associated with cognitive decline, yet the underlying neural mechanisms remain largely unclear. This study sought to explore the potential association between cognitive function and auditory cortex functional connectivity in patients with unilateral sensorineural hearing loss (USNHL) and investigate the possible neural mechanisms underlying subtle cognitive decline. Sixty patients with moderate-to-severe USNHL (disease duration ≥3 years; 30 with right-sided USNHL and 30 with left-sided USNHL) and 30 age-matched normal-hearing c

A physiologically inspired meta-pattern generator bridging intention and motor primitives for human voluntary locomotion.

Understanding the hierarchical organization and transmission characteristics of neural signals-from motor intention, through spinal integration, to lower limb joint motions-is critical for developing physiologically plausible, intention-driven motion generation models. Such models not only advance our comprehension of the neurophysiological mechanisms underlying lower limb locomotion but also provide a biological foundation for the direct neural control of lower limb prostheses and rehabilitation robots, offering both theoretical significance and practical utility. This challenge has persisted

Multispectral brain connectivity during visual attention distinguishes controlled from uncontrolled hypertension.

Hypertension-related changes in brain function place individuals at higher risk for cognitive impairment and Alzheimer's disease. The existing functional neuroimaging literature has identified important neural and behavioural differences between normotensive and hypertensive individuals. However, previously-used methods (i.e. magnetic resonance imaging, functional near-infrared spectroscopy) rely on neurovascular coupling, which is a useful but indirect measure of neuronal activity. Furthermore, most studies fail to distinguish between controlled and uncontrolled hypertensive individuals, who

The interaction of oxytocin and nicotine addiction on psychosocial stress: an fMRI study.

The anxiolytic effect of oxytocin (OXT) on psychosocial stress has been well documented, but the effectiveness under the interference of other factors still requires in-depth research. Previous studies have shown that nicotine addiction interacts with OXT on psychosocial stress on the behavioral level. However, the underlying neural mechanism of interaction between OXT and nicotine addiction on psychosocial stress has not been examined, and we conducted two experiments to reveal it. Firstly, after intranasal administration of randomized OXT or placebo (saline), a group of healthy participants

Prenatal opioid exposure: The next neonatal neuroinflammatory disease.

The rates of opioid use disorder during pregnancy have more than quadrupled in the last decade, resulting in numerous infants suffering exposure to opioids during the perinatal period, a critical period of central nervous system (CNS) development. Despite increasing use, the characterization and definition of the molecular and cellular mechanisms of the long-term neurodevelopmental impacts of opioid exposure commencing in utero remains incomplete. Thus, in consideration of the looming public health crisis stemming from the multitude of infants with prenatal opioid exposure entering school age,

Optical stimulation of auditory neurons: effects of acute and chronic deafening.

In developing neural prostheses, particular success has been realized with cochlear implants. These devices bypass damaged hair cells in the auditory system and electrically stimulate the auditory nerve directly. In contemporary cochlear implants, however, the injected electric current spreads widely along the scala tympani and across turns. Consequently, stimulation of spatially discrete spiral ganglion cell populations is difficult. In contrast to electrical stimulation, it has been shown that extremely spatially selective stimulation is possible using infrared radiation (e.g. [Izzo, A.D., S

The state of motor neural networks in low back pain: a systematic review and meta-analysis.

Low back pain (LBP) is one of the most prevalent musculoskeletal disorders and often shows poor recovery. Although symptom persistence is multifactorial, trunk motor control changes are frequently observed and suggested to result from reorganisation of motor neural networks. Understanding these mechanisms may help guide more effective interventions. This systematic review with meta-analysis (PROSPERO: CRD42023417238) aims to compare motor neural network state between individuals with LBP and pain-free controls, and under experimental LBP conditions. Four databases (CINAHL, Medline, Embase, Web

Amantadine modulates novel macrophage phenotypes to enhance neural repair following spinal cord injury.

BACKGROUND: Spinal cord injury (SCI) triggers a complex inflammatory response that impedes neural repair and functional recovery. The modulation of macrophage phenotypes is thus considered a promising therapeutic strategy to mitigate inflammation and promote regeneration. METHODS: We employed microarray and single-cell RNA sequencing (scRNA-seq) to investigate gene expression changes and immune cell dynamics in mice following crush injury at 3 and 7 days post-injury (dpi). High-dimensional gene co-expression network analysis (hdWGCNA) and slingshot trajectory analysis were employed to identif

Birth prevalence and parental stress associated with neural tube defects in Amhara's public comprehensive specialized hospitals, Ethiopia, 2024.

BACKGROUND: Neural tube defects (NTDs) are severe congenital anomalies resulting from the incomplete closure of the embryonic neural tube, affecting around 300,000 newborns globally each year and leading to significant mortality and disability. While high-income countries have seen a reduction in NTD prevalence, developing nations like Ethiopia continue to face high rates. Families impacted by NTDs often endure emotional challenges, including grief, anxiety, and social isolation. This study aims to investigate the birth prevalence of NTDs and the associated parental stress, emphasizing the wid
