Nutritional and environmental determinants of maturation and disruption of the early life gut microbiome: A narrative review.

The first 1000 days of life represent a critical window for gut microbiome assembly, with lifelong implications for child growth, immune development, and disease risk. This review synthesizes evidence on maternal, perinatal, and especially nutritional factors that influence early-life intestinal colonization and highlights the consequences of microbial disruptions during this period. In utero exposures and birth-associated factors can profoundly shape microbial development, reducing diversity and beneficial taxa. Nutrition exerts a particularly dominant and modifiable influence: breastfeeding

From physiology to food applications: Kluyveromyces marxianus as a key player for innovation in probiotic attributes, fermented foods and functional food ingredients.

Kluyveromyces marxianus is a non-conventional yeast of growing interest in food biotechnology due to rapid growth, thermotolerance, broad substrate use, and metabolic versatility. Current knowledge remains fragmented across genomics, physiology, probiotic traits, metabolite production, and food applications. This review summarizes recent advances in K. marxianus with a focus on its food applications and strain-level variability, highlighting how ecological origin, genomic background, ploidy, lactose-utilization systems, fructan-hydrolyzing capacity, stress responses, and genome-scale metabolic

YB-1 inhibition suppresses osteosarcoma growth and reverses tumor immunosuppression.

Osteosarcoma (OS), the most common primary bone malignancy in adolescents and young adults, is still marked by poor long-term survival and poor mortality. Although immune checkpoint blockade (ICB) has transformed treatment for several cancers, its benefit in OS has been minimal, largely due to OS's "immune-cold" phenotype characterized by scarce tumor-infiltrating lymphocytes. Novel strategies are urgently needed to overcome this resistance. Y-box binding protein 1 (YB-1), a pro-oncogenic member of the cold-shock protein superfamily, represents a promising target to sensitize OS to ICB. Here,

The combined toxic effects of long-term exposure to environmentally relevant concentrations of imidacloprid and chromium on Xenopus laevis tadpoles: Growth, oxidative stress, and molecular mechanisms.

Imidacloprid (IMI) and chromium (Cr), as widely co-occurring pesticide and heavy metal pollutants in freshwater ecosystems, pose potential threats to non-target aquatic organisms. Despite their frequent detection in the environment, systematic investigations into their combined toxicity mechanisms remain limited, particularly in ecologically significant amphibians. Therefore, in this study, the mechanisms of single and combined toxicity of IMI and Cr in Xenopus laevis tadpoles were investigated at environmentally relevant concentrations (41.4 μg/L IMI; 54.6 μg/L Cr). The results showed that th

Ketogenic Diet and β-Hydroxybutyrate Suppress Bladder Cancer Growth and Enhance Anti-PD-L1 Immunotherapy Efficacy.

The limited clinical efficacy of immune checkpoint inhibitors (ICIs) remains a major challenge in the treatment of bladder cancer (BCa). Here, we report that a ketogenic diet (KD) and its principal circulating metabolite, β-hydroxybutyrate (β-HB), suppress bladder tumor growth and enhance responses to PD-L1 inhibitor. In a syngeneic MB49 model, KD reduced tumor growth compared with normal diet (ND). KD increased plasma β-HB and was associated with higher intratumoral PD-L1 expression. Exogenous β-HB supplementation under ND recapitulated the antitumor effect of KD, further enhancing anti-PD-L1

Childhood-Onset Lupus Nephritis in the Era of Triple Therapy and Steroid Minimization.

Childhood-onset lupus nephritis (cLN) should no longer be framed as a smaller version of adult lupus nephritis. It is a high-stakes pediatric kidney disease in which immune injury, treatment toxicity, growth, puberty, fertility, adherence, and transition to adult care intersect over decades. Approximately 10-20% of systemic lupus erythematosus begins in childhood, and 40-60% of affected children develop lupus nephritis. Regional cohorts report even higher renal involvement in some populations, including 65-73% among Saudi children with SLE. Contemporary guidance has moved from prolonged high-d

Orally administered biomimetic nanovesicles engineered with FGF2 orchestrate mucosal healing and microbiome remodeling in inflammatory bowel disease.

Inflammatory bowel disease (IBD) is characterized by chronic intestinal inflammation and profound microbial dysbiosis, presenting significant therapeutic challenges. While fibroblast growth factor 2 (FGF2) possesses potent regenerative capabilities, its oral administration is severely hindered by rapid gastrointestinal degradation. To overcome these delivery barriers, this study investigates a novel, targeted therapeutic strategy utilizing nanoscale outer membrane vesicles (OMV/FGF2) naturally secreted during the normal growth of FGF2-engineered Gram-negative bacteria. The isolated OMV/FGF2 (1

Parasitic isopods in fish aquaculture: A review of current knowledge and future perspectives.

Aquaculture has become a dominant contributor to global seafood production, providing essential protein and supporting livelihoods while alleviating pressure on wild fish stocks. However, cultured fish face numerous health challenges, including infestations by parasitic isopods, notably cymothoid and gnathiid species, which cause significant economic losses through reduced growth, compromised health, and mortality. Infestations are usually influenced by host factors, environmental conditions, parasite traits, and aquaculture management practices, including cage design, stocking density, water

Allee Effect and Oncolytic Virotherapy: Different Therapeutic Outcomes for Different Tumour Growth.

Tumour recurrence after oncolytic virotherapy is a critical clinical challenge, as solid cancers tend to persist and spread after such therapies alone. Often, mathematical models consider tumour growth without taking possible cooperative cell behaviours into account. Instead, the well-known Allee effect has the ability to drive low-density tumour dynamics and affect therapeutic outcomes. For this reason, we here explore the contribution of Allee effects into two of the most common cancer growth paradigms, e.g. the logistic and the Gompertz frameworks. Our analysis reveals that density-dependen

Epidermal growth factor receptors unveiled: a comprehensive survey on mutations, clinical insights of global inhibitors, and emergence of heterocyclic derivatives as EGFR inhibitors.

Mutations that overexpress the epidermal growth factor receptor (EGFR) are linked to cancers like breast (15-20%), head and neck (10-15%), colorectal (5-8%), and non-small cell lung cancer (10-50%), especially in East Asian populations. EGFR activation stimulates 'RAS/RAF/MEK/ERK, PI3K/Akt, and MAPK' pathways, which enhance cell division, survival, angiogenesis, and tumour growth while inhibiting apoptosis and metastasis. Secondary mutations (e.g. 'T790M', 'C797S'), off-target effects, and resistance due to alternate pathway activation reduce the efficacy of currently available EGFR inhibitors

In Vitro Hepatocyte Differentiation of Human Adipose-Derived Stem Cells Under Hypoxia and Photobiomodulation Irradiation.

Stem cells may be manipulated in vitro to induce hepatic differentiation. We investigated the effect of hypoxia and photobiomodulation therapy (PBMT) on the hepatogenic differentiation of human adipose-derived stem cells (hASCs). hASCs were exposed to different carbon dioxide concentrations with photobiomodulation (PBM) using low-level light. Cell survival and secretion of hepatocyte growth factor (HGF) of the hASCs were evaluated by immunostaining and Western blot analyses. Hepatic differentiation was assessed via immunocytochemical staining, fluorescence-activated cell sorting, and Western b

Engineered bacterial extracellular vesicles mediate pyroptosis to counteract m6A methylation-based immunosuppression after insufficient radiofrequency ablation of hepatocellular carcinoma.

Insufficient radiofrequency ablation (IRFA) of hepatocellular carcinoma (HCC) leads to alterations in epigenetic properties such as N 6-methyladenosine (m6A) RNA methylation in tumor cells, which creates an immune-suppressive tumor microenvironment capable of promoting residual tumor growth and recurrence and affecting the efficacy of RFA. In this study, the constructed STM-Mn@OMVs, which were produced through the rational functionalisation of bacterial-derived OMVs with Mn2+ ions and the methylation inhibitor STM2457, were found to effectively activate antitumor immunity. Our study shows that

Leprosy reactions: New knowledge on pathophysiology, diagnosis, treatment and prevention.

Leprosy, or Hansen's disease, caused by Mycobacterium leprae and Mycobacterium lepromatosis, is a chronic granulomatous infectious disease. Leprosy reactions, characterised by neurocutaneous inflammation, complicate the disease's indolent course, leading to significant morbidity. However, limited knowledge of reaction pathophysiology stems from a lack of experimental models and the abrupt onset of reactional episodes, posing challenges in delineating initial pathogenic steps. In type 1 reactions, ongoing studies explore the roles of interferon-gamma which results in increased interleukin (IL)-

The Role of Dietary Fatty Acids in Modulating Blue Crab (Callinectes sapidus) Physiology, Reproduction, and Quality Traits in Captivity.

The invasive blue crab is challenging the Mediterranean basin, progressively declining local populations. This reflects a lower prey availability and suitability of dietary nutrients (mainly n-3 polyunsaturated fatty acids, PUFA). The present study aimed to challenge blue crab males and females with a feed source low in n-3 PUFA with respect to one showing a proper fatty acid profile and to investigate the responses in terms of growth, welfare, lipid characterization of target tissues, and reproductive status. Blue crabs were divided into three groups as follows: (i) Marine: crabs fed sardinel

Towards zero excess sludge discharge with built-in ozonation for wastewater biological treatment.

In this study, a biological treatment process, which used a built-in ozonation bypass to achieve sludge reduction, was built to treat the industrial antifreeze production wastewater (mainly composed of ethylene glycol). The results indicated there is a positive correlation between ozone dosage and sludge reduction. At the laboratory level, the MLSS in the system can be stably controlled at around 3400 mg MLSS L-1 under the dosage of 0.18 g O3 g-1 MLSS. Ozonation can increase the compactness of sludge flocs (fractal dimension increased from 1.89 to 1.92). Ozone destroys microbial cell membranes

Tracking the progeny of bacterial persisters using a CRISPR-based genomic recorder.

The rise of antimicrobial failure is a global emergency, and causes beyond typical genetic resistance must be determined. One probable factor is the existence of subpopulations of transiently growth-arrested bacteria, persisters, that endure antibiotic treatment despite genetic susceptibility to the drug. The presence of persisters in infected hosts has been successfully established, notably through the development of fluorescent reporters. It is proposed that infection relapse is caused by persisters resuming growth after cessation of the antibiotic treatment, but to date, there is no direct

An Unusual Endometrial Stromal Neoplasm With JAZF1-BCORL1 Rearrangement.

Endometrial stromal tumors represent the second most common category of uterine mesenchymal tumors. Several different histologic variants and underlying genetic alterations have been recognized, one such being a group associated with BCORL1 rearrangements. They are usually high-grade endometrial stromal sarcomas, often associated with prominent myxoid background and aggressive behavior. Here, we report an unusual endometrial stromal neoplasm with JAZF1-BCORL1 rearrangement and briefly review the literature. The neoplasm formed a well-circumscribed uterine mass in a 50-yr-old woman and had an u

Metagenomics Reveals the Microbiome Multifunctionalities of Environmental Importance From Termite Mound Soils.

The ecological deterioration caused by the continuous and excessive use of synthetic inputs in agriculture has prompted the search for environmentally favorable resources for crop production. Many have advocated for the use of soils from termite mounds to improve soil and plant health; therefore, the purpose of this study was to characterize the microbiome multifunctionalities that are important for plant health and growth in termite mound soil. The metagenomics of soil from termite mounds revealed taxonomic groups with functional potentials associated with promoting the growth and health of p

Melatonin in Health and Disease: A Perspective for Livestock Production.

Mounting evidence in the literature indicates an important role of endogenous and exogenous melatonin in driving physiological and molecular adaptations in livestock. Melatonin has been extensively studied in seasonally polyestrous animals whereby supplementation studies have been used to adjust circannual rhythms in herds of animals under abnormal photoperiodic conditions. Livestock undergo multiple metabolic and physiological adaptation processes throughout their production cycle which can result in decreased immune response leading to chronic illness, weight loss, or decreased production ef

Aggregation of alpha-synuclein in enteric neurons does not impact function in vitro.

Recent evidence implicates a gut-first pathogenesis in the enteric nervous system (ENS) within a portion of PD patients, yet in vitro investigations have primarily focused on the central nervous system. Here, the preformed fibril (PFF) PD model is applied with co-administered groups of butyrate and lipopolysaccharide to model the effects of the local gut microbiome. Significant PFF uptake and retention occur in isolated rat enteric neurons compared to untreated controls resulting in increasing immunostained aggregate conformation-specific, alpha-synuclein (a-Syn) average intensity between 6 µg
