Fatal encephalomyocarditis virus (EMCV) infections in a ring-tailed lemur (Lemur catta), a squirrel monkey (Saimiri sciureus), three mandrills (Mandrillus sphinx), a chimpanzee (Pan troglodytes), a pygmy hippopotamus (Choeropsis liberiensis), and two Goodfellows tree kangaroos (Dendrolagus goodfellowi) occurred at Taronga Zoo. This is the first description of EMCV in a zoological collection outside of the United States. Regardless of species, the most common clinical presentation was sudden death. The gross pathologic changes were diffuse or focal pallor of the myocardium with occasional marke

The production of neutrophil granulocytes is a complex and dynamic process during which a small number of self-renewing stem cells give rise to lineage-specific progenitors. One hematopoietic glycoprotein that appears to specifically control the survival, cycle activation, proliferation and terminal maturation of this myeloid lineage is granulocyte colony-stimulating factor (G-CSF). Some topics discussed are its molecular characterization, biochemical purification, sources, and effects of administration.

A female Bornean orangutan (Pongo pygmaeus) aged 11 years and 6 months was examined by veterinarians after caretakers observed lethargy and facial grimacing. Within 72 h the primate had left-sided hemiparesis that worsened over the next week. An MRI revealed a focal right-sided cerebral mass suspected to be a neoplasm. Ten days after onset of clinical signs, the orangutan died. On postmortem exam, the medial right parietal lobe was replaced by a 7 × 4 × 3.5 cm focus of neuromalacia and hemorrhage that displaced the lateral ventricle and abutted the corpus callosum. Histopathology of the cerebr

Animal models of inherited bleeding disorders are important for understanding disease pathophysiology and are required for preclinical assessment of safety prior to testing of novel therapeutics in human and veterinary medicine. Experiments in these animals represent important translational research aimed at developing safer and better treatments, such as plasma-derived and recombinant protein replacement therapies, gene therapies and immune tolerance protocols for antidrug inhibitory antibodies. Ideally, testing is done in animals with the analogous human disease to provide essential safety i

There are no currently approved treatments for filovirus infections. In this study we report the discovery process which led to the development of antisense Phosphorodiamidate Morpholino Oligomers (PMOs) AVI-6002 (composed of AVI-7357 and AVI-7539) and AVI-6003 (composed of AVI-7287 and AVI-7288) targeting Ebola virus and Marburg virus respectively. The discovery process involved identification of optimal transcript binding sites for PMO based RNA-therapeutics followed by screening for effective viral gene target in mouse and guinea pig models utilizing adapted viral isolates. An evolution of

There is considerable evidence to suggest that adverse early-life experiences have a profound effect on the developing brain. Neurobiological changes that occur in response to untoward early-life stress can lead to lifelong psychiatric sequelae. Children who are exposed to sexual or physical abuse or the death of a parent are at higher risk for development of depressive and anxiety disorders later in life. Preclinical and clinical studies have shown that repeated early-life stress leads to alterations in central neurobiological systems, particularly in the corticotropin-releasing factor system

It has been demonstrated over the last decade that some higher animals can learn sign language and communicate with humans. This finding radically alters some earlier conceptions of animals as being non-sentient, and forces a re-evaluation of the use of higher animals in research. The ethics of using animals are examined in this paper in relation to postulated levels of consciousness, and in relation to experimental design and experimental goals. In particular, an attempt is made to classify experimental situations with regard to potential psychological trauma, and to examine in detail the eth

Infectious disease severity, shedding, and within-host kinetics can vary substantially with exposure dose, inoculation route, and host factors. However, the relative contributions of these variables to infection heterogeneity remain poorly quantified because small sample sizes in controlled in vivo experiments limit their statistical power and scientific scope. Here, by compiling and jointly analyzing the largest published database of nonhuman primate challenge experiments (107 studies; 721 animals), we show that exposure route drives SARS-CoV-2 infection kinetics more strongly than dose, age,

BACKGROUND: The ability to study in vivo microvascular flow within the placenta is limited. We sought to compare two techniques for evaluation of placental perfusion in a translational nonhuman primate model.
METHODS: We measured placental microvascular perfusion in six pregnant African green monkeys (Chlorocebus aethiops sabaeus) using both Laser Doppler Flowmetry (LDF) and contrast-enhanced ultrasound (CEUS) in 2-4 discrete placental locations per animal at early- (EG), mid- (MG), and/or late-gestation (LG). We assessed correlation between matched LDF and CEUS, with the hypothesis that we w

The arcuate fasciculus (af) is a crucial dorsal pathway underpinning human language, yet its weak frontal connectivity in macaques-the standard primate model-creates an evolutionary puzzle. Here, we investigate the common marmoset, a distantly related platyrrhine with high vocal complexity, to test for convergent neural adaptations. By integrating retrograde and anterograde tracing with ultra-high-resolution diffusion MRI, we identified a robust af homolog in marmosets that is anatomically distinct from the superior longitudinal fasciculus. Comparative mapping across marmosets, macaques, chimp

Precise and up-to-date information about animal location and identity allows us to better quantify individual behaviors in studies of neural activity, cognition, and animal health. In socially housed laboratory animals, identification is usually defined by observation or invasive markers, making the data collection time-consuming, variable across experimenters, and disruptive to animals. We established an automatic pipeline for real-time identification of common marmosets in captivity using a close-view camera. It uses the supervised deep-learning YOLOv8 model to localize individuals, detect f

Memory formation relies on the reorganization of neural activity patterns during experience that persist in subsequent sleep. How these processes promote learning while preserving established memories remains unclear. Here we recorded neural ensemble activity from the hippocampal and associated regions in freely moving macaques as they recalled item sequences presented that day ('new'), 1 day earlier ('recent') or more than 2 weeks earlier ('old'). Cell assemblies biased for old sequences showed less drift, greater network connectivity and stronger sleep reactivation than new-biased assemblies

Cerebellar ataxias (CAs) encompass a wide spectrum of genetic and sporadic origins, of which a portion is driven by immune-mediated pathomechanisms. While some patients harbor known autoantibodies, assisting diagnosis and treatment, many remain seronegative. Here, we identify synaptophysin (SYP) as an autoantigen in CA by serum IgG staining on primate tissue, combined with human protein array and cell-based assay (CBA). SYP is abundant in presynaptic vesicles and is transiently exposed on the cell surface. Out of 43 patients with CA with synaptic serum reactivity on cerebellar sections, SYP an

How does the primate brain coordinate plasticity when learning to discriminate new objects? We measured consequences of object learning on inferior temporal (IT) cortex, a key waypoint supporting object recognition in the ventral visual stream, in male macaques. Neural activity in task-trained monkeys' IT showed increased object selectivity, enhanced linear separability across objects, and more object-invariant representations compared to task-naïve monkeys. To model these differences, we developed a computational framework using anatomically-mapped artificial neural network (ANN) models of th

CLN1 Batten disease is caused by mutations in the CLN1 gene which codes for the lysosomal enzyme palmitoyl-protein thioesterase-1 (PPT1). Disease progression is marked by intellectual and motor deterioration, seizures, vision loss, and early mortality. There are no approved treatments for this severe pediatric condition. We describe the development and characterization of recombinant human PPT1 (rhPPT1) suitable for use as a clinical enzyme replacement therapy in CLN1 Batten patients. rhPPT1 displays similar mannose-6-phosphate receptor (M6PR)-dependent uptake kinetics in neuronal cell lines f
