Histological and genetic analysis and risk assessment for chromosomal aberration after ICSI for patients presenting with CBAVD.

Intracytoplasmic sperm injection (ICSI) has opened a new field in the treatment of male infertility, leading to a debate concerning its genetic safety. In this study we present an analysis of 11 patients presenting congenital bilateral absence of the vas deferens (CBAVD). In all 11 cases, genetic counselling, histological analysis of testicular biopsies, cystic fibrosis transmembrane conductance regulator (CFTR) mutation screenings of both partners and spermatozoa three-colour fluorescent in-situ hybridization (FISH) analysis were performed. A total of 31 CFTR mutations were screened and mutat…

In mice, a population of male germ cells show characteristics of nonapoptotic cell death during G0 arrest.

In mammals, a small population of spermatogonial stem cells (SSCs) is established shortly after birth. These cells self-renew and produce sperm for the male's reproductive lifespan, passing the genome on to the next generation. Establishment of a population of SSCs with high genomic integrity is essential. SSCs are derived from a much larger precursor population of male germ cells (MGCs) that differentiate during fetal life. During the last third of gestation, MGCs undergo a prolonged period of G0 cell cycle arrest during which they sustain high levels of transcription and acquire epigenetic p

Study of the effect of breeding programs in dairy production on cryopreserved ram (Ovis aries) sperm parameters of Manchega sheep.

Breeding programs in dairy sheep (Ovis aries) have significantly increased milk production but have led to a decline in other functional aspects such as reproductive efficiency. While including functional traits in these programs has mitigated this decline, enhancing productivity remains the main goal due to its importance for sector sustainability. The present study aims to assess changes in reproductive physiology resulting from increased productivity, particularly focusing on the structural and functional characteristics of sperm that occurred over the last 25 yr, which are closely linked t

Frozen Boar Semen Can Be Successfully Stored at -80°C, Reducing Need for LN2 Storage Containers.

This study explores alternative freezing approaches aimed at reducing or avoiding the use of liquid nitrogen (LN2) to store boar semen. Sperm-rich fractions from three boars were pooled and frozen in 0.25- and 0.5-mL straws using four different protocols: a conventional method (Control; LN2 vapours followed by immersion and storage in LN2), FT1 (as Control but stored at -80°C), FT2 (LN2 vapours followed by storage at -80°C), and FT3 (direct transfer from 5°C to -80°C plus storage at this temperature). Sperm quality and intracellular H2O2 production were evaluated at 30- and 150-min after thawi

Simulated microgravity alters sperm navigation, fertilization and embryo development in mammals.

As human space exploration extends toward long-duration missions and off-Earth settlements, understanding the impact of microgravity on reproduction is vital for sustaining life beyond Earth. This study investigates how simulated microgravity influences sperm navigation, fertilization, and subsequent early embryo development across three mammalian species; human, mouse, and pig, using a dual-axis 3D clinostat and clinically relevant in vitro culture systems. Sperm function was assessed via microchannels, revealing that microgravity impaired directional navigation and fertilization capacity in

Pentoxifylline and EDTA can modulate the capacitation of frozen/thawed donkey sperm.

BACKGROUND: Capacitation is a process in which sperm undergo a series of modifications that allow it to fertilize the oocyte. Capacitation inducers are agents or substances that promote these changes essential for in vitro fertilization. AIMS/OBJECTIVES: To evaluate the use of capacitation inducers in frozen/thawed donkey semen. METHODS: Fifteen semen samples from five Creole donkeys were frozen. Post-thawing, semen samples were supplemented with pentoxifylline (PTX) at 2.5, 5.0, and 7.5 mM; ethylenediaminetetraacetic acid (EDTA) at 0.5, 1.0, and 1.5 mM; heparin (HEP) at 1.1 mM (positive con

Cryopreservation of Aseel rooster sperm using different glycerol-iodixanol combinations and evaluation of post-thaw quality.

BACKGROUND: Cryopreservation of avian semen is a critical tool for genetic conservation and poultry breeding programs. However, the use of glycerol, the most common penetrating cryoprotectant, is associated with dose-dependent impairments in sperm function and potential contraceptive effects in the female reproductive tract. In this study, we investigated the potential of iodixanol, a non-penetrating and biocompatible medium, to improve post-thaw sperm quality when used alone or in combination with reduced glycerol concentrations. Semen samples from ten Aseel roosters were pooled and cryoprese

Evaluation of Oxidative Profiling in Semen Fractions and Its Relationship With Sperm Function in Kangal Akkaraman Rams.

The determination of sperm oxidative stress provides significant information related to semen quality. Most studies investigate oxidative stress markers only at the level of seminal plasma, but this approach may provide an incomplete overview of the oxidative status of semen. The aim of the study is to determine the relationship between protein-normalized oxidative stress markers and spermatological parameters in ram semen fractions. A total of 38 ejaculates obtained from 24 rams were fractionated into seminal plasma, whole semen (non-centrifuged), and pellet at different concentrations. Total

Myo-inositol in assisted reproductive technologies: Roles in sperm function, oocyte maturation, and embryo development across species.

Assisted reproductive technologies (ARTs) are widely used in animal and human reproduction; however, their efficiency remains limited due to oxidative stress-induced damage to gametes and embryos during in vitro manipulation. Myo-inositol (Myo-Ins), a naturally occurring cyclitol and key precursor of phosphatidylinositol signaling molecules, has emerged as a promising modulator of reproductive performance due to its roles in intracellular signaling, redox regulation, and metabolic homeostasis. This review summarizes and critically evaluates current evidence regarding the role of Myo-Ins in ART

Glycine improves boar sperm linear motility by promoting heme biosynthesis to regulate mitochondrial energy metabolism.

Boar sperm are prone to functional deterioration during incubation, which is commonly associated with reduced motility, impaired mitochondrial function, and insufficient adenosine triphosphate (ATP) production. This study investigated whether glycine supplementation improves boar sperm progressive and linear motility by enhancing mitochondrial energy metabolism through the 5-aminolevulinic acid (5-ALA)/heme-associated mitochondrial pathway. Boar sperm samples were incubated with different concentrations of glycine (0, 0.1, 1, and 10 mM) at 37 °C for 6 h, and sperm motility was assessed at 0, 1

A systematic review and meta-analysis of astaxanthin efficacy in male infertility: evidence from clinical and preclinical studies.

Astaxanthin (AST), a potent antioxidant carotenoid, has been investigated for its potential to improve male reproductive outcomes; however, findings remain inconsistent. This systematic review and meta-analysis aimed to evaluate the ability of AST to improve male infertility-related outcomes in both animal models and human clinical trials. A comprehensive search was carried out in Web of Science, Scopus, and PubMed. Eligible studies assessed the effects of AST on semen quality or oxidative stress markers. The risk of bias was assessed via the SYRCLE for animal studies and Cochrane RoB 2.0 for

Standardized Cryopreservation Protocols for European Perch (Perca fluviatilis) and Pikeperch (Sander lucioperca) Semen Enabling Hatchery-Scale Fertilization.

Cryopreservation of fish semen ensures the year-round availability of gametes from species with seasonal spawning, supports genetic resource banking, and underpins selective breeding programs. However, percid species such as European perch and pikeperch pose specific challenges, notably the spontaneous activation of sperm motility in seminal plasma, which can reduce sperm motility reserves during cryopreservation. Building on systematic studies, here we present standardized protocols for semen collection, sperm quality evaluation, extender preparation, freezing, thawing, and post-thaw handling

Advancing paternal age: Effects on sperm quality and offspring health.

BACKGROUND: Over recent decades, socioeconomic and cultural changes have led to a progressive increase in paternal age at conception, prompting growing concern regarding its potential effects on male reproductive function, pregnancy outcomes, and offspring health. In contrast to the extensive literature on female reproductive aging, the biological and clinical consequences of paternal aging have received less attention. OBJECTIVE: This narrative review summarizes and critically discusses current evidence on the impact of advanced paternal age on conventional and biofunctional sperm parameters

The zinc contraceptive effect: targets, timing, and quantitative thresholds.

There is a critical need for novel non-hormonal contraceptives. Zinc-based intrauterine devices (IUDs) demonstrate 100% efficacy in rats, yet the mechanism remains unclear. We investigated whether zinc targets sperm, embryos, or the endometrium. Rats with zinc, copper, or nylon IUDs were mated. All retrieved embryos were fertilised, indicating no primary spermicidal effect. However, embryos from rats with a zinc IUD failed to reach the blastocyst stage, unlike those from rats with a copper IUD. In vitro embryo culture confirmed that zinc significantly inhibited mouse embryo development in a do

DNA Fragmentation Analysis in Human Sperm-Technical Instructions to Prevent False Positives and Negatives in Angle-Modulated Two-Dimensional Single-Cell Pulsed-Field Gel Electrophoresis.

Over the past two decades, numerous studies have examined the etiological significance of DNA fragmentation in human sperm using methods such as the comet assay (CA), the sperm chromatin structure assay, the sperm chromatin dispersion assay, and the TUNEL assay. We developed single-cell pulsed-field gel electrophoresis techniques, including one-dimensional (1D-SCPFGE) and angle-modulated two-dimensional (2D-SCPFGE), to detect early signs of naturally occurring DNA fragmentation. Comparative studies using purified human sperm with and without DNA fragmentation revealed some technical limitation

Machine learning-driven proteomic profiling reveals seminal plasma biomarkers linking trihalomethane exposure to impaired sperm quality.

The decline in sperm quality has emerged as a major public health concern with significant implications for male fertility and population sustainability. However, high-accuracy biomarkers for sperm quality are lacking, and the environmental drivers remain obscure. Trihalomethanes (THMs), ubiquitous disinfection by-products in drinking water, have been linked to declining semen quality worldwide, yet the molecular mechanisms underlying THM-induced reproductive toxicity remain underexplored. In this propensity score-matched case-control study designed to maximize internal validity, we identified

Niosomal l-carnitine and quercetin improve sperm quality and testicular function in atrazine-induced reproductive toxicity in rats.

Atrazine (ATZ), a frequently employed herbicide, is classified as one of the environmental pollutants that play a pivotal role in progression of male infertility. L-carnitine (LC) and quercetin (QT) possess antioxidant properties, rendering them viable additional treatments for male infertility. Additionally, specialized drug delivery systems, such as niosomes, enhance the distribution of hydrophilic pharmaceuticals. This study aimed to investigate the ameliorative potential of LC and QT, administered in conventional and niosomal formulations, against ATZ-induced testicular dysfunction in adul

Epididymosome-Supplemented Extender Induces Changes in Morpho-Functional Traits and microRNA Levels of Post-Thaw Sperm and Improves Embryo Developmental Potential.

While molecular signature contributes to sperm quality and fertility potential, sperm may lost key molecules during cryopreservation. Since sperm cells are transcriptionally inert and rely on interactions with epididymosomes (epEVs) to acquire molecules, herein, we investigated the effects of the addition of epEVs to the cryopreservation extender on post-thaw sperm quality and fertility potential. Epididymal sperm were obtained from six bulls (n = 6) and a pool of epEVs was formed from the epididymal fluid of 15 bulls. After, a total of 30 × 106 sperm was cryopreserved in the presence (epEVs)
