Virulence of two Streptococcus pyogenes strains (types M1 and M3) associated with toxic-shock-like syndrome depends on an intact mry-like gene.

The major virulence factor of Streptococcus pyogenes, the M protein, is positively regulated at the transcriptional level by mry in the M type 6 strain studied. We show here that in two S. pyogenes strains isolated from cases of toxic-shock-like syndrome, a type M1 strain and a type M3 strain, an mry-like gene is also required for resistance to phagocytosis.

Molecular identification with virulence gene profiles and in-vitro antifungal susceptibility of Candida species associated with subclinical mastitis in Anatolian buffaloes and dairy cows.

Mastitis is a serious multifactorial disease of dairy animals. Etiologically, bacterial mastitogens are intensively investigated. Conversely, yeast-associated mastitis, particularly Candida species, has received comparatively less attention in buffaloes. The present study aimed to perform species-level molecular identification of Candida isolates obtained from Anatolian buffaloes and dairy cows with subclinical mastitis, to characterize the selected virulence gene profiles, and to evaluate antifungal susceptibility patterns. Milk samples were collected on an udder-quarter basis from 1188 quart…

Immune protection in chickens via lipid nanoparticle-encapsulated VP2 DNA vaccine against very virulent infectious bursal disease virus.

Infectious bursal disease (IBD), caused by infectious bursal disease virus (IBDV), is an acute and highly contagious avian infection that primarily causes severe immunosuppression in chickens. Since the 1980s, very virulent IBDV (vvIBDV) has triggered a global crisis in the poultry industry due to its high mortality. Although IBD has become largely sporadic in recent years following widespread vaccination, the persistent threat of vvIBDV underscores the critical need for novel vaccines that are both safer and more effective. In this study, a lipid nanoparticle (LNP)-encapsulated DNA vaccine en

Expression of bluetongue virus core proteins VP2 and VP7 by bovine herpesvirus-4 confers protection against virulent challenge in a mouse model.

Bluetongue virus (BTV) remains a major threat to livestock health and agricultural economies worldwide due to its multiple serotypes, vector transmission, and wide geographic distribution. Vaccination is essential for controlling BTV outbreaks; however, developing vaccines that provide broad protection while complying with the requirements of differentiating infected from vaccinated animals (DIVA) remains challenging. In this study, we evaluated the antigenicity and protective efficacy of two BTV proteins, VP2 and VP7, delivered using a bovine herpesvirus-4 (BoHV-4)-based viral vector. Groups

Vaccination with a PRRSV-1 modified live vaccine provides protection against a highly virulent PRRSV-1.1 Spanish strain challenge in piglets.

In 2020, atypical outbreaks of porcine reproductive and respiratory syndrome virus type 1 strain (PRRSV-1) were reported in northeastern Spain, characterized by high abortion rates, severe respiratory signs, and elevated mortality in affected nurseries. Sequencing analyses identified a highly virulent PRRSV-1.1 strain (Rosalia). This study evaluated the efficacy of a commercial live attenuated PRRSV-1 vaccine against this strain. Seventy PRRSV-naïve, 3-week-of-age piglets were randomly assigned to vaccinated (intradermal) and non-vaccinated groups. Five weeks post-vaccination, all piglets were

Discovering host-viral protein interactions in autophagy: A LIR discovery pipeline for identifying LC3-interacting region motifs in highly virulent viruses.

Hemorrhagic fever virus (HFV) infections are highly fatal, posing a significant global pandemic threat as they continue to emerge in new locations. HFVs and other highly virulent viruses (HVVs), such as Nipah virus, exploit host cell pathways, including the conserved host autophagy pathway, to promote viral replication. The Atg8/microtubule-associated protein 1 light chain 3 (LC3) proteins are necessary for autophagosome formation and maturation. Proteins interact with Atg8-family proteins through LC3-interacting region (LIR) motifs, which is a short linear motif (SLiM) found in intrinsically

Phenotypic and Genomic Divergence in Biofilm Formation in STEC and Non-STEC Escherichia coli: The Impact of Genomic Plasticity on the Virulence and Persistence of Bovine Isolates.

Shiga toxin-producing Escherichia coli (STEC) is a zoonotic pathogen of global relevance whose persistence in food processing environments is frequently mediated by biofilm formation. The acquisition of stx prophages and associated regulatory mutations can impose adaptive restrictions on this phenotype. This study analyzed 216 whole-genome sequences (WGS) to investigate the genomic determinants of biofilm formation in a diverse collection of E. coli isolates (STEC and non-STEC) isolated from cattle feedlots. Genomes were characterized for serogroup, MLST, stx subtyping, integrity of the mlrA a
