[Rhino-cerebral fungal infection successfully treated with supplementary hyperbaric oxygen therapy].

Rhino-cerebral fungal infections are rare and difficult disorders to cure. We report the case of a woman presenting a left trigeminal neuralgia complicated by ophthalmoplegia and blindness. MRI demonstrated a lesion of the left orbital apex with extension into the cavernous sinus. Fungal infiltration (aspergillosis or mucormycosis), was seen on biopsy. High-dose liposomal Amphotericin B (5mg/kg/day) for six weeks was unsuccessful. Adjunctant hyperbaric oxygen therapy led to clinical and radiological improvement. Hyperbaric oxygen therapy is discussed in the medical management of rhino-cerebral

Massive septic thrombus formation on a superior vena cava indwelling catheter following Torulopsis (Candida) glabrata fungemia.

Fungal endocarditis is an exceedingly rare complication of indwelling central venous catheters in adults. Here we describe what appears to be the first case of a right atrial thrombus superinfected with the yeast Torulopsis (Candida) glabrata and attached to an indwelling superior vena cava catheter that was not used for parenteral nutrition. A large vegetation-like mass adherent to the catheter tip was visualized by transesophageal echocardiography in a patient who presented with signs of septic pulmonary embolism. Following open-heart surgery, the definitive diagnosis was established by hist

Evaluation of the fungitest kit by using strains from human immunodeficiency virus-infected patients: study of azole drug susceptibility.

One hundred eighteen Candida clinical isolates from human immunodeficiency virus-infected patients were tested for their susceptibilities to fluconazole and itraconazole by Fungitest and the National Committee for Clinical Laboratory Standards MIC method. Fungitest results depended on both yeast species and antifungal agents. This test is able to detect sensitive strains (97% agreement with results of the MIC method in tests with fluconazole and 84% agreement in tests with itraconazole) but has a poor capacity to detect resistant strains (26% agreement in tests with fluconazole and 5% agreemen

Cutaneous lymphatic sporotrichosis.

The first case of cutaneous lymphatic sporotrichosis from Nagaland and a case of cutaneous sporotrichosis from Kerala who had acquired infection from Assam are reported. The diagnosis in both cases were established by isolating Sporothrix schenckii from multiple cutaneous lesions. The dimorphic nature of fungus was established in vitro by demonstrating the mycelial phase at 25-30 degrees C and yeast phase at 37 degrees C and pathogenicity to white mice. Both the patients were successfully treated with oral administration of potassium iodide for 3 months.

[Value of hypertonic media for the isolation of aberrant forms of Candida albicans from women with lower genital tract infections].

Women presenting symptoms and/or signs of lower genital tract infection were examined mycologically and to increase the sensitivity of the culture methods and to estimate the aberrant forms of fungi, hypertonic media was used. 47 yeast species were isolated from 86 patients. On the other hand 8 Candida albicans strains were isolated by using hypertonic culture media in which no growth was observed in any of the media used for routine culture. In relation with this study 8 cases were evaluated and the culture methods that have been used were discussed.

Noncandidal, fungal infections of the burn wound.

Candidal (yeast) and noncandidal (filamentous fungal) wound infections have become an increasingly important cause of burn-associated morbidity and mortality. However, these two diseases differ markedly in their epidemiology, onset, appearance, diagnosis, and treatment. In the last four years, we have had five cases of noncandidal sepsis (four mucoraceae and one aspergillus) that are illustrative of these differences. Early recognition and diagnosis are essential. Radical excision when the infection is confined to superficial invasion of only one anatomic area is key to decreasing mortality fr

Acquired immunodeficiency syndrome manifested as disseminated cryptococcosis.

A 32-year-old male homosexual presented to the emergency department (ED) with the clinical picture of a nonspecific illness. While in the ED, he experienced a first-time seizure. Computed tomography (CT) showed an enhancing mass lesion. Antibacterial therapy was started and continued until a second lumbar puncture (LP), 36 hours after admission, showed distinct yeast forms. Subsequent institution of appropriate therapy did not prevent the patient's death. The cause of death was disseminated cryptococcosis secondary to acquired immunodeficiency syndrome (AIDS).

Effect of chloramphenicol on the electron systems in Ustilago cynodontis.

The mycelial cells of Ustilago cynodontis possess at least two electron transport systems: a cyanide-sensitive cytochrome pathway, which represents the major route for electron transport, and an alternative cyanide-insensitive pathway, inhibited by salicylhydroxamic acid. In the presence of chloramphenicol in the culture medium, mycelial cells respire only by the alternatuve chain. The stable induced yeast-like cells, obtained by prolonged chloramphenicol treatment of the mycelial cells, respire as the untreated mycelial cells; this result indicates that the phenotypic change induced by chlora

A CEREBELLAR DISORDER IN CHICKS, APPARENTLY OF NUTRITIONAL ORIGIN.

Growing chicks maintained on a diet consisting of milk powder, casein, starch, yeast, cod liver oil, salts and filter paper develop ataxia, tremors, retraction or twisting of the head, clonic spasms of the legs, and stupor. These symptoms may appear suddenly, usually between the 18th and 25th day, and may end in death. If recovery takes place, the chicks may go on to normal development. Definite lesions are found in the cerebellum of the affected chicks. These consist of edema, necrosis and hemorrhages. Hyaline thrombi are found in the capillaries in and about the degenerated areas.

STUDIES ON BACTERIAL NUTRITION : II. GROWTH ACCESSORY SUBSTANCES IN THE CULTIVATION OF HEMOPHILIC BACILLI.

The hemophilic bacteria of which Bacillus influenzae serves as a type require for their growth two distinct and separable substances, both of which are present in blood and neither of which alone suffices. These substances are (a) a vitamine-like substance which can be extracted from red blood corpuscles, from yeast, and from vegetable cells, which is relatively heat-labile and absorbed from solution by certain agents; (b) a so called X substance which is present in red blood cells, is heat-stable, and acts in minute amounts.

First report of Candidozyma auris (Candida auris) in a cat: A case of persistent shedding.

Candidozyma (Candida) auris is a multidrug-resistant fungal pathogen of growing concern. We report the first isolation of C. auris in a domestic cat in Italy. The yeast was repeatedly recovered from feces over six months, suggesting persistent intestinal colonization. Isolation occurred during and after immunosuppressive treatment for eosinophilic granuloma complex. Antifungal susceptibility testing revealed resistance to azoles, amphotericin B and capsofungin. This case highlights the potential role of companion animals as reservoirs of C. auris, supporting a One Health perspective in fungal

Protocol for adaptive laboratory evolution of S. cerevisiae by PEG/LiAc transformation and sequencing.

Gene deletions perturb cellular homeostasis, affecting gene expression and phenotypes. Here, we present a protocol for serial transfer experiments in Saccharomyces cerevisiae strains following targeted gene knockouts. We describe steps for gene deletion, serial passage optimization, and growth kinetics analysis. We then detail procedures for genomic and transcriptomic profiling. This protocol enables the investigation of changes in yeast strains resulting from gene loss. For complete details on the use and execution of this protocol, please refer to Jiang et al.1.

Candida auris Diagnostics: Identification and Screening.

Candida auris is an emerging yeast species classified as an urgent threat by the Centers for Disease Control and Prevention due to its ability to colonize numerous surfaces, its ability to spread through health care systems, its elevated antifungal resistance, and its high mortality rate. Accurate detection of C auris from patient specimens is crucial for containing, preventing the spread of, and managing patients with C auris. Here, we review currently available diagnostics, current screening guidance, and identify areas where diagnostics could improve.

Cation Transporters of Candida albicans-New Targets to Fight Candidiasis?

Candidiasis is the wide-spread fungal infection caused by numerous strains of yeast, with the prevalence of Candida albicans. The current treatment of candidiasis is becoming rather ineffective and costly owing to the emergence of resistant strains; hence, the exploration of new possible drug targets is necessary. The most promising route is the development of novel antibiotics targeting this pathogen. In this review, we summarize such candidates found in C. albicans and those involved in the transport of (metal) cations, as the latter are essential for numerous processes within the cell; henc

Saccharomyces cerevisiae-Based Probiotics as Novel Antimicrobial Agents to Prevent and Treat Vaginal Infections.

Vaginal infections affect 70% of women during their lifetimes and account for millions of annual doctors' visits. These infections are predominantly represented by vulvovaginal candidiasis (VVC) and bacterial vaginosis (BV). Although standard antimicrobial agents remain the major strategy for the prevention and treatment of vaginal infections, both VVC and BV are difficult to treat due to high rates of resistance and recurrence, high probability of complications, and negative effects on the vaginal microbiota. This review focuses on a new approach of yeast-based probiotics for the prevention a

Granulomatous mastitis due to coinfection with Histoplasma sp. and Paracoccidioides sp.: A case report.

A 51 years old female patient with granulomatous mastitis diagnosis, was referred to the Medical Mycology Department "Dr. Dante Borelli" at the Instituto de Medicina Tropical, Universidad Central de Venezuela (IMT-UCV). Mycological diagnosis demonstrated the presence of intracellular yeast structures suggesting Histoplasma sp. and also multi-budding structures compatible with Paracoccidioides sp. To the best of our knowledge, this is the first case report of a granulomatous mastitits due to coinfection of both fungi. 2012 Elsevier Ltd. All rights reserved.

A recurrent GARS mutation causes distal hereditary motor neuropathy.

We found a p.Gly327Arg mutation in GARS in two unrelated women, both of whom had a similar phenotype - motor weakness that began in late childhood, distal weakness in the arms and legs, a motor greater than sensory neuropathy with slowing of motor and not sensory conduction velocities. A de novo mutation was proven in one patient and suspected in the other. The p.Gly327Arg GARS variant did not support yeast growth in a complementation assay, showing that this variant severely impairs protein function. Thus, the p.Gly327Arg GARS mutation causes a distal motor neuropathy.

Effect of Antiobiotic-Induced Disbiosis and Its Correction with Probiotics on Myocardial Tolerance to Ischemia-Reperfusion Injury in SPF Rats.

Changes in morphofunctional parameters of the isolated heart subjected to global ischemia-reperfusion were studied in SPF Wistar rats with antibiotic-induced dysbiosis (AID) treated with lyophilized yeast Saccharomyces boulardii and inactivated probiotic bacteria Lactobacillus reuteri KR2017. In contrast to S. boulardii, correction of dysbiosis with L. reuteri KR2017 against the background of gastric hypersecretion and standard antimicrobial therapy led to an increase in fat content and a decrease in free and bound water in tissues and to a significant reduction in myocardial infarct size caus

A Rare Case of White Piedra Caused by Candida parapsilosis in the Sub-Himalayan Region of North India.

White piedra is a superficial fungal infection of the hair shaft, caused by Trichosporon, a noncandidal yeast characterized by the presence of numerous, discrete, asymptomatic nodules attached to the infected hair shafts. White piedra is considered a disease of tropical regions and occasionally reported from temperate countries. Although Candida parapsilosis such as Candida albicans is well known to cause cutaneous infections, it has been reported as a co-isolate for white piedra along with Trichosporon. We report a case of white piedra from a temperate region caused by C. parapsilosis.

[Biological activity of Penicillium sp. 10-51 exometabolites].

Silica gel column chromatography (silica gel "L" II kind of activity 100/160 mkm) of the chloroform extract from the cultural filtrate of Penicillium sp. 10-51 gave two fractions (chloroform and chloroform-acetone, 5:1) having biological activity. Recrystallization yielded two compounds. On the basis of physico-chemical and spectral data these compounds were identified as curvularin and hydroxycurvularin, which have a large spectrum of biological action as to bacteria, yeast, blue-green algae and phytopathogenic micromycetes.
