Acute kidney injury (AKI) is a leading cause of distant organ dysfunction among critically ill patients. Mitochondrial dysfunction is considered a key factor driving the damage after renal ischemia-reperfusion (IR) injury. Damaged mitochondria release mitochondrial damage-associated molecular patterns (mtDAMPs) into the cytosol, which initiate a systemic inflammatory response. To better understand the underlying mechanism, mice were challenged with 30 min of bilateral renal ischemia followed by 24 h of reperfusion. The cytokine profiling in mouse lung tissues revealed that TREM-1 was significa

One of the main goals of vaccine research is the development of adjuvants that can enhance immune responses and are both safe and biocompatible. We explored the application of the natural polymer hyaluronan (HA) as a promising immunological adjuvant for protein-based vaccines. Chemical conjugation of HA to antigens strongly increased their immunogenicity, reduced booster requirements, and allowed antigen dose sparing. HA-based bioconjugates stimulated robust and long-lasting humoral responses without the addition of other immunostimulatory compounds and proved highly efficient when compared to

The neuroinflammation in the ischemic brain could occur as sterile inflammation in response to damage-associated molecular patterns (DAMPs). However, its long-term dynamic transcriptional changes remain poorly understood. It is also unknown whether this neuroinflammation contributes to the recovery or just deteriorates the outcome. The purpose of this study is to characterize the temporal transcriptional changes in the post-stroke brain focusing on DAMPs-related genes by RNA-sequencing during the period of 28 days. We conducted the RNA-sequencing on day 1, 3, 7, 14, 28 post-stroke in the mouse

Nonalcoholic fatty liver disease (NAFLD) is a multifactorial pathological condition, which recognizes a certain sexual dimorphism. Experimental and clinical studies provided evidence for a critical role of macrophages in NAFLD development and progression. Especially, liver-resident macrophages (also known as Kupffer cells) are likely the common final pathway of several pro-steatosic signals. A huge amount of danger-associated molecular patterns recognized by Kupffer cells is provided within the liver by lipid and glucose toxicity. Other pro-inflammatory signals come from surrounding tissues in

Air pollution is a major environmental risk factor contributing to the development, exacerbation, and persistence of asthma. A wide range of airborne pollutants, including particulate matter (PM), ozone (O₃), nitrogen dioxide, sulfur dioxide, diesel exhaust particles (DEPs), volatile organic compounds, and biological contaminants, can interact with the airway epithelium and immune system, leading to dysregulated inflammatory responses. Increasing evidence indicates that pollutant exposure induces epithelial injury, disrupts epithelial barrier integrity, promotes oxidative stress, and activates

PURPOSE: Radiation-induced intestinal injury (RIII) is a serious complication of radiation therapy without effective therapies. The alarmin, S100A8/A9, a biomarker of gastrointestinal inflammation, is highly elevated following abdominal irradiation (ABI). However, it remains unclear whether it functions as an active driver of RIII pathogenesis. The study aims to assess the effects of S100A8/A9 on the pathogenesis of RIII. Our goal is to identify its dynamic alteration during RIII and determine whether it can be used as a therapeutic target in the injury and repair phases of acute RIII.

BACKGROUND: Osteoarthritis (OA) is among the common chronic health conditions which impacts many aspects of an individual's life from physical function to mental health. Inflammation, in the joint as well as system inflammation is among the many mechanisms hypothesized to be involved in OA. IL-33 which belongs to the IL-1 family, is considered as an alarmin in OA. IL-33 binds to a group of receptors to stimulate signaling among which is the ST2L receptor that induced the activation of NFƙb, thus, working as a booster of the inflammatory cascade. Many polymorphisms have been identified on the I

BACKGROUND AND AIMS: The triggering factors of sepsis-induced myocardial dysfunction (SIMD) are poorly understood and are not addressed by current treatments. S100A8/A9 is a pro-inflammatory alarmin abundantly secreted by activated neutrophils during infection and inflammation. We investigated the efficacy of S100A8/A9 blockade as a potential new treatment in SIMD.
METHODS: The relationship between plasma S100A8/A9 and cardiac dysfunction was assessed in a cohort of 62 patients with severe sepsis admitted to the intensive care unit of Linköping University Hospital, Sweden. We used S100A8/A9 b

Traumatic brain injury (TBI) is a devastating condition due to its long-term sequelae on neurological functions. Inflammatory responses after TBI are critical for injury expansion and repair. Recent research in central nervous system (CNS) disorders reveals the importance of IL-33 and its receptor (ST2) as an alarmin system to initiate immune responses. This study explored the role of IL-33/ST2 signaling in TBI. TBI was induced in adult male C57BL/6J mice using a controlled cortical impact (CCI) model. We found that the expression of IL-33 increased in the injured brain and blood, and ST2 was

Ehlers-Danlos syndromes (EDS) are clinically and genetically heterogeneous disorders characterized by soft connective tissue alteration like joint hypermobility and skin hyper-extensibility. We previously identified heterozygous missense mutations in the C1R and C1S genes, coding for the complement C1 proteases, in patients affected by periodontal EDS, a specific EDS subtype hallmarked by early severe periodontitis leading to premature loss of teeth and connective tissue alterations. Up to now, there is no clear molecular link relating the nominal role of the C1r and C1s proteases, which is to

INTRODUCTION: Chronic Periodontitis (CP) is an inflammatory disease of bacterial origin that results in alveolar bone destruction. Porphyromonas gingivalis (Pg), one of the main periopathogens, initiates an inflammatory cascade by host immune cells thereby increasing recruitment and activity of osteoclasts, the bone resorbing cells, through enhanced production of the crucial osteoclastogenic factor, RANK-L. Antibodies directed against some cytokines (IL-1β, IL-6 and TNF-α) failed to exhibit convincing therapeutic effect in CP. It has been suggested that IL-33, could be of interest in CP.

Chronic osteoarthritis (OA) is a degenerative disease of the articular cartilage. DNA-binding high mobility group protein B1 (HMGB1) is released on cellular death/activation and acts as an endogenous danger signal and a proinflammatory cytokine. Matrix metalloproteinase (MMP)-2 and in MMP-9 are induced to mediate proteolytic degradation/remodelling of joint tissues. Collagen degradation in the bone and synovium leads to release of type I collagen-derived cross-linked carboxy-terminal telopeptide (ICTP). These molecules have been linked to the pathogenesis of OA and could have potential as syno

BACKGROUNDS: Pediatric patients with congenital heart disease (CHD) often require surgical repair using cardiopulmonary bypass. Despite advancements, mortality and complication rates remain significant.
METHODS & RESULTS: We prospectively examined 101 patients undergoing congenital cardiac surgery, identifying a mortality rate of 4.0% and a complication rate of 31.6%. Neonates and infants exhibited multiple complications more frequently. Prolonged bypass time was significantly associated with complications, with each additional 30 min increasing the odds by 1.46 times (95% CI 1.01-2.10, p = 0

Damage-associated molecular patterns (DAMPs) and pathogen-associated molecular patterns (PAMPs) are key triggers of inflammation in sepsis. However, they have rarely been studied simultaneously. Thus, in the present study of patients with bacteraemic infection, we aimed to study how DAMP dynamics are linked to disease severity and outcome and to compare diagnostic and prognostic properties of a DAMP and a previously analysed PAMP (16S rDNA). In a prospective study of adult patients hospitalized with culture-proven community-onset bacteraemic infection, caused by Streptococcus pneumonia (n = 30

Toll-like receptors (TLRs) are discovered as crucial pattern recognition receptors (PRRs) involved in the recognition of pathogen-associated molecular patterns (PAMPs). Later studies showed their involvement in the recognition of various damage/danger-associated molecular patterns (DAMPs) generated by host itself. Thus, TLRs are capable of recognizing wide-array of patterns/molecules derived from pathogens and host as well and initiating a proinflammatory immune response through the activation of NF-κB and other transcription factors causing synthesis of proinflammatory molecules. The process

Viral proteins mimic host protein structure and function to redirect cellular processes and subvert innate defenses. Small basic proteins compact and regulate both viral and cellular DNA genomes. Nucleosomes are the repeating units of cellular chromatin and play an important part in innate immune responses. Viral-encoded core basic proteins compact viral genomes, but their impact on host chromatin structure and function remains unexplored. Adenoviruses encode a highly basic protein called protein VII that resembles cellular histones. Although protein VII binds viral DNA and is incorporated wit

Resolution of inflammation is an active process that requires efferocytosis, the engulfment of apoptotic cells by macrophages, mediated by receptors such as MerTK. IL-33 is an alarmin that initiates type 2 immune responses, including increased production of IL-13, which promotes MerTK expression. The ability of IL-33 to promote efferocytosis in vivo was examined. Intraperitoneal administration of IL-33 to mice increased local MerTK+ macrophage numbers within 48h. MerTK+ macrophages were not similarly induced by free mitochondria, an alternative cell damage associated signal. Efferocytotic acti

BACKGROUND: Interleukin-33 (IL-33), a tissue-derived alarmin released during sterile and infectious stress, modulates inflammatory responses in an organ-specific manner. Although IL-33 is implicated in hepatic pathophysiology, its functional mechanism in sepsis-induced liver injury (SILI) remains insufficiently characterized.
METHOD: SILI was modeled in vivo via cecal ligation and puncture (CLP) and in vitro using lipopolysaccharide (LPS)-stimulated primary hepatocytes. The multimodal evaluations included oxidative stress analysis, inflammatory cytokine measurements, liver functional biomarke

BACKGROUND: Severe asthma (SA) is characterized by corticosteroid insensitivity, persistent airway inflammation, and excessive oxidative stress, underscoring an urgent unmet therapeutic need. Meroterpenoids derived from Rhododendron capitatum Maxim have demonstrated potent anti-inflammatory properties; however, their therapeutic potential in SA has not been elucidated.
METHODS: A meroterpenoid-rich fraction was isolated from R. capitatum Maxim, and cannabichromeorcin (CBCO) was identified as its most abundant constituent. The therapeutic effects of CBCO were evaluated in murine models of SA i

Spinal cord injury (SCI) triggers a complex and evolving cascade of pathological events in which neuroinflammation and microenvironmental imbalance critically constrain repair. Among the cellular mediators, microglia exhibit remarkable functional plasticity, transitioning across diverse states that can either support tissue repair or exacerbate secondary damage. However, the mechanisms governing this dynamic reprogramming remain incompletely understood, limiting the development of effective immunomodulatory strategies. Recent evidence identifies the interleukin-33 (IL-33)/ST2 axis as an emergi
