Amphetamine blocks long-term synaptic depression in the ventral tegmental area.

The mesolimbic dopamine system is essential for reward-seeking behavior, and drugs of abuse are thought to usurp the normal functioning of this pathway. A growing body of evidence suggests that glutamatergic synapses on dopamine neurons in the ventral tegmental area (VTA) are modified during exposure to addictive drugs, producing sensitization, a progressive augmentation in the rewarding properties of psychostimulant drugs with repeated exposure. We have tested the hypothesis that psychostimulant exposure interferes with the synaptic plasticity of glutamatergic inputs to the VTA. We find that

Identification and validation of an explainable model of methamphetamine use disorders with anxiety symptoms: a multicenter cross-sectional study.

AIMS: Negative reinforcement process triggered by anxiety is an important factor in promoting the relapse behavior of substance use disorder. Objective identification of the anxiety level of methamphetamine (METH) use disorder (MUD) and other addiction behaviors becomes particularly important, yet the current understanding of this issue remains unclear. METHODS: This multicenter cross-sectional study in China was conducted on METH patients between September 1, 2019 and September 1, 2021 for the identification and validation of the prediction model. MUD with anxiety symptoms was defined mainly

Focus on Cognitive Enhancement: A Narrative Overview of Nootropics and "Smart Drug" Use and Misuse.

Whilst "nootropics" are meant to treat a range of medical disorder-related cognitive impairments, the typically healthy "smart drugs" user ingests a range of drugs/molecules to achieve improved mental performance. Given the increasing levels of related concerns, this study aimed to provide an overview of the clinical pharmacological issues relating to both the most popular nootropics and the vast range of drugs that are being used as putative cognitive enhancers/smart drugs. In terms of the cognitive decline associated with neurological degenerative disorders, a significant variation in resear

CST, an Herbal Formula, Exerts Anti-Obesity Effects through Brain-Gut-Adipose Tissue Axis Modulation in High-Fat Diet Fed Mice.

The brain, gut, and adipose tissue interact to control metabolic pathways, and impairment in the brain-gut-adipose axis can lead to metabolic disorders, including obesity. Chowiseungcheng-tang (CST), a herbal formulation, is frequently used to treat metabolic disorders. Here, we investigated the anti-obesity effect of CST and its link with brain-gut-adipose axis using C57BL/6J mice as a model. The animals were provided with a normal research diet (NRD) or high-fat diet (HFD) in absence or presence of CST or orlistat (ORL) for 12 weeks. CST had a significant anti-obesity effect on a number of v

Experiences in managing problematic crystal methamphetamine use and associated depression in gay men and HIV positive men: in-depth interviews with general practitioners in Sydney, Australia.

BACKGROUND: This paper describes the experiences of Australian general practitioners (GPs) in managing problematic crystal methamphetamine (crystal meth) use among two groups of male patients: gay men and HIV positive men. METHODS: Semi-structured qualitative interviews with GPs with HIV medication prescribing rights were conducted in Sydney, Adelaide and a rural-coastal town in New South Wales between August and October 2006. Participants were recruited from practices with high caseloads of gay and HIV positive men. RESULTS: Sixteen GPs were recruited from seven practices to take part in in

Development of an addiction index and delineation 15-year trends of illicit drugs from the Taiwan national drug enhancement database.

Illicit drug use contributes to substantial morbidity and mortality. Drug scheduling, a legal measure in drug enforcement, is often structured as a hierarchy based on addiction tendency, abuse trends, and harm, but may lack data-driven evidence when classifying substances. Our study aims to measure addiction tendency and use trends based on real-world data. We used the open access database of National Police Agency, Ministry of the Interior in Taiwan and analyzed all daily criminal cases of illicit drugs from 2013 to 2017 and monthly illicit drug enforcement data from the same database from 20

Short-term cessation of sex work and injection drug use: evidence from a recurrent event survival analysis.

OBJECTIVE: This study quantitatively examined the prevalence and correlates of short-term sex work cessation among female sex workers who inject drugs (FSW-IDUs) and determined whether injection drug use was independently associated with cessation. METHODS: We used data from FSW-IDUs (n=467) enrolled into an intervention designed to increase condom use and decrease sharing of injection equipment but was not designed to promote sex work cessation. We applied a survival analysis that accounted for quit-re-entry patterns of sex work over 1-year stratified by city, Tijuana and Ciudad Juarez, Mexi

Sex differences in disinhibition and its relationship to physical abuse in a sample of stimulant-dependent patients.

BACKGROUND: Research suggests that impulsivity is a vulnerability factor for developing stimulant dependence, that women develop dependence more quickly than men, and that physical abuse can increase impulsivity and may have greater adverse health consequences in women. This study sought to tie these findings together by evaluating: (1) sex differences in disinhibition prior to lifetime initiation of stimulant abuse and (2) the relationship between physical abuse and disinhibition in stimulant-dependent patients. METHOD: The Frontal Systems Behavior Scale (FrSBe) is a reliable and valid self-

Non-suicidal Self-Injury in the Context of Psoriasis, Post-traumatic Stress Disorder, and Acute Polysubstance Intoxication.

Non-suicidal self-injury (NSSI) during acute intoxication has been reported in association with stimulant and polysubstance use. Chronic somatic pain and trauma exposure may further increase vulnerability to impulsive self-injury. However, the intersection of inflammatory skin disease, addiction, and trauma-related pathology remains underdescribed in the clinical literature. We describe an unusual case of a hospitalized patient with psoriasis, post-traumatic stress disorder, and polysubstance use disorder who intentionally ignited psoriatic plaques during acute intoxication with cocaine, amphe

Approach With Caution: A Pilot Randomised Controlled Trial of Approach Bias Modification for People Undergoing Residential Treatment for Methamphetamine Use Disorder.

INTRODUCTION: Less than one-quarter of people who attend residential rehabilitation for methamphetamine use disorder (MUD) remain abstinent for 1 year. Approach bias modification (ApBM), a cognitive training intervention, reduces relapse rates when delivered during residential rehabilitation for alcohol use disorder. However, there has been little research on ApBM for MUD. METHODS: Twenty-four patients with MUD attending four residential treatment services in Melbourne were randomised, between March 2020 and August 2024, to 6 sessions of either ApBM (n = 10) or sham-control training (n = 14).

Multi-metric resting-state fMRI reveals brain network abnormalities underlying cognitive impairment in male patients during methamphetamine abstinence.

BACKGROUND: Methamphetamine (MA) use disorder leads to persistent cognitive impairment, but the underlying neural mechanisms during abstinence remain unclear. This study aimed to investigate brain functional abnormalities in male methamphetamine-abstinent patients and their associations with cognitive impairment using multiple resting-state fMRI metrics. METHODS: Twenty-five male MA-abstinent patients (Disease Group, DG) and twenty-nine healthy controls (Healthy Control Group, HC) were recruited, and rs-fMRI data were collected. Multimodal analysis methods, including regional homogeneity (ReH

Unwanted effects, self-management strategies, and consequences of unwanted effects of methamphetamine use.

UNLABELLED: Background Acute methamphetamine toxicities are poorly characterized. We sought to describe unwanted physical and mental effects from methamphetamine use, consequences of unwanted effects (e.g., altercations), and self-management strategies. METHODS: We used data from two cross-sectional surveys conducted from 2023 to 2024 in San Francisco and Oakland, CA (N = 1385), of people over 18 years of age who used unregulated drugs. Among participants reporting past 90-day methamphetamine use (n = 955), we determined the proportion with past 90-day unwanted effects; consequences of the mo

Methamphetamine intoxication and suicidal ideation/behavior in the emergency department.

BACKGROUND: This study investigates the association between Methamphetamine (MA) intoxication and suicidal ideation/behavior in patients presenting to emergency departments. Amidst rising MA use and co-use with opioids, this "twin epidemic" has manifested in increasing admissions for MA intoxication, often accompanied by psychiatric symptoms that can escalate to suicidal behaviors. METHOD: This retrospective study utilized patient records and analyzed data from 629 patients admitted to a Texas emergency department in 2020, with MA intoxication confirmed via urine tests and patient interviews.
