Anthelmintic-induced destrobilation and its influence on calculated drug efficacy in Hymenolepis diminuta infections in rats.

Anthelmintic efficacy studies typically involve direct counts of worms remaining in the host shortly after drug treatment. Few such studies, however, have considered the phenomenon of tapeworm destrobilation when determining effective dosages. The present study reports on the frequency of drug-induced destrobilation and the subsequent regeneration of Hymenolepis diminuta in rats following treatment with niclosamide or praziquantel and its implications with respect to the apparent efficacy of these anthelmintics. Drug efficacies very similar to those reported in the literature were determined u…

The impact of repeated doses of anthelmintic on the intensity of infection and age structure of Fasciola hepatica populations in sheep.

A mathematical model of ovine fascioliasis is used to assess the likely impact of repeated doses of 2 anthelmintic types on the intensity of infection in the ewes. When chemotherapy is applied in the autumn and early winter, the type of anthelmintic that kills proportionately more of the immature parasites brings about a greater reduction in the predicted overall mean intensity of infection than the type of anthelmintic that kills proportionately more of the mature parasites, and leads to a lower predicted burden of mature flukes. In both cases, each dose of flukicide has proportionately less

Clinical evaluation of a new anthelmintic - C. 9333 Go./CGP 4540 in human hookworm infection.

A new antihookworm compound, C.9333 Go./CGP 4540, was evaluated clinically in 65 adult patients with hookworm infection. Several dosage schedules were evaluated to determine an effective, well tolerated and convenient regime. The lowest effective dose was 125 mg administered at four-hourly intervals for three doses. The compound was generally well tolerated; mild giddiness was observed in nine patients and severe giddiness was present in one patient. Clinical and biochemical monitoring revealed no other toxicity of the compound.

Preliminary efficacy of anthelmintic closantel as a potential antimicrobial against Gram-positive bacteria: An assessment of the enantioselectivity.

OBJECTIVES: Approved salicylamide agents exhibit promising activity against multidrug-resistant Gram-positive bacteria (GPB). The objectives of this study were to investigate whether the antimicrobial efficacy of closantel (CLO) is related to its enantiomeric properties, and whether the CLO enantiomers contribute the efficacy significantly. METHODS: The in vitro antibacterial activities of R-CLO and S-CLO against staphylococcus and enterococcus were explored using minimum inhibitory concentrations, growth inhibition curves, time-killing assays, and postantibiotic effects. The potential effect

Anthelmintic medication with focus on first-trimester exposure: an evaluation of pregnancy outcomes based on the Embryotox cohort.

The present study aims to contribute to the available experience on the systemic use of five different anthelmintics during pregnancy (albendazole, mebendazole, praziquantel, pyrantel and pyrvinium). Particular focus was placed on the occurrence of major birth defects and pregnancy loss following first-trimester exposure. As the assessed agents differ considerably in terms of their chemical structure and mechanism of action, they were analyzed separately. A total of 282 pregnancies with exposure to the study medication were recorded in the Embryotox database (January 1, 2000-February 28, 2023)…

Fasciolosis, a foodborne zoonotic trematode infection in cattle in Bangladesh: multifaceted validation of parthenogenecity and anthelmintic efficacy.

Parthenogenic Fasciola (Trematoda: Fasciolidae) flukes have been developed by the hybridization of Fasciola hepatica and Fasciola gigantica. They are aspermic (asF) but capable of clonal expansion through parthenogenesis and are spreading rapidly throughout the globe. Here, we unambiguously prove the occurrence of parthenogenic Fasciola in cattle in Bangladesh, along with their ex vivo culture protocol and anthelmintic efficacy. By employing multiple conventional and molecular tools, we confirmed the presence of both the spermic F. gigantica (sFg) (31.7%; 814/2575) and asF (68.3%; 1761/2575) i

In vivo and in vitro anthelmintic activity of quebracho-chestnut tannin extract against Haemonchus contortus in lambs.

Haemonchus contortus is considered the most pathogenic gastrointestinal nematode of sheep and, due to the increasing resistance to synthetic anthelmintics, it poses a serious challenge to small ruminant production systems. The aim of this work was to evaluate the in vivo and in vitro anthelmintic activity of quebracho-chestnut tannin extract against Haemonchus contortus in lambs. The in vitro assays (egg hatching inhibition and larval migration inhibition) were performed to determine the effective concentrations required to inhibit 50% of egg hatching and larval migration (EC50), which were 3.…

Integrated epidemiology and multi-assay evidence of anthelmintic resistance in ovine gastrointestinal nematodes of Northern India.

Gastrointestinal parasitic (GIP) infections are a major constraint to sustainable small ruminant production worldwide due to increasing anthelmintic resistance caused by extensive reliance on chemical anthelmintics. In this study, a total of 2308 faecal samples were collected and examined from naturally infected sheep across northern India, encompassing 15 districts of Punjab and 07 of Himachal Pradesh. An overall prevalence of 96.4% for GIP infections was recorded with strongyles being predominant followed by coccidia. Quantitative faecal examination revealed 62.6% light, 10.2% moderate and 2…

Three-year evaluation of anthelmintic treatment efficacy against equine strongyles in France using FECRT.

Reduced anthelmintic efficacy against strongyles in equids is a widespread concern, but up-to-date field data are scarce. The aim of this study was to evaluate, over a three-year period, the efficacy of the anthelmintic (AH) molecules available in France (fenbendazole, pyrantel, ivermectin and moxidectin) against strongyles in equids using faecal egg count reduction tests (FECRTs). A total of 104 complete FECRTs (i.e., required sample size reached) were conducted in 98 groups, involving 749 equids kept in 70 facilities. Fenbendazole-resistant populations were detected in 8/9 groups (89%) from

The anthelmintic activity of the white wormwood (Artemisia herba Alba) against Haemonchus contortus in beef cattle.

The objective of the study was to determine the efficacy of white wormwood on helminthes in beef cattle production. Water extracts of white wormwood of different levels of phytotoxicity were used to treat female adult H. contortus over 8 h under controlled laboratory conditions. The experiment was designed in a completely randomized design with six treatments replicated three times. Treatments 3-6 showed a reduction in worm motility over time (P < 0.05) while it did not change much for T1 and T2 (P < 0.05). Even after 8 h of incubation, more than 50% of the worms were still active in T1, T2 an
