Spaceflight regulates ryanodine receptor subtype 1 in portal vein myocytes in the opposite way of hypertension.

Gravity has a structural role for living systems. Tissue development, architecture, and organization are modified when the gravity vector is changed. In particular, microgravity induces a redistribution of blood volume and thus pressure in the astronaut body, abolishing an upright blood pressure gradient, inducing orthostatic hypotension. The present study was designed to investigate whether isolated vascular smooth muscle cells are directly sensitive to altered gravitational forces and, second, whether sustained blood pressure changes act on the same molecular target. Exposure to microgravity

Facing ambiguous threats.

On February 1, 2003, the world watched in horror as the Columbia space shuttle broke apart while reentering the earth's atmosphere, killing all seven astronauts. Some have argued that NASA's failure to respond with appropriate intensity to the so-called foam strike that led to the accident was evidence of irresponsible or incompetent management. The authors' research, however, suggests that NASA was exhibiting a natural, albeit unfortunate, pattern of behavior common in many organizations. The foam strike is a prime example of what the authors call an ambiguous threat-a signal that may or may …

Extraterrestrial regolith is hemostatic and potentially suitable for hemorrhage control in space.

BACKGROUND: Long-duration space missions beyond low Earth orbit pose increased risks of injury to astronauts. Traumatic hemorrhage will be a cause of preventable death. Due to payload constraints, in situ production of medical materials is essential. Regolith from the Moon, Mars, and asteroids is rich in silicates, which may serve as a hemostatic agent. OBJECTIVES: Here, we aimed to evaluate whether extraterrestrial regolith simulants, their mineral components, and meteorites can activate coagulation through factor (F)XII and control bleeding. METHODS: The procoagulant potential of Lunar and…

Baicalein Attenuates Disuse-Driven Skeletal Senescence in Association with Gut Microbiota Modulation.

The progression of skeletal senescence under mechanical unloading conditions elevates fracture risk, emerging as a critical health challenge for the general population and astronauts during spaceflight. Nevertheless, current therapeutic strategies demonstrate persistent limitations in efficacy and safety, with unresolved challenges in long-term sustainability. Baicalein, a natural flavonoid compound, exhibits favorable anti-aging and anti-inflammatory effects, while its therapeutic potential and underlying mechanism in bone disorders remain unknown. In this study, a hind limb unloading (HLU) r…

Hypergravity reduces F-actin accumulation in osteoclasts, with attenuated bone resorption.

Bone loss occurs in astronauts during prolonged spaceflight, thus indicating the sensitivity of skeletal homeostasis to altered gravitational environments. Previous studies have shown that microgravity affects osteoclast differentiation and bone resorption, which suggests that osteoclasts possess mechanisms to sense and respond to gravity-generated mechanical forces. For testing of the related mechanisms, hypergravity can be experimentally reproduced with use of a centrifuge. In the present study, osteoclasts derived from mouse bone marrow were subjected to hypergravity under three conditions:…

Dose and LET-dependence of gastric cancer risk in Apc1638N/+ mice exposed to space-relevant radiation types.

Exposure to high-linear energy transfer (LET) heavy ions (Z > 2) present in galactic cosmic radiation (GCR) is predicted to pose a significant but uncertain risk of gastric (stomach) cancer among astronauts. Uncertainties in current risk estimate models are primarily attributed to the lack of in vivo radiobiological data on dose- and LET-responses of high-LET heavy-ions. To address this gap, we investigated dose- and LET-dependent gastric tumorigenesis in male Apc1638N/+ mice following exposure to γ rays and 28Si ions (69 keV/μm) at doses of 5, 10, and 50 cGy. Additionally, a separate cohort o…

Spaceflight-Associated Epiphora Mechanisms and Countermeasures.

INTRODUCTION: Epiphora, or excessive tearing, is a commonly reported symptom among those with dry eyes in the terrestrial environment. It is therefore not surprising that a higher frequency of epiphora was demonstrated among astronauts onboard both Space Transportation Shuttle and International Space Station missions. In this review paper, we discuss the pathophysiology and mechanisms of epiphora and consider the additional risk factors due to the extreme environments of spaceflight, including microgravity and, to a lesser extent, radiation. We also discuss potential strategies to mitigate spa…

Comparing Hypothermic and Thermal Neutral Conditions to Induce Metabolic Suppression.

Suppressing metabolism in astronauts could decrease CO2 production. It is unknown whether active cooling is required to suppress metabolism in sedated patients. We hypothesized that hypothermia would have an additive effect with dexmedetomidine on suppressing metabolism. This is a randomized crossover trial of healthy subjects receiving sedation with dexmedetomidine and exposure to a cold (20°C) or thermal neutral (31°C) environment for 3 hours. We measured heart rate, blood pressure, core temperature, resting oxygen consumption (VO2), resting carbon dioxide production (VCO2), and resting ener…

Heavy-ion radiation-induced colitis and colorectal carcinogenesis in Il10-/- mice display co-activation of β-catenin and NF-κB signaling.

Space radiation-induced gastrointestinal (GI) cancer risk models for future interplanetary astronauts are being developed that primarily rely on quantitative animal model studies to assess radiation-quality effects of heavy-ion space radiation exposure in relation to γ-rays. While current GI-cancer risk estimation efforts are focused on sporadic GI-cancer mouse models, emerging in-vivo data on heavy-ion radiation-induced long-term GI-inflammation are indicative of a higher but undetermined risk of GI-inflammation associated cancers, such as colitis-associated cancer (CAC). Therefore, we aimed …

Altered cerebral perfusion in response to chronic mild hypercapnia and head-down tilt Bed rest as an analog for Spaceflight.

PURPOSE: Following prolonged stays on the International Space Station (ISS), some astronauts exhibit visual acuity changes, ophthalmological findings, and mildly elevated intracranial pressures as part of a novel process called spaceflight-associated neuro-ocular syndrome (SANS). To determine the pathophysiology of SANS, NASA conducted a multi-investigator study in which 11 healthy participants underwent head-down tilt bed rest, mimicking microgravity-induced cephalad fluid shifts, combined with elevated ambient CO2 levels similar to those on the ISS (HDT+CO2). As part of that study, we examin…

Cerebral hypoperfusion precedes nausea during centrifugation.

INTRODUCTION: Nausea and motion sickness are important operational concerns for aviators and astronauts. Understanding the underlying mechanisms associated with motion sickness may lead to new treatments. The goal of this work was to determine if changes in cerebral blood flow precede the development of nausea in subjects susceptible to motion sickness. METHODS: Cerebral flow velocity in the middle cerebral artery (transcranial Doppler), BP, and end-tidal CO2 were measured while subjects were rotated on a centrifuge (250 degrees x s(-1)). Following 5 min of rotation, subjects were translated …

Acute hemodynamic responses to weightlessness during parabolic flight.

Pilots and astronauts experience fluid shifts in variable gravity. Acute effects of fluid shifts on the cardiovascular system were monitored on NASA's KC-135 aircraft during parabolic flight. The variability of R-R intervals in the electrocardiogram was measured as an indication of vagal cardiac neural activity. R-R intervals were measured during the gravity transition from 2-G to 0-G produced by parabolic flight to assess the involvement of the autonomic nervous system in regulating the acute effects of fluid shifts. In seven subjects, a BoMed noninvasive continuous cardiac output monitor (NC…

Interplanetary travel and permanent injury to normal heart.

This hypothesis is that some crewmen on prolonged space flights may develop permanent myocardial injury despite the absence of coronary atherosclerosis and even without the hazards of radiation beyond orbit. This may resuIt from atrophy of skeletal muscle and bone resulting in magnesium ion deficiency predisposing to a vicious cycle with catecholamine elevations, with the latter aggravated by stress, dehydration-provoked angiotensin elevations, unremitting endurance exercise, and in turn a second vicious cycle with severe ischemia. Toxic free radicals can develop complicating ischemia and pote

UHAB-1: Physiological responses to a 48-hour spaceflight analog mission underwater.

Underwater habitats (UHABs) are a promising research platform to investigate human performance in spaceflight because they combine isolation, confinement, limited physical activity, and technical life-support demands with real environmental hazards. UHABs accurately replicate several challenges of long-duration spaceflight, including real over perceived isolation, dependency on habitat and technology for survival, and a platform for evaluating novel countermeasures. Here we report the first physiological case study from a 48-hour, single-occupancy underwater habitat (UHAB-1), designed as a pro
