Huoxue Jiedu formula attenuates myocardial ischemia-reperfusion injury by modulating LAPTM4B/mTORC1/TFEB pathway-mediated autophagic flux.

BACKGROUND: Myocardial ischemia-reperfusion injury (MIRI) severely limits the benefits of revascularization in acute myocardial infarction, with impaired autophagic flux being a central pathological mechanism. The Huoxue Jiedu Formula (HXJDF), a traditional Chinese medicine prescription, has demonstrated cardioprotective potential, yet its underlying mechanisms remain unclear. PURPOSE: This study aimed to determine whether HXJDF ameliorates MIRI by restoring impaired autophagic flux and to elucidate the underlying mechanisms. METHODS: MIRI-related genes were identified from GEO transcriptomi

Daily heat stress treatment rescues denervation-activated mitochondrial clearance and atrophy in skeletal muscle.

KEY POINTS: Traumatic nerve injury or nerve disease leads to denervation and severe muscle atrophy. Recent evidence shows that mitochondrial loss could be a key mediator of skeletal muscle atrophy. Here, we show that daily heat stress treatment rescues denervation-induced loss of mitochondria and concomitant muscle atrophy. We also found that denervation-activated autophagy-dependent mitochondrial clearance (mitophagy) was suppressed by daily heat stress treatment. The molecular basis of this observation is explained by our results showing that heat stress treatment attenuates the increase of

Degradation of Mutant Protein Aggregates within the Endoplasmic Reticulum of Vasopressin Neurons.

Misfolded or unfolded proteins in the ER are said to be degraded only after translocation or isolation from the ER. Here, we describe a mechanism by which mutant proteins are degraded within the ER. Aggregates of mutant arginine vasopressin (AVP) precursor were confined to ER-associated compartments (ERACs) connected to the ER in AVP neurons of a mouse model of familial neurohypophysial diabetes insipidus. The ERACs were enclosed by membranes, an ER chaperone and marker protein of phagophores and autophagosomes were expressed around the aggregates, and lysosomes fused with the ERACs. Moreover,…

Autophagy in lysosomal myopathies.

Lysosomal myopathies are hereditary myopathies characterized morphologically by the presence of autophagic vacuoles. In mammals, autophagy plays an important role for the turnover of cellular components, particularly in response to starvation or glucagons. In normal muscle, autolysosomes or autophagosomes are typically inconspicuous. In distinct neuromuscular disorders, however, lysosomes become structurally abnormal and functionally impaired, leading to the accumulation of autophagic vacuoles in myofibers. In some instances, the accumulation of autophagic vacuoles can be a prominent feature, …

Protein aggregation and autophagy dysfunction: new lessons from mucopolysaccharidoses.

Mucopolysaccharidoses (MPS) are inherited metabolic diseases with strong neurological involvement. MPSs are caused by defects in lysosomal enzymes involved in the degradation of glycosaminoglycans (GAGs), which consequently accumulate into the lysosomes as primary storage. Macroautophagy/autophagy impairment is well known to drive neurodegeneration in MPSs, however, mechanisms underlying such dysfunction are still poorly understood. Recently, by studying a mouse model for MPS-III (Sanfilippo syndrome) we have shown that the progressive aggregation of amyloid proteins in neuronal cell bodies oc

Antigen B from Echinococcus granulosus regulates autophagy-mediated macrophage polarization to alleviate immune thrombocytopenia.

BACKGROUND: Immune thrombocytopenia (ITP) is an acquired autoimmune disease characterized by a low platelet count (< 100 × 109/L) induced by … AgB primarily acts by influencing the expression of LC3II/LC3I and p62, increasing the formation of autophagosomes and enabling lysosomes to identify and consume autophagosomes more accurately. AgB also inhibits macrophage polarization towards M1. These results suggested that AgB reduced hemorrhage in the ITP mouse model by regulating autophagy-mediated macropha …

African swine fever virus B66L drives extracellular mitochondrial release to promote systemic inflammation in mice.

Systemic inflammation is a hallmark of viral infection, but the upstream signals that initiate it remain poorly defined. Here we show that extracellular mitochondria act as inflammatory mediators during infection by vesicular stomatitis virus, influenza A virus, rabies virus, herpes simplex virus 1 and African swine fever virus (ASFV). In ASFV-infected primary porcine alveolar macrophages, the viral protein B66L promotes the capture of damaged mitochondria by autophagosomes while blocking their fusion with lysosomes, causing mitochondria to accumulate outside cells. Extracellular mitochondria …

The FTLD Risk Factor TMEM106B Regulates the Transport of Lysosomes at the Axon Initial Segment of Motoneurons.

Genetic variations in TMEM106B, coding for a lysosomal membrane protein, affect frontotemporal lobar degeneration (FTLD) in GRN- (coding for progranulin) and C9orf72-expansion carriers and might play a role in aging. To determine the physiological function of TMEM106B, we generated TMEM106B-deficient mice. These mice develop proximal axonal swellings caused by drastically enlarged LAMP1-positive vacuoles, increased retrograde axonal transport of lysosomes, and accumulation of lipofuscin and autophagosomes. Giant vacuoles specifically accumulate at the distal end and within the axon initial seg…

Autophagy dysregulation in cell culture and animals models of spinal muscular atrophy.

Abnormal autophagy has become a central thread linking neurodegenerative diseases, particularly of the motor neuron. One such disease is spi … induction of autophagy accompanied by attenuated autophagic flux, resulting in the accumulation of autophagosomes and their associated cargo. Expression of the SMN-binding protein a-COP, a known modulator of autophagic flux, can ameliorate this autophagic traffic jam.

A Salmonella protein causes macrophage cell death by inducing autophagy.

Salmonella enterica, the causative agent of food poisoning and typhoid fever, induces programmed cell death in macrophages, a process found to be dependent on a type III protein secretion system, and SipB, a protein with membrane fusion activity that is delivered into host cells by this system. When expressed in cultured cells, SipB caused the formation of and localized to unusual multimembrane structures. These structures resembled autophagosomes and contained both mitochondrial and endoplasmic reticulum markers. A mutant form of SipB devoid of membrane fusion activity localized to mitochondr…

Autophagy-mediated reduction of miR-345 contributes to hepatic cystogenesis in polycystic liver disease.

BACKGROUND & AIMS: Polycystic liver disease (PLD) is characterised by increased autophagy and reduced miRNA levels in cholangiocytes. Given that autophagy has been implicated in miRNA regulation, we tested the hypothesis that increased autophagy accounts for miRNA reduction in PLD cholangiocytes (PLDCs) and accelerated hepatic cystogenesis. METHODS: We assessed miRNA levels in cultured normal human cholangiocytes (NHCs), PLDCs, and isolated PLDC autophagosomes by miRNA-sequencing (miRNA-seq), and miRNA targets by mRNA-seq. Levels of miR-345 and miR-345-targeted proteins in livers of animals a…
