Mechanisms for axon maintenance and plasticity in motoneurons: alterations in motoneuron disease.

In motoneuron disease and other neurodegenerative disorders, the loss of synapses and axon branches occurs early but is compensated by sprouting of neighboring axon terminals. Defective local axonal signaling for maintenance and dynamics of the axonal microtubule and actin cytoskeleton plays a central role in this context. The molecular mechanisms that lead to defective cytoskeleton architecture in two mouse models of motoneuron disease are summarized and discussed in this manuscript. In the progressive motor neuropathy (pmn) mouse model of motoneuron disease that is caused by a mutation in th

A few axonal proteins distinguish ventral spinal cord neurons from dorsal root ganglion neurons.

A series of proteins putatively involved in the generation of axonal diversity was identified. Neurons from ventral spinal cord and dorsal root ganglia were grown in a compartmented cell-culture system which offers separate access to cell somas and axons. The proteins synthesized in the neuronal cell somas and subsequently transported into the axons were selectively analyzed by 2-dimensional gel electrophoresis. The patterns of axonal proteins were substantially less complex than those derived from the proteins of neuronal cell bodies. The structural and functional similarity of axons from dif

Initial stages of retinofugal axon development in the hamster: evidence for two distinct modes of growth.

In order to characterize differences in growth patterns of axons as they elongate toward their targets and during the initial stages of terminal arbor formation within the targets, we examined the primary visual system of fetal and newborn hamsters using three morphological methods: the Cajal-deCastro reduced silver method, the rapid Golgi technique, and anterograde transport of HRP. Axons emerge from the retina between the 10th and 11th embryonic days (E10-E11). The front of retinal axons crosses the chiasm, extends over the primitive dorsal nucleus of the lateral geniculate body (LGBd) by E1

Vesicular glutamate transporter 2-immunoreactive synapses onto phrenic motoneurons in the neonatal rat.

The synaptic organization between vesicular glutamate transporter 2 (VGLUT2)-immunoreactive (ir) axon terminals and phrenic motoneurons in the neonatal rat was examined using a combined retrograde tracing and immunohistochemistry for VGLUT2. The phrenic nucleus (PhN) contained large numbers of VGLUT2-ir axon terminals, some of which made axosomatic and axodendritic synapses with PhN motoneurons. These terminals were of asymmetrical type and contained spherical clear synaptic vesicles. The results suggest that in the neonatal rat glutamatergic synapses onto PhN motoneurons exist and mediate exc

Electron microscopic identification of trigeminal nerve fibers in the ciliary body and the iris.

The trigeminal nerve fibers in the ciliary body and the iris have been identified electron microscopically by using intra-axonally transported horseradish peroxidase injected into the trigeminal ganglion. The tracer revealed unmyelinated fiber profiles in the stroma of the ciliary body and the iris. These were not found to be associated with any particular organ but rather may terminate as free endings. Large profiles of varicosities contained mitochondria. Thin profiles, containing neurotubules, probably represent axons between the varicosities or endings. An interaction between the sensory a

Continuous low amplitude direct current stimulation of the crushed peripheral nerve accelerates the early recovery of choline acetyltransferase but not of acetylcholinesterase activity in fast and slow muscles.

We investigated if continuous 1 µA direct current stimulation of the injured nerve, with the cathode electrode at the distal end of the nerv … mber of motor axons entering the muscles. It seems possible that cathode stimulation increased ChAT axonal transport, causing the early increase of ChAT content in the nerve endings. This raises the possibility that the axon transport and subsequent secretion of a trophic factor(s) from the nerve to the reinnervated muscle are enhanced as well, thus shortening the …

Targeting and anchoring the mechanosensitive ion channel Piezo to facilitate its inhibition of axon regeneration.

Mechanical force orchestrates a myriad of cellular events including inhibition of axon regeneration, by locally activating the mechanosensitive ion channel Piezo enriched at the injured axon tip. However, the cellular mechanics underlying Piezo localization and function remains poorly characterized. We show that the RNA repair/splicing enzyme Rtca acts upstream of Piezo to modulate its expression and transport/targeting to the periphery of the soma via the Rab10 GTPase, whose expression also relies on Rtca. In vitro evidence that covalent crosslinking of neurofilaments occurs in gamma-diketone neuropathy.

We have postulated that the toxic neuropathies associated with neurofilament-filled axonal swellings have a common pathogenesis, the covalent crosslinking of neurofilaments during anterograde transport. The newly described gamma-diketone, 3,4-dimethyl-2,5-hexanedione (DMHD), is a more potent analogue of the toxic metabolite of n-hexane, 2,5-hexanedione. The axonal swellings observed in DMHD toxicity are in the proximal axon, as seen in intoxication with beta, beta'-iminodipropionitrile, rather than in the distal axon, where neurofilamentous swellings are observed in n-hexane, carbon disulfide,

Axonal Protection by Netarsudil, a ROCK Inhibitor, Is Linked to an AMPK-Autophagy Pathway in TNF-Induced Optic Nerve Degeneration.

PURPOSE: Netarsudil, a Rho kinase inhibitor with norepinephrine transport inhibitory effect, lowers intraocular pressure, however, its effect on axon damage remains to be elucidated. The aim of the current study was to investigate the effect of netarsudil on TNF-induced axon loss and to examine whether it affects phosphorylated-AMP-activated kinase (p-AMPK) and autophagy in the optic nerve. METHODS: Intravitreal administration of TNF or TNF with netarsudil was carried out on rats and quantification of axon number was determined. Electron microscopy determined autophagosome numbers. In vivo confocal neuroimaging (ICON): non-invasive, functional imaging of the mammalian CNS with cellular resolution.

With in vivo confocal neuroimaging (ICON), single retinal ganglion cells (RGCs) can be visualized non-invasively, repeatedly, in real-time and under natural conditions. Here we report the use of ICON to visualize dynamic changes in RGC morphology, connectivity and functional activation using calcium markers, and to visualize nanoparticle transport across the blood-retina barrier by fluorescent dyes. To document the versatility of ICON, we studied the cellular response to optic nerve injury, and found evidence of reversible soma swelling, recovery of retrograde axonal transport and a difference…

Aging-Related Changes in the Injury Response of the Peripheral Nervous System.

Peripheral nerve injury (PNI) significantly impairs patients' quality of life, with elderly individuals experiencing particularly severe consequences due to aging-related declines in neuronal injury response and repair capabilities. Processes of the generation and transmission of injury signals, axonal disruption, initiation of regeneration, and the elongation of regenerating axons, as well as the subsequent reinnervation by these axons, are all significantly influenced by aging. These alterations are closely associated with changes in mitochondrial function, neuronal transport systems, a pers

Receptor cell regeneration and connectivity in olfaction and taste.

The capacity of adult mammalian gustatory and olfactory receptor cells to regenerate and make synaptic reconnections provides examples that may be useful in initiating replacement of other kinds of sensory receptor cells. The sensory code for taste quality may not be degraded by taste receptor cell turnover because axons probably recouple to the appropriate type of new receptor cell by axon-receptor cell affinity. Experiments on the development and regeneration of taste receptor cells suggest that they regenerate and turn over by recapitulating the late but not the early steps in taste bud dev

Taxol-induced neuropathy: chronic effects of local injection.

The long-term neurotoxic effects of taxol, a compound known to promote microtubule protein polymerization, injected subepineurially into rat sciatic nerve were studied up to 10 weeks post-injection. At the site of injection, taxol caused local axonal reactions and degeneration which were causally related to the slow progressive accumulation of microtubules and other axoplasmic constituents. This culminated in the appearance of giant axonal spheroids and profiles similar to the retraction bulbs of Wallerian degeneration. From these axonal bulbs, many of which arose at nodes of Ranvier, groups o

Cholinergic function in lumbar aluminum myelopathy.

To determine whether perikaryal neurofilamentous accumulation in cholinergic neurons is associated with a deficit in cholinergic function, we developed a new model of aluminum-induced neurofibrillary degeneration, referred to as focal lumbar aluminum myelopathy. The model is produced by direct intramedullary microinjection of AlCl3, which results in a characteristic neurological syndrome. Four weeks after injections, affected rabbits show extensive neurofilamentous lesions of both large and small neurons in the lumbar spinal cord, including a majority of anterior horn cells. These animals are

A different type of vertebrate thalamic organization.

The presence of a reptilian thalamic reticular nucleus was investigated in Caiman crocodilus. Injections of horseradish peroxidase made into the dorsal thalamus produced retrogradely labeled neurons in the nucleus of the dorsal peduncle of the lateral forebrain bundle. A separate population of thalamic neurons located in the nucleus of the dorsal peduncle of the lateral forebrain bundle with morphology different from those neurons projecting to the dorsal thalamus were found to be immunoreactive for glutamic acid decarboxylase. Taken together, these findings suggest that Caiman, like mammals,

Movement of choline acetyltransferase in axons disconnected from their cell bodies.

1. Phrenic nerves of rats were cut out and divided into two parts of equal length, one proximal and one distal. The activity of choline acetyltransferase of the two pieces was estimated. Four groups of rats were studied: (1) normal rats, and rats in which 3 days earlier, (2) the right nerve had been cut in the uppermost part of the thorax, (3) a ligature had been tied round the right nerve just above the diaphragm, and (4) these two procedures had been combined.2. In normal rats the activity of the enzyme was about the same in proximal and distal pieces. Ligation of the nerve caused an increas

SARM1 deletion delays cerebellar but not spinal cord degeneration in an enhanced mouse model of SPG7 deficiency.

Hereditary spastic paraplegia is a neurological condition characterized by predominant axonal degeneration in long spinal tracts, leading to weakness and spasticity in the lower limbs. The nicotinamide adenine dinucleotide (NAD+)-consuming enzyme SARM1 has emerged as a key executioner of axonal degeneration upon nerve transection and in some neuropathies. An increase in the nicotinamide mononucleotide/NAD+ ratio activates SARM1, causing catastrophic NAD+ depletion and axonal degeneration. However, the role of SARM1 in the pathogenesis of hereditary spastic paraplegia has not been investigated.

Ventricular Netrin-1 deficiency leads to defective pyramidal decussation and mirror movement in mice.

The corticospinal tract (CST) is the principal neural pathway responsible for conducting voluntary movement in the vertebrate nervous system. Netrin-1 is a well-known guidance molecule for midline crossing of commissural axons during embryonic development. Families with inherited Netrin-1 mutations display congenital mirror movements (CMM), which are associated with malformations of pyramidal decussation in most cases. Here, we investigated the role of Netrin-1 in CST formation by generating conditional knockout (CKO) mice using a Gfap-driven Cre line. A large proportion of CST axons spread la
