STUDIES ON BACILLUS WELCHII WITH SPECIAL REFERENCE TO GAS GANGRENE.

1. Spores of the Bacillus welchii group of bacteria were found on 100 per cent of the uniforms of Belgian soldiers who had come directly from the trenches, and in the meshes of all the samples examined of the new cloth from which the uniforms were made. 2. In fifteen out of twenty fresh war wounds members of this group of bacteria were found. Of the fifteen patients, only three later developed gas gangrene. Once the spores of Bacillus welchii have been carried into a wound the deep-lying lacerated muscle tissue appears to be the most important factor in the onset of gas gangrene. 3. Bacillus w…

Natural Endotoxemia in Dogs-A Hidden Condition That Can Be Treated with a Potential Probiotic Containing Bacillus subtilis, Bacillus licheniformis and Pediococcus acidilactici: A Study Model.

Spore-based Bacillus spp. products are considered to have a higher probiotic potential compared to products containing only lactic acid bacteria because their viability in the gastrointestinal (GI) tract is higher, even when GI environmental conditions are unfavorable. The aim of this study was to assess the effect of a Bacillus subtilis, Bacillus licheniformis and Pediococcus acidilactici spore-based potential probiotic on the natural levels of postprandial endotoxemia. A total of 11 dogs completed the study: group 1-healthy dogs: n = 5; group 2-dogs with apparent dysbiosis: n = 6. For 30 day…

Pathologic and genomic characterization of an outbreak of anthrax-like disease caused by Bacillus tropicus (formerly atypical Bacillus cereus) in red kangaroos (Macropus rufus).

Atypical Bacillus cereus strains (currently classified as Bacillus tropicus) capable of causing anthrax-like disease in humans and animals have emerged in the last 2 decades. These emerging strains are characterized by the acquisition of virulence plasmids pBCXO1 and pBC210, which are homologous to the pXO1 and pXO2 virulence plasmids of Bacillus anthracis, the agent of anthrax. The aim of this study was to describe the gross, histologic, microbiologic, and molecular features of an outbreak of anthrax-like septicemia caused by B. tropicus in red kangaroos (Macropus rufus). Three red kangaroos …

Immunization against anthrax with aromatic compound-dependent (Aro-) mutants of Bacillus anthracis and with recombinant strains of Bacillus subtilis that produce anthrax protective antigen.

The safety and efficacy of five prototype, live anthrax vaccines were studied in Hartley guinea pigs and CBA/J and A/J mice. Two of the strains, Bacillus anthracis FD111 and FD112, are Aro- mutants derived by Tn916 mutagenesis of B. anthracis UM23-1. Bacillus subtilis PA1 and PA2 contain a recombinant plasmid, pPA101 or pPA102, respectively, that carries the gene from B. anthracis encoding synthesis of protective antigen (PA). The final strain, B. subtilis PA7, was isolated in this study from B. subtilis DB104 transformed with pPA101. All five strains were less virulent in guinea pigs and A/J …

CREST: phase III study of sasanlimab and Bacillus Calmette-Guérin for patients with Bacillus Calmette-Guérin-naïve high-risk non-muscle-invasive bladder cancer.

Bacillus Calmette-Guérin (BCG) is the standard of care for patients with high-risk non-muscle-invasive bladder cancer (NMIBC) after transurethral resection of bladder tumor (TURBT). BCG in combination with programmed cell death-1 (PD-1) inhibitors may yield greater anti-tumor activity compared with either agent alone. CREST is a phase III study evaluating the efficacy and safety of the subcutaneous PD-1 inhibitor sasanlimab in combination with BCG for patients with BCG-naive high-risk NMIBC. Eligible participants are randomized to receive sasanlimab plus BCG (induction ± maintenance) or BCG al…

A Nitroreductase-Activatable Lapachol Against Bacillus subtilis Unveils Antimicrobial Specificity.

Lapachol is a natural product with antimicrobial activity. Though promising for combating antimicrobial resistance, lapachol has toxicity to mammalian cells, which will cause severe side effects in vivo. To mitigate this, we developed alapachol prodrug which is dependent on the action of nitroreductase-an enzyme expressed in bacteria but not healthy mammalian cells. We observed release of lapachol in vitro by purified nitroreductase and in the gram-positive bacterial strain, Bacillus subtilis. Colony formation assays in Bacillus subtilis indicated release of lapachol, achieving comparable leve…

Nisin against Bacillus cereus spores: activity, characterization, and beef application.

Heat-resistant spore-forming bacteria are a primary cause of spoilage in ready-to-eat (RTE) foods, and effective control of spore contamination is critical for food preservation. Nisin, a GRAS antimicrobial peptide, exhibits potent activity against spore-forming bacteria, but its stage-specific effects during spore germination and outgrowth remain unclear. Heat-resistant Bacillus species were identified as the dominant spoilage bacteria in spoiled cooked beef, and B. cereus was selected to evaluate inhibitory effects of nisin via phenotypic and transcriptomic assays. The minimum inhibitory con…

Lipopeptides efficiently disperse spore crystal aggregates in Bacillus thuringiensis israelensis.

The separation of Bacillus thuringiensis (Bt) spores and crystals is often compromised by dense aggregates, particularly in Bt subsp. israelensis. This study evaluates (sonication), chemical (SDS), and biological (lipopeptide) treatments to improve aggregate dissociation in BUPM98, using AR23 as a reference and Lip (Bt subsp. kurstaki) as a naturally dissociated control. Initial hexane-based purification highlighted the impact of aggregation, with significantly lower recovery rates for israelensis compared to kurstaki. Electron microscopic analysis revealed tightly integrated spore crystal com…

Oscillatory expansion of Bacillus subtilis biofilms on MSgg agar medium.

We investigated the oscillatory expansion of Bacillus subtilis biofilms on minimal salts glycerol glutamate (MSgg) agar by modulating the concentration of the nutrient glutamate and the rigidity of the surface via agar concentration. The concentration of glutamate and agar influenced the parameters of biofilm oscillation. Specifically, elevated glutamate levels boosted the initial growth rate (within the first 15 h) and prolonged the oscillation period. Conversely, increasing the agar concentration physically restrained colony expansion and dampened the amplitude of growth rate variations, lea…

Purification of active staphylococcal alpha-toxin produced in Bacillus subtilis.

The Gram-positive bacterium Staphylococcus aureus, a multidrug-resistant pathogen, poses a significant threat to human and animal health. Its virulence is largely attributed to the production of alpha-toxin (Hla), a potent cytotoxic protein that forms pores in cell membranes, leading to lysis. This is the first study to report the successful expression and purification of wild-type Hla (HlaWT) using Bacillus subtilis-a safe, genetically stable, and endotoxin-free host system. A vector was constructed harboring the hla gene fused to Strep-tag II at the C-terminus under the control of a strong i…

Bacillus Pre-Pasteurization Screening Protocol Reduces Donor Human Milk Loss.

Introduction: Human milk is the optimal source of nutrition and bioactives for infants, particularly preterm infants, who are at higher risk of morbidity and mortality. Many premature infants require pasteurized donor human milk (PDHM) to supplement their diet. Given the increased susceptibility of preterm infants to infections, PDHM is strictly regulated to ensure safety. Pasteurization of human milk eliminates most pathogens. However, spore-forming bacteria, notably Bacillus cereus and other Bacillus species, are capable of surviving pasteurization. These bacteria contribute significantly to…

Bacillus Subtilis-Derived Exopolysaccharide Halts Depigmentation and Autoimmunity in Vitiligo.

Vitiligo has a complex multifactorial etiology involving a T-cell-mediated autoimmune response to cutaneous melanocytes. Microbial dysbiosis has been assigned a contributing role in vitiligo etiology. Treating vitiligo can be a challenging task, and finding novel treatment approaches is crucial. In this study, we tested exopolysaccharides (EPSs) isolated from Bacillus subtilis as a microbiome-based therapy. Vitiligo-prone h3TA2 mice were treated by weekly intraperitoneal EPS injection for 18 weeks. Depigmentation was evaluated over time, measuring immune responses at end point. EPS treatment s…
