In vivo comparison of avirulent Vwa- and Pgm- or Pstr phenotypes of yersiniae.

The abilities of Yersinia pestis to undergo restriction in Ca2+-deficient medium with concomitant production of V and W antigens (Vwa+) and to absorb exogenous pigments (Pgm+) are established virulence factors. Mutation of Y. pestis to Pgm- is known to promote resistance to pesticin (Pstr) and reduced lethality by peripheral routes of injection. Vwa+ Pgm- isolates of Y. pestis were shown in this study to retain virulence in mice when injected intravenously. Although Pgm- in appearance, wild-type cells of Yersinia pseudotuberculosis and Yersinia enterocolitica may also be sensitive to pesticin.

Characterization of Escherichia coli serogroups causing meningitis, sepsis and enteritis. II. Classification of Escherichia coli O78 strains by phage sensitivity, colicin type and antibiotic resistance.

Escherichia coli O78: K80 strains isolated from an outbreak of meningitis, sepsis and enteritis in infants, were compared with O78: K80 strains from sporadic cases of enteritis, healthy carriers and animals. The strains were uniform in antigenic structure and phage pattern but differed in colicinogenicity. The epidemic strains and calf-pathogenic cultures produced colicin V, the remaining isolates were characterized by other types of colicin or were not colicinogenic. Col V+ strains multiplied in the mouse peritoneal cavity more readily and killed the animals at significantly lower doses than

Unnoticed spread of linear VanA-plasmids in vancomycin-variable Enterococcus faecium strains across different regions: a diagnostics challenge.

BACKGROUND: Vancomycin-variable enterococci (VVE) are van-positive Enterococcus faecium strains with a vancomycin-susceptible phenotype that can revert to resistance upon exposure to vancomycin, posing an emerging diagnostic challenge. METHODS: We screened VVE within a large collection of E. faecium from hospitalised patients (n = 319) and healthy human volunteers (n = 60) during 2009-2022. VVE prevalence, antimicrobial resistance, plasmid replicases, virulence factors and bacteriocins were investigated using culture-based methods and WGS. vanA-carrying clusters and plasmids were analysed and

Microbial community structure, functional potential, probiotic signatures, and MAG reconstruction of fermented bamboo shoots from Northeast India.

Fermented bamboo shoot (FBS) products are widely consumed traditional foods across the Northeast region (NER) of India, yet their microbiome structure, functional capacity, biosynthetic potential, and safety attributes remain insufficiently explored. Here, comparative shotgun metagenomics of ten traditional FBS products from six NER states was used to address these gaps integrating previously generated metagenomic data from Tripura with newly generated datasets from Manipur, Meghalaya, Arunachal Pradesh, Nagaland, and Sikkim thereby bringing the total number of samples to 24. Taxonomic profili

Antibacterial and antibiofilm activities of pyocins produced by Pseudomonas aeruginosa against gram-negative bacteria: a systematic review (2015-2025).

BACKGROUND: Antimicrobial resistance among Gram-negative bacteria, particularly Pseudomonas aeruginosa, Escherichia coli, Klebsiella pneumoniae, and Acinetobacter baumannii, has reduced the effectiveness of conventional antibiotics. Biofilm formation further increases bacterial tolerance and exacerbates resistance to multiple antibiotics. Pyocins, bacteriocins produced by P. aeruginosa, are emerging as promising targeted antibacterial and antibiofilm agents. However, there is no comprehensive systematic literature synthesis of pyocin extraction, characterization, antibacterial, and antibiofilm

Mechanisms of probiotic action in poultry gut: Colonization resistance, bioactive metabolites, quorum sensing activity, and immunomodulation.

Probiotics have profound effects on the host's physiological functions and gut mucosal immunity. The in ovo and early post-hatch probiotic supplementation provides a better opportunity for commensal bacteria to reside in the avian gut and minimize the invasion of pathogenic microbes. Probiotics colonize the gut epithelial cells through adhesion structures and mucus surface-binding proteins. The successfully colonized probiotic bacteria use direct and indirect mechanisms to resist the invasion of other bacteria to develop colonization resistance. Probiotics produce the secondary metabolites, su

Secondary metabolite biosynthetic gene clusters and microbial diversity as predicted by shotgun metagenomic sequencing approach in hospitals and pharmaceutical industry wastes.

Hospital and pharmaceutical industry wastes harbor a diverse microbial community influenced by their complex chemical composition, including antibiotic-rich compounds originating from soil microorganisms. This study employs a shotgun metagenomic approach to comprehensively characterize the microbial composition of hospital and Pharmaceutical industry wastes for the first time in Ethiopia. Metagenomic DNA was extracted and used to construct a whole-genome shotgun library, which was subsequently sequenced using the Illumina HiSeq1500 platform. Taxonomic analysis revealed that bacteria were the p

Preliminary pediatric clinical evaluation of the oral probiotic Streptococcus salivarius K12 in preventing recurrent pharyngitis and/or tonsillitis caused by Streptococcus pyogenes and recurrent acute otitis media.

BACKGROUND: The oral probiotic Streptococcus salivarius K12 has been shown clearly to antagonize the growth of Streptococcus pyogenes, the most important bacterial cause of pharyngeal infections in humans, by releasing two bacteriocins named salivaricin A2 and salivaricin B. Unpublished observations indicate that it can also antagonize the growth of other bacteria involved in acute otitis media. Because of its ability to colonize the oral cavity and its safety profile, we have tested its efficacy in reducing the incidence of streptococcal pharyngitis and/or tonsillitis and episodes of acute ot

Comprehensive appraisal of the extracellular proteins from a monoderm bacterium: theoretical and empirical exoproteomes of Listeria monocytogenes EGD-e by secretomics.

Defined as proteins actively transported via secretion systems, secreted proteins can have radically different subcellular destinations in monoderm (Gram-positive) bacteria. From degradative enzymes in saprophytes to virulence factors in pathogens, secreted proteins are the main tools used by bacteria to interact with their surroundings. The etiological agent of listeriosis, Listeria monocytogenes, is a Gram-positive facultative intracellular foodborne pathogen, whose ecological niche is the soil and as such should be primarily considered as a ubiquitous saprophyte. Recent advances on protein

Cre-inducible human CD59 mediates rapid cell ablation after intermedilysin administration.

Cell ablation is a powerful tool for studying cell lineage and/or function; however, current cell-ablation models have limitations. Intermedilysin (ILY), a cytolytic pore-forming toxin that is secreted by Streptococcus intermedius, lyses human cells exclusively by binding to the human complement regulator CD59 (hCD59), but does not react with CD59 from nonprimates. Here, we took advantage of this feature of ILY and developed a model of conditional and targeted cell ablation by generating floxed STOP-CD59 knockin mice (ihCD59), in which expression of human CD59 only occurs after Cre-mediated re

The contribution of transcriptomic and proteomic analysis in elucidating stress adaptation responses of Listeria monocytogenes.

The foodborne transmission of Listeria monocytogenes requires physiological adaptation to various conditions, including the cold, osmotic, heat, acid, alkaline, and oxidative stresses, associated with food hygiene, processing, and preservation measures. We review the current knowledge on the molecular stress adaptation responses in L. monocytogenes cells as revealed through transcriptome, proteome, genetic, and physiological analysis. The adaptation of L. monocytogenes to stress exposure is achieved through global expression changes in a large number of cellular components. In addition, the cr

Emergence of optrA-mediated linezolid-resistance in human clinical Enterococcus faecalis: an alert for a shifting resistance landscape in hospitals.

OBJECTIVE: The emergence of transferable linezolid resistance genes as optrA represents an increasing concern. This study characterizes the first optrA-positive E. faecalis (Efs) isolates associated with human infections in Portugal, while highlighting undetected colonization as a reservoir for dissemination within healthcare settings. METHODS: Between July 2024-February 2025, three Efs isolates, classified as linezolid-resistant, were identified from different patients at a central hospital in Porto, Portugal. Two isolates were available for phenotypic/molecular testing and whole-genome sequ

Nonliving biotherapeutics for veterinary enteric disease: Postbiotics, bacteriophage, and antivirulence strategies to reduce antibiotic dependence.

Enteric diseases in poultry and neonatal calves remain major drivers of productivity loss and therapeutic antimicrobial use, thereby contributing to antimicrobial resistance. Necrotic enteritis in poultry is estimated to cost the global industry US $5-6 billion annually, while neonatal calf diarrhea is reported as a leading cause of morbidity (∼20-40%) and mortality (up to 8%). Nonliving biotherapeutics have attracted increasing attention because they can reduce antibiotic dependence without requiring live microbial engraftment and can be formulated for stability and consistent delivery.
