Aldehyde dehydrogenase 2 (ALDH2) is best known for its critical detoxifying role in liver alcohol metabolism. However, ALDH2 dysfunction is also involved in a wide range of human pathophysiological situations and is associated with complications such as cardiovascular diseases, diabetes mellitus, neurodegenerative diseases and aging. A growing body of research has shown that ALDH2 provides important protection against oxidative stress and the subsequent loading of toxic aldehydes such as 4-hydroxy-2-nonenal and adducts that occur in human diseases, including ischemia reperfusion injury (IRI).

Post-MI depression is a critical clinical problem, the comorbidity of which complicates depression treatment and worsens cardiovascular outcomes. However, which antidepressant is the best to lower the risk of cardiovascular events in persons with depression was still unknown. Recently, it has been proposed that the activation of ALDH2 by Alda-1 can effectively reduce depressive-like behaviors and improve the prognosis of coronary heart disease. In the present study, we investigated the effect of Alda-1 on the expression of VEGF in the hippocampus of a rat model with post-MI depression, as well

Cystic fibrosis patients exhibit chronic Pseudomonas aeruginosa respiratory infections and sustained proinflammatory state favoring lung tissue damage and remodeling, ultimately leading to respiratory failure. Loss of cystic fibrosis transmembrane conductance regulator (CFTR) function is associated with MAPK hyperactivation and increased cytokines expression, such as interleukin-8 [chemoattractant chemokine (C-X-C motif) ligand 8 (CXCL8)]. Recently, new therapeutic strategies directly targeting the basic CFTR defect have been developed, and ORKAMBI (Vx-809/Vx-770 combination) is the only Food

BACKGROUND: 20(S)-protopanaxadiol (PPD), similar to several other anticancer agents, has low oral absorption and is extensively metabolized. These factors limit the use of PPD for treatment of human diseases.
METHODS: In this study, we used cubic nanoparticles containing piperine to improve the oral bioavailability of PPD and to enhance its absorption and inhibit its metabolism. Cubic nanoparticles loaded with PPD and piperine were prepared by fragmentation of glyceryl monoolein (GMO)/poloxamer 407 bulk cubic gel and verified using transmission electron microscopy and differential scanning ca

Inhibition of the endocannabinoid catabolic enzymes, monoacylglycerol lipase (MAGL) or fatty acid amide hydrolase (FAAH) attenuates naloxone-precipitated opioid withdrawal signs in mice via activation of CB1 receptors. Complete FAAH inhibition blocks only a subset of withdrawal signs, whereas complete MAGL inhibition elicits enhanced antiwithdrawal efficacy, but is accompanied with some cannabimimetic side effects. Thus, the primary objective of the present study was to determine whether combined, full FAAH inhibition and partial MAGL represents an optimal strategy to reduce opioid withdrawal.

BACKGROUND: Therapy with elexacaftor/tezacaftor/ivacaftor (ETI) works to improve the functionality of the cystic fibrosis (CF) transmembrane conductance regulator (CFTR) protein and has revolutionized CF treatment. However, the implications of ETI for airway barrier and systemic T cell immunobiology remain relatively little studied. METHODS: Here, we investigated the immunological effects of ETI at systemic and local pulmonary levels, using paired peripheral blood and sputum sampling, in relation to key clinical parameters. Samples were taken longitudinally at baseline (n = 27), and at three (

INTRODUCTION: Elexacaftor/Tezacaftor/Ivacaftor (ETI) has revolutionized cystic fibrosis (CF) care. In Argentina, the absence of patent restrictions allowed the production of a generic ETI formulation (ETIgf), significantly reducing costs.
OBJECTIVE: This study evaluates the real-world clinical impact of ETIgf in people with CF (pwCF).
METHODS: This prospective, single-center observational study in Buenos Aires, Argentina, included 71 pwCF (≥ 6 years) with responsive variants. Participants were categorized as: Group 1 (modulator-naïve) or Group 2 (previously on lumacaftor/ivacaftor). Clinical

Chloride channel potentiator and corrector elexacaftor/tezacaftor/ivacaftor (ETI) has reduced the burden of disease in people with cystic fibrosis (PwCF) and led to some patients ceasing high-burden treatment regimens. This study was a prospective survey and qualitative interview that evaluated medications PwCF had ceased after ETI. It demonstrated that a majority of PwCF have ceased medications, which prompts further research to rationalise medication regimens for PwCF.

Multidrug-resistant (MDR) Agrobacterium tumefaciens threatens healthcare and agriculture. Toxicity and environmental risks limit conventional plasmid-curing agents, prompting natural alternatives. Curcumin, piperine, and quercetin were tested for plasmid-curing against clinically isolated MDR A. tumefaciens. Morphological and 16S rRNA gene sequencing were used for identification of the isolate. Antimicrobial susceptibility and biofilm formation were measured pre- and post-treatment. Molecular docking was performed to analyze phytocompound-plasmid maintenance protein interactions. Broad-spectru

As a traditional medicinal-food ingredient, pepper roots were investigated for bioactive dimeric amide alkaloids. Eighteen pairs of novel anti-inflammatory dimeric amide alkaloids (1a/1b-18a/18b) and two known analogues (19 and 20) were isolated under the guidance of a feature-guided molecular networking (FGMN) strategy, with their structures established by extensive spectroscopic analyses and X-ray diffraction. A visible light photocatalytic method enabled the sustainable synthesis of the dimeric scaffold directly from piperine, a key component of Piper nigrum. In vitro assays demonstrated si

A key aspect of cystic fibrosis pathophysiology is the significant role played by neutrophils, central to the inflammatory response in cystic fibrosis airways. Neutrophil-dominated inflammation greatly influences clinical outcomes, including airway damage and lung function decline. People with cystic fibrosis have an increase in circulating neutrophils that exhibit numerous functional and phenotypic abnormalities. We used spectral flow cytometry to delineate the expression of 24 cell surface markers on neutrophils from 24 healthy donors and 45 adults with cystic fibrosis, before and after elex

BACKGROUND: The impact of CFTR modulator therapy on host immunity and outcomes in people with Cystic Fibrosis (CF)-related Aspergillus lung disease is poorly defined. We aimed to characterise fungal-relevant clinical outcomes post-CFTR modulators and assess effects on the Aspergillus-dependent Type I/III interferome.
METHODS: Biomarkers of Aspergillus-related lung disease (Aspergillus-specific IgE/IgG), anti-fungal and corticosteroid therapy were analysed in a retrospective cohort of people with CF pre and post Elexacaftor/Tezacaftor/Ivacaftor (ETI) modulator therapy. Homozygous F508del (CF)

BACKGROUND: Patient-derived intestinal organoids (PDIOs) are an in vitro tool used to predict clinical responses to CFTR modulators in people with cystic fibrosis (pwCF). Although strong genotype-based correlations have been observed, individual responses among F508del homozygous pwCF remain variable. We aimed to assess this correlation specifically within this group.
METHODS: A retrospective multi-center study included 60 F508del homozygous pwCF from six Belgian CF centers. The 15 highest and 15 lowest PDIO responders to tezacaftor/ivacaftor (TI) and elexacaftor/tezacaftor/ivacaftor (ETI) we

The advent of CFTR modulators has marked a breakthrough in cystic fibrosis management. Current monitoring relies on clinical data, spirometry, and CT. We report the interest of functional assessment using ventilation/perfusion (V/Q) SPECT/CT with 99m Tc in 3 patients treated with Elexacaftor/Tezacaftor/Ivacaftor (ETI). Two patients exhibited improvement in lung function on SPECT imaging, consistent with their clinical response, which was more pronounced than the structural changes observed on CT. The third patient exhibited no scintigraphic improvement, consistent with no clinical benefit. The

Chronic inflammatory conditions, such as psoriasis, require effective therapeutic strategies to manage both local and systemic inflammation. This study aimed to develop a thermosensitive hydrogel (Gel) based on Pluronic F127 and F68, incorporating NLC (Gel-NLC) loaded with anti-inflammatory phytochemicals, such as curcuminoids, resveratrol, and/or piperine, for the topical treatment of psoriasis. Phytochemicals-loaded NLC presented encapsulation efficiency > 90%, particle size values between 39-49 nm, and polydispersity indices <0.211. Compared to curcuminoids and resveratrol from NLC and Gel-

BACKGROUND: Piperine, a secondary metabolite, affects the antihyperlipidemic effect of Ezetimibe (EZ). Hyperlipidemia is one of the independent risk factors for cardiovascular disorders such as atherosclerosis. Antihyperlipidemic drugs are essential for reducing cardiovascular events and patient mortality. Our study aimed to improve the solubility of EZ, a lipid-lowering drug that belongs to BCS II and has low solubility. Piperine, a bioenhancer, can increase the bioavailability of other pharmaceuticals without modifying their fundamental characteristics or enhancing their efficacy. The object

Cystic fibrosis (CF), the most common hereditary lung disease in Caucasians, is caused by dysfunction of the cystic fibrosis transmembrane conductance regulator (CFTR). We evaluated CFTR function using a newly developed Ussing chamber system, the Multi Trans Epithelial Current Clamp (MTECC), in an in vitro model of human airway epithelia. Air-liquid interface (ALI) cultures were established from nasal brushings of healthy controls (HC) and CF patients with biallelic CFTR variants. ALI layer thickness was similar between groups (HC: 62 ± 13 µm; CF: 55 ± 9 µm). Immunofluorescence showed apical C

BACKGROUND: Clinical trials showed improved faecal elastase (FE-1) levels in younger children with cystic fibrosis (cwCF) on elexacaftor/tezacaftor/ivacaftor (ETI). Lower sweat chloride has been linked to better clinical outcomes in people with CF. Our previous work showed increased vitamin A levels in cwCF aged 5-15 years. We aimed to evaluate changes in FE-1 and vitamin levels and explore associations between sweat chloride and FE-1, in 2-6-year-olds starting ETI in a real-world setting.
METHODS: In a large UK paediatric specialist CF centre, cwCF newly eligible for ETI when UK licensing ag
