A graphene quantum dot-based method for the highly sensitive and selective fluorescence turn on detection of biothiols.

A facile assay method for the highly sensitive and selective sensing of biothiols based on graphene quantum dots (GQDs) has been developed. GQDs emitted strong blue fluorescence in an aqueous buffer solution. It was observed that mercury(II) ions could efficiently bind and quench the fluorescence of the GQDs. When a biothiol compound (glutathione, cysteine, or homocysteine) was added to the assay mixture of GQDs and mercury(II), it bound to mercury(II) ions. Hg(2+)-GQD complex dissociated, and a fluorescence turn-on signal was detected. The emission intensity changes of the GQDs could be direc

Regression modeling to inform cell incorporation into therapies for craniosynostosis.

Designing an appropriate tissue engineering solution for craniosynostosis (CS) necessitates determination of whether CS-derived cells differ from normal (wild-type, WT) cells and what assays are appropriate to test for differences. Traditional methodologies to statistically compare cellular behavior may not accurately reflect biologically relevant differences because they poorly address variation. Here, logistic regression was used to determine which assays could identify a biological difference between WT and CS progenitor cells. Quantitative alkaline phosphatase and MTS proliferation assays

Applicability of amino acid derivative reactivity assay (4 mM) for the prediction of skin sensitization by combining multiple alternative methods to evaluate key events.

The amino acid derivative reactivity assay (ADRA) is an alternative method for evaluating key event 1 (KE-1) in the skin sensitization mechanism included in OECD TG442C (OECD, 2021). Recently, we found that ADRA with a 4-mM test chemical solution had a higher accuracy than the original ADRA (1 mM). However, ADRA (4 mM) has yet to be evaluated using integrated approaches to testing and assessment (IATA), a combination of alternative methods for evaluating KE. In this study, the sensitization potency of three defined approaches (DAs) using ADRA (4 mM) as KE-1 was predicted and compared with thos

Von Willebrand factor (vWF) antigen levels and function in healthy Thais.

Type I von Willebrand disease (vWD) is very common in caucasians. Its genetic basis is possibly heterogenous, lying both within and out of the vWF gene locus. We sought to investigate vWF levels in the Thai population, to compare with those of western countries. The vWF antigen and activity were measured using ELISA and Collagen Binding Assay (CBA), respectively, in 311 healthy Thai volunteers. The mean age was 32.3, ranging from 18 to 75 years. Fifty-four percent were female. Low vWF antigen and activity (below 50 U/dl) were found in 3.5% and 10.2%, respectively. Around 75% and 20% of these c

Radioprotective effect of sulphydryl group containing triazole derivative to modulate the radiation- induced clastogenic effects.

Protection of biological systems against radiation damage is of paramount importance during accidental and unavoidable exposure to radiation. Several physico-chemical and biological factors collectively contribute to the damage caused by radiation and are, therefore, targets for developing radioprotectors. Chemicals capable of scavenging free radicals, relieving oxidative stress, promoting antioxidant activity and modulating immune response have been some of the radioprotectors extensively investigated with limited success. It has long been known that some of the most effective radioprotective

Dual Evidence of Trichoderma pleuroti Antagonism against Pleurotus pulmonarius: Direct Inhibition and Volatile-Mediated Bioassays with Volatilome Fingerprinting.

Trichoderma is known to cause green mold contamination, which severely threatens Pleurotus pulmonarius cultivation in tropical regions, including Malaysia, and can lead to major yield and economic losses. In this study, the contact-dependent and contactless antagonistic interactions between Trichoderma pleuroti and P. pulmonarius were investigated, and associated morphological changes and volatile profiles were characterized. Dual-culture assays recorded rapid overgrowth of P. pulmonarius by T. pleuroti, leading to growth suppression and mycelial discoloration. Light and scanning electron micr

From raw data to meaningful information: a robust but flexible method to assess in vitro assay responses-lessons learned from a novel Dicentrarchus labrax estrogen screen test.

Statistical analysis of in vitro assay data is a critical step towards a good interpretation of biological responses. However, it is still frequently undermined due to inappropriate statistical procedures, misinterpretation, insufficient statistical power-often resulting from small sample sizes-or poorly defined methodologies, in case of standardized tests. In line with practices commonly adopted in clinical studies and more recently in biomonitoring research, the use of thresholds for interpreting results may improve the robustness of conclusions. This work presents the application of a metho

Incorporating a dermal absorption route into high throughput toxicokinetic modeling.

BACKGROUND: Dermal absorption of chemicals represents an important route of exposure in pharmaceutical, occupational, and environmental settings. Thousands of chemicals with little toxicity or toxicokinetic (TK) data are in use. However, in many cases, in vitro bioactivity data are available. While it is not feasible to collect in vivo TK data, TK may be estimated using high-throughput methods. OBJECTIVE: This study developed a generalized physiologically-based TK (PBTK) dermal exposure model for in vitro-in vivo extrapolation (IVIVE). This model estimates dermal exposures that result in syst

Ecotoxicity assessment of primary producers in metal mining areas: biological indicators for ecosystem restoration.

Abandoned mines represent a considerable risk to ecosystems surrounding former exploitation sites. In metal mining, the exposure of waste significantly increases the mobility and bioavailability of potentially toxic elements (PTEs), affecting adjacent soils and organisms. Assessing the toxicity of mining waste involves challenges related to selecting appropriate bioassays and those recommended by current environmental regulations. Considering the expected increase in metal and metalloid extraction to supply critical raw materials, improving our understanding of the advantages and limitations o

Read-across prediction of nongenotoxic thyroid carcinogenicity of pesticides in rats using mechanism-based in vitro assay data.

Developing alternatives to rat carcinogenicity studies remains a major challenge. In our previous study, we found that while structural similarity provided a basis for grouping carcinogens, integrating mechanistically relevant in vitro cytotoxicity data substantially increased the accuracy of read-across predictions for tumors in the nasal cavity, stomach, and bladder/urethra. In this study, using the same dataset of 126 pesticides, we investigated whether a toxicity mechanism-based read-across approach could be applied to predict thyroid carcinogenicity. We evaluated nuclear receptor activati

GzESTY as an optimized cell-based assay for initial steps in GPCR deorphanization.

G protein-coupled receptors (GPCRs) are key pharmacological targets, yet many remain underutilized due to unknown activation mechanisms and ligands. Orphan GPCRs, lacking identified natural ligands, are a high priority for research, as identifying their ligands will aid in understanding their functions and potential as drug targets. Most GPCRs, including orphans, couple to Gi/o/z family members, however current assays to detect their activation are limited, hindering ligand identification efforts. We introduce GzESTY, a sensitive, cell-based assay developed in an easily deliverable format desi

High-throughput direct profiling of natural products in plant leaves using ambient ionization high-resolution mass spectrometry.

Plant leaves contain natural products that may have nutritional and medicinal values. In this work, a modified ambient ion source of angled direct analysis in real time (aDART) and high-resolution mass spectrometry (HRMS) was used for direct surface analysis of different plant leaves. Untreated leaves, leaves with the wax layer removed, and leaf imprints were all successfully analyzed by aDART-HRMS in min, observing abundant natural product information including esters, amino acids, saccharides, flavonoids, alkaloids, alcohols, and ketones/aldehydes. Different plant samples have their unique f

Phytochemical Analysis and Chymotrypsin Inhibitory Potential of Galium sp. and Solidago sp. via Effect-Directed HPTLC Bioassay.

Chymotrypsin inhibitors were initially considered mainly as anti-nutritional factors. However, the potential for their use as therapeutics has been recognized, particularly in the control of cancer, neurodegenerative diseases, and inflammatory processes. The search for new, effective, and safe chymotrypsin inhibitors has become important not only for food and feed safety reasons, but also in the search for new compounds with potential for use in the pharmaceutical industry. Oxidative stress is also an integral etiological factor in the development of the aforementioned pathological conditions.

The comet assay as a tool in human biomonitoring studies: Effects of confounding factors.

The comet assay is widely used in human biomonitoring studies of environmental and occupational exposures. However, it is clear from multiple studies that various types of confounding factors might affect the direct relationship between exposure and DNA damage in the comet assay. In addition to common confounders such as age, sex, and smoking, other factors considered to be important determinants for background levels of DNA damage in the comet assay include exhaustive physical exercise, chronic diseases, medical treatment, and diet. These are typically controlled in biomonitoring studies by r

Standardization of larval bioassays for determination of fluralaner susceptibility in Rhipicephalus microplus.

Fluralaner is an isoxazoline acaricide with potent activity against Rhipicephalus (Boophilus) microplus, a major tick species affecting cattle in South America. In this study, larval bioassays were performed to evaluate the baseline susceptibility of R. microplus populations from Brazil and Argentina to fluralaner. The larval packet test (LPT) was applied to Brazilian laboratory and field populations, while the larval immersion test (LIT) was used in Argentina. Both tests were carried with technical grade and a commercial formulation of fluralaner. LPT results showed consistent LC₅₀ values acr

Rapid visual detection of Giardia duodenalis in faecal samples using an RPA-CRISPR/Cas12a system.

Giardiasis is a common intestinal infection caused by Giardia duodenalis, which is a major economic and health burden for humans and livestock. Currently, a convenient and effective detection method is urgently needed. CRISPR/Cas12a-based diagnostic methods have been widely used for nucleic acid-based detection of pathogens due to their high efficiency and sensitivity. In this study, a technique combining CRISPR/Cas12a and RPA was established that allows the detection of G. duodenalis in faecal samples by the naked eye with high sensitivity (10-1 copies/μL) and specificity (no cross-reactivity

Chromatin Immunoprecipitation Sequencing (ChIP-Seq) Assay in Food Allergy Research.

Chromatin immunoprecipitation (ChIP) analysis followed by genome-wide sequencing represents the most efficient method for studying the complex interactions between nuclear proteins and their associated genomic DNA. This chapter aims to present a ChIP-Sequencing (ChIP-Seq) analysis protocol used for the investigation of different epigenetic modifications in diseases such as food allergy. Starting by treating cells with cross-linking reagents such as formaldehyde and ending up with profound bioinformatic analysis have made it plausible to understand how transcription factors and histone modifica

Detection of prions in matching post-mortem skin and cerebrospinal fluid samples using second-generation real-time quaking-induced conversion assay.

Real-time quaking-induced conversion assay (RT-QuIC) exploits templating activity of pathogenic prion protein for ultrasensitive detection of prions. We have utilized second generation RT-QuIC assay to analyze matching post-mortem cerebrospinal fluid and skin samples of 38 prion disease patients and of 30 deceased neurological controls. The analysis of cerebrospinal fluid samples led to 100% sensitivity and 100% specificity, but some samples had to be diluted before the analysis to alleviate the effect of present RT-QuIC inhibitors. The analysis of the corresponding skin samples provided 89.5%

Efficient Lung Cancer Molecular Diagnostics by Combining Next Generation Sequencing with Reflex Idylla Genefusion Assay Testing.

Molecular diagnostics for lung cancer is a well-established standard of care, but how to use the available diagnostic tools for optimal and cost-effective patient care remains unresolved. Here, we show that DNA-only, small gene next-generation sequencing (sNGS) panels (<50 genes) combined with ultra-rapid reflex testing for common fusion transcripts using the Idylla Genefusion assay provide a cost-effective and sufficiently comprehensive testing modality for the majority of lung cancer cases. We also demonstrate the need for additional reflex testing capability on larger DNA and fusion panels
