[Proteins involved in invasion of human red blood cells by malaria parasites].

Malaria is a disease caused by parasites of Plasmodium species. It is responsible for around 1-2 million deaths annually, mainly children under the age of 5. It occurs mainly in tropical and subtropical areas. Malaria is caused by five Plasmodium species: P. falciparum, P. malariae, P. vivax, P. knowlesi and P. ovale. Mosquitoes spread the disease by biting humans. The malaria parasite has two stages of development: the human stage and the mosquito stage. The first stage occurs in the human body and is divided into two phases: the liver phase and the blood phase. The invasion of erythrocytes b

Lectin-induced aggregates of blood cells from patients with acute coronary syndromes.

BACKGROUND: Cell surface glycoligands and circulating glycoproteins are believed to contribute to the pathogenesis of acute coronary syndromes (ACS) through cell aggregation/adhesion mechanisms. To characterize the glycobiological status of blood cells from patients with ACS, we used an advanced lectin-mediated aggregation technique allowing for detection of not only conventional lectin-induced cell aggregates but also their fraction resistant to haptenic/inhibitory sugars. METHODS: Peripheral blood samples were obtained from 24 patients with acute myocardial infarction or unstable angina and…

A simple method to irradiate blood cells in vitro with radon gas.

The design and dosimetry of a novel in vitro radon irradiation facility to investigate cytogenetic damage induced by radon and progeny is described. The system offers a 4pi geometry for uniform irradiation, proving a versatile and convenient facility for irradiation of whole blood cells in suspension or media. Doses can be controlled as exact volumes of the gas can be dispensed and measured by the Lucas cell. Irradiation of blood samples could be carried out in a unique and safe manner and the present study is an attempt to demonstrate the usefulness of a facility to expose blood lymphocytes i…

Characterization of mesenchymal stromal cells derived from full-term umbilical cord blood.

BACKGROUND: Multipotent mesenchymal stromal cells (MSC) are of interest for their potential to repair bone and cartilage, and also their immunosuppressive properties. Umbilical cord blood (UCB) is reported to contain MSC, and therefore may be a useful source of these cells for clinical applications. METHODS: We evaluated protocols for isolating MSC from UCB and characterized the surface phenotype, differentiation potential and immunoregulatory properties of the cells obtained. RESULTS: Ten of 25 UCB units processed yielded MSC-like colonies, with depletion of lineage+ cells providing a highe

Derivation of lung epithelium from human cord blood-derived mesenchymal stem cells.

RATIONALE: Recent studies have suggested that both embryonic stem cells and adult bone marrow stem cells can participate in the regeneration and repair of diseased adult organs, including the lungs. However, the extent of airway epithelial remodeling with adult marrow stem cells is low, and there are no available in vivo data with embryonic stem cells. Human umbilical cord blood contains both hematopoietic and nonhematopoietic stem cells, which have been used clinically as an alternative to bone marrow transplantation for hematologic malignancies and other diseases. OBJECTIVES: We hypothesize

Osmotic selection of human mesenchymal stem/progenitor cells from umbilical cord blood.

The isolation of undifferentiated adult stem/progenitor cells remains a challenging task primarily due to the rare quantity of these cells in biological samples and the lack of unique markers. Herein, we report a relatively straightforward method for isolation of human mesenchymal stem cells (MSCs) based on their unusual resistance to osmotic lysis, which we term "osmotic selection" (OS). MSCs can remarkably withstand significant exposure to hypotonic conditions (> 30 min) with only a reversible impairment in cell proliferation and with no loss of stem cell potential after exposure. Comparison

Blood cells cholinesterase activity in early stage Alzheimer's disease and vascular dementia.

Blood acetylcholinesterase (AChE) and butyrylcholinesterase (BuChE) activities have been studied as markers for Alzheimer's disease (AD), but their usefulness as a disease marker is controversial. To determine cholinesterase (ChE) activity during AD progression and whether ChE changes associate to other dementias, ChE activity was measured in lymphocytes, erythrocytes and platelets. Subjects underwent extensive medical and neuropsychological examination. Both early-AD and AD patients had lower AChE activity in lymphocytes compared to control subjects (p < 0.0001). In contrast, erythrocyte AChE

Diagnosis of human cytomegalovirus intrauterine infection using fetal cells from maternal blood.

OBJECTIVE: The sensitivity and specificity for the noninvasive prenatal diagnosis of human cytomegalovirus intrauterine infection were estimated by using isolating single fetal cells from maternal peripheral blood. METHODS: Micromanipulation techniques were employed to isolate single fetal nucleated erythroblasts from 273 maternal blood samples. SRY gene and HCMV-DNA in single fetal cells were detected by multiple primed in situ labeling (PRINS) from 76 HCMV-DNA positive samples of maternal peripheral blood. 273 samples of maternal peripheral blood were tested for SRY gene and HCMV-DNA in sin

[Immunological studies on the peripheral blood mononuclear cells in metal allergy patients].

Recently, the number of patients with dental metal allergy has risen significantly. The aim of this study was to evaluate the clinical availability of the lymphocyte stimulation test (LST) using metal reagents as a method for diagnosing metal allergy. In vitro, peripheral blood mononuclear cells (PBMC) from 18 patients with metal allergy and five healthy volunteers were examined by LST using 12 metal reagents (HAuCl₄, PdCl₂, NiSO₄, CoCl₂, CuSO₄, SnCl₄, K₂Cr₂O₇, MoCl₅, H₂PtCl₆, IrCl₄, AgBr, FeCl₃). To define metal allergy, a patch test (PT) was performed with metal reagents, and then the compon

Sensitivity of human cord blood cells to tetrachloroethylene: cellular and molecular endpoints.

The International Agency for Research on Cancer (IARC) currently lists tetrachloroethylene [perchloroethylene (PCE)] as being carcinogenic in animals. PCE is listed as possibly carcinogenic to humans upon occupational exposure. Human exposure to PCE can produce oesophageal cancer, cervical cancer, non-Hodgkin's lymphoma, urinary bladder cancer and leukemia. This work shows that PCE modulates the expression of some genes implicated in cancer induction, cell differentiation, cell-cycle progression, and the survival and clonogenic potential of human cord blood cells. After exposure to the compoun…

Pretransplant blood transfusions revisited: a role for CD(4+) regulatory T cells?

Pretransplant blood transfusions have been shown to improve organ allograft survival. However, the immunologic mechanism leading to this beneficial effect of blood transfusions is still unknown. The observation that transfusions sharing at least one HLA-DR antigen (human leukocyte antigen) with the recipient are more effective than HLA-mismatched transfusions has led to the hypothesis that CD(4+) regulatory T cells are induced that recognize allopeptides of the blood transfusion donor in the context of the self-HLA-DR molecule on the donor cells. In vitro studies showed that CD(4+) T cells rec

Antioxidant enzymes in red blood cells and lymphocytes of ataxia-telangiectasia patients.

Toxic oxygen metabolites may contribute to the development of tissue damage, and play a role in the pathogenesis of malignancies, some acute and chronic pulmonary diseases, and in cell damage by radiomimetic agents, which can be seen in patients with ataxia-telangiectasia (A-T). Oxidative stress resulting from increased free radical production and/or defects in antioxidant defences is also involved in neurodegenerative disorders. Thus, oxidative stress could account for several aspects of the pleiotropic phenotype of A-T patients. The aim of this study was to determine the activities of the en

Human amnion-isolated cells normalize blood glucose in streptozotocin-induced diabetic mice.

Whole pancreas or beta-cell transplantation has opened the way for the treatment of advanced stage of diabetes mellitus. However, it is always limited by the scarcity of transplantation materials. The amniotic membrane is part of the fetal membrane and is composed of amniotic epithelium (HAE) and mesenchymal (HAM) cells that are derived from the inner cell mass in the blastocyst. Thus, HAE and HAM cells may have the potential to differentiate into various organs. The aim of our study was to assess the possibility of HAE cells differentiating into insulin-producing cells. In vitro, HAE cells st

Effects of anoxia on megakaryocyte progenitors derived from cord blood CD34pos cells.

BACKGROUND: Severe hypoxic insults to the fetus and neonate are associated with the development of thrombocytopenia. The thrombocytopenia in some cases is the result of disseminated intravascular coagulation, but that mechanism fails to account for all, perhaps the majority, of cases. OBJECTIVE: We hypothesized that human fetal megakaryocyte (Mk) progenitors are directly adversely affected by transient anoxia. DESIGN AND METHODS: To test this, we isolated CD34pos cells from the umbilical cord blood of 10 healthy term neonates, and exposed these to 0% or 20% O2 for 24 h, with or without recom

Determination of the elastic shear modulus of cultured human red blood cells.

In this study we investigated the mechanical properties of in vitro cultured red blood cells (RBCs) in a liquid system. We used human umbilical cord blood as a highly efficient source of hematopoietic stem cells (HS). Our first goal was to establish an optimal medium composition in order to yield finally differentiated RBCs, i.e. enucleated and hemoglobin-filled cells. Different stages of cell differentiation were distinguished based on morphological observations and flow cytometry measurements. By means of the micropipette aspiration technique we estimated the deformability characteristics of…

Ex vivo expansion of CD56+ cytotoxic cells from human umbilical cord blood.

The immune-mediated effect of natural killer (NK) and cytotoxic T cells against residual tumor cells previously was shown to prevent relapse and reinduce remission after bone marrow transplantation. Human umbilical cord blood is a rich source of cytotoxic CD56+ cells including fetal NK cells (CD16(-)CD56+1) with high lytic capabilities upon activation with interleukin-2 (IL-2). Cord blood transplantations are reported to be associated with lower risk of graft-vs-host disease, which may jeopardize the graft-vs-leukemia effect. Therefore, our goal was to expand and amplify, ex vivo, cord blood-d

Peripheral blood T, B lymphocytes and NK cells in primary biliary cirrhosis.

Primary biliary cirrhosis (PBC) is an autoimmune disease with progressive destruction of the bile ducts and cholestasis. The most of epithelial cells in the bile ducts has molecule MHC I and II class expression which induced intensive immunological reaction and produced antimitochondrial autoantibodies (AMA). It leads to the destruction of own cells in PBC. The main part in the development of the disease have T cells, B cells, and NK cells. We analyzed subpopulation of lymphocytes in peripheral blood in 16 patients with PBC by flow cytometry. We noted statistically significant increase of NK C

Neopterin production in SCID mice injected with human peripheral blood mononuclear cells.

Intraperitoneal transfer of peripheral blood mononuclear cells (PBMC) from human EBV+ donors into severe combined immunodeficiency (SCID) mice is a suitable model for studying some aspects of lymphomagenesis and immune activation. Neopterin is a soluble immune marker which was found to be a useful indicator for immune activation processes in humans, e.g. to monitor immunological complications in allograft recipients or to predict prognosis in HIV-infected individuals. In contrast, this pteridine compound is normally synthesized in murine organism in only very low amounts. The measurement of ne
