Complementary Genetic Targeting and Monosynaptic Input Mapping Reveal Recruitment and Refinement of Distributed Corticostriatal Ensembles by Cocaine.

Drugs of abuse elicit powerful experiences that engage populations of neurons broadly distributed throughout the brain. To determine how synaptic connectivity is organized to enable robust communication between populations of drug-activated neurons, we developed a complementary targeting system for monosynaptic rabies virus (RV) tracing that identifies direct inputs to activated versus nonactivated neuronal populations. Analysis of over 100,000 synaptic input neurons demonstrated that cocaine-activated neurons comprise selectively connected but broadly distributed corticostriatal networks. Ele

Control of Feeding Behavior by Cerebral Ventricular Volume Transmission of Melanin-Concentrating Hormone.

Classical mechanisms through which brain-derived molecules influence behavior include neuronal synaptic communication and neuroendocrine signaling. Here we provide evidence for an alternative neural communication mechanism that is relevant for food intake control involving cerebroventricular volume transmission of the neuropeptide melanin-concentrating hormone (MCH). Results reveal that the cerebral ventricles receive input from approximately one-third of MCH-producing neurons. Moreover, MCH cerebrospinal fluid (CSF) levels increase prior to nocturnal feeding and following chemogenetic activat

Noninvasive ultrashort wave neuromodulation enhances glymphatic clearance to alleviate traumatic brain edema and Alzheimer's pathology.

The glymphatic system clears cerebral waste, yet strategies to enhance its function remain elusive. Here, we show that 40.68 MHz ultrashort wave (USW) rapidly enhances glymphatic bulk flow in healthy mice within 10 minutes. USW potentiates both glymphatic influx and efflux, concomitant with increased Delta wave neuronal synchronization, as confirmed by local field potentials and electroencephalograms. Chemogenetic silencing confirms that USW-driven glymphatic enhancement depends on neuronal activity, while pharmacological blockade of aquaporin-4 (AQP4) abolishes the effects, revealing dual rel

A cross-species AAV platform for precise microglial interrogation.

Targeting microglia with adeno-associated viruses (AAVs) remains challenging because of inefficient transduction, off-target expression, and inconsistent payload performance. Here, we develop an integrated, Cre-independent AAV platform that combines a compact 446-bp microglia-specific regulatory element (MSRE), a high-yield AAV2/9.m capsid, microRNA-mediated neuronal de-targeting, and a microglia-tailored codon-optimization strategy. Following direct intracranial delivery, the optimized vector achieves robust, long-lasting transgene expression with greater than 90% microglial specificity acros

Transcriptomic landscape of microglia in mouse models of social dysfunction and oxytocin-mediated recovery.

Atypical sociability is a hallmark of neurodevelopmental disorders arising from genetic susceptibility and prenatal environmental perturbations that can affect diverse brain cell types. Using single-cell transcriptomics, we previously identified selective vulnerability of parvocellular oxytocin (OT) neurons in the paraventricular hypothalamus (PVH) following embryonic exposure to valproic acid (VPA), a teratogen that induces social deficits. Neonatal chemogenetic activation of OT neurons rescued these behavioral abnormalities and partially restored dysregulated gene expression. However, the ef

Male and female mice scent mark during social communication regardless of sexual motivation or partner identity.

Many species communicate via pheromone signals that orchestrate key social and survival behaviors. In laboratory assays, only male mice are observed to broadcast pheromones through urine scent marking. This has contributed to the view that males have specialized circuits and anatomy to intentionally transmit urine pheromones and actively participate in socio-chemical communication, while females remain mute recipients in laboratory settings. Here, we find that simple modifications to standard assays reveal exuberant urine marking by laboratory females, enabling reexamination of its dimorphic f

Astrocyte-to-microglia purinergic signaling mediates synaptic shielding and promotes neuronal activity.

How glial cells sense and regulate neuronal hypoactivity remains poorly understood. Using in vivo imaging in anesthetized, sleeping, and behaving mice, we identified a glial purinergic signaling mechanism that converts neuronal hypoactivity into local circuit regulation. Cortical hypoactivity increased spatially confined ATP release through astrocytic pannexin-1 hemichannels, which were structurally enriched near perisomatic parvalbumin-positive boutons. Local ATP release directed microglial process movement and stabilized bulbous endings through microglial P2Y12 signaling, accompanied by loca

GABAergic Inhibition from the Nucleus Tractus Solitarius to Ventrolateral Medulla Phox2b Neurons Modulates Central Respiratory Chemoreflex and Ventilatory Homeostasis.

Central respiratory chemoreception is a vital homeostatic mechanism maintaining arterial blood gas levels. Central respiratory chemoreceptors in the retrotrapezoid nucleus (RTN) and nucleus tractus solitarius (NTS) form interconnected circuits to regulate respiratory homeostasis. We hypothesized that NTS GABAergic neurons (NTSGABA) modulate hypercapnic ventilatory responses by targeting ventrolateral medulla Phox2b neurons (VLMPhox2b), particularly Phox2b-expressing RTN neurons. Stimulation of NTSGABA neurons significantly attenuated CO2-evoked ventilatory responses, accompanied by a reduction

Chronic alcohol exposure contributes to postoperative cognitive dysfunction via NR2B upregulation in the hippocampus of adult mice.

Postoperative cognitive dysfunction (POCD) is a common perioperative complication of the central nervous system, and patients with chronic alcohol exposure frequently require surgery under anesthesia. However, the contribution of chronic alcohol exposure to POCD and its underlying mechanisms remains unclear. In this study, eight-week-old female and male mice were subjected to chronic alcohol exposure using the intermittent access two-bottle choice (IA2BC) paradigm. Compared with controls, mice with chronic alcohol exposure exhibited greater susceptibility to POCD. Specifically, chronic alcohol

Preoptic kisspeptin-nNOS-GnRH (KiNG) neuronal network regulates LH rhythmicity through activation-inhibition in mice.

Gonadotropin-releasing hormone (GnRH) neurons are the final target of a complex network regulating reproduction. The balance between excitatory and inhibitory inputs is essential for rhythmic GnRH secretion, including pulses and surges, yet the underlying mechanisms remain unresolved. Here, using adult animals of both sexes, we test the hypothesis that excitatory kisspeptin and inhibitory neuronal nitric oxide (NO) synthase (nNOS)-derived inputs orchestrate GnRH release within a microcircuit of kisspeptin, nNOS and GnRH neurons (the "KiNG" network). We focus on nNOS neurons of the organum vasc

Activation by Alcohol of Prefrontal Layer 5 Pyramidal Neurons Depends on Ascending Dopaminergic Input.

Short-term alcohol exposure modulates activity in reward-related regions such as the ventral tegmental area (VTA) and executive cortical regions such as the prefrontal cortex (PFC), potentially contributing to alcohol use disorders. Although alcohol induces dopamine release from VTA projection neurons, how dopamine signaling contributes to the acute effects of alcohol in the PFC at the cellular level remains unclear. Using in vivo Ca2+ imaging and in vitro slice electrophysiology in mice of either sex, we found that moderate-to-high doses of alcohol increased the activity of layer 5 (L5) pyram

PACAP neurons of the ventral dentate gyrus are not involved in fear acquisition.

The neural mechanisms underlying learned affective responses remain a core question in affective neuroscience. The ventral dentate gyrus (vDG) has been implicated in variants of fear conditioning (i.e., contextual and cued) and may rely on pituitary adenylate cyclase-activating polypeptide (PACAP) pathways, given their role in regulating stress responses and the formation of fear memories. The current study investigated whether PACAP neurons in the vDG are also involved in fear learning by chemogenetically exciting or inhibiting them in PACAP-Cre mice during fear acquisition. We first confirme

Engineering of genetically encoded programmable calcium channel inhibitory binders.

Store-operated Ca2+ release-activated Ca2+ (CRAC) channels, composed of STIM and ORAI, are essential for immune and developmental processes, and their dysregulation underlies channelopathies such as Stormorken syndrome. Here, we report the engineering of genetically encoded CRAC channel inhibitory binders (CRABs) derived from the ORAI C-terminal tail. Guided by deep mutational scanning, we optimize a membrane-anchored CRAB variant that potently inhibits Ca2+ influx and NFAT signaling, and rescues thrombocytopenia-like phenotypes in a zebrafish model of Stormorken syndrome. To enable tunable in

Norepinephrine acts through radial astrocytes in the developing optic tectum to enhance threat detection and escape behavior.

The ability to switch behavioral states is essential for animals to adapt and survive. Here, we demonstrate how norepinephrine (NE) activation of radial astrocytes alters visual processing in the optic tectum (OT) of developing Xenopus laevis. NE activates calcium transients in radial astrocytes through α1-adrenergic receptors. NE and radial astrocyte activation shift OT response selectivity to preferentially respond to looming stimuli, associated with predation threat. NE-mediated astrocytic release of ATP/adenosine reduces excitatory transmission by retinal ganglion cell axons, without affec

Activation of SNr GABA neurons drives liver-brain-eye axis dysfunction in hepatic encephalopathy.

Hepatic encephalopathy (HE) is frequently accompanied by visual dysfunction, yet the mechanisms underlying the liver-brain-eye axis remain unclear. We established mouse models of acute hepatic encephalopathy (AHE) using thioacetamide and chronic hepatic encephalopathy (CHE) using bile duct ligation, confirming hyperammonemia and visual impairment by electroretinogram (ERG) and visual evoked potentials (VEPs). Retinal analyses revealed preserved structure in AHE, whereas CHE induced significant thinning of the ganglion cell layer (GCL), inner nuclear layer (INL), and outer plexiform layer (OPL)

Locus coeruleus-spinal noradrenergic signaling drives sleep deprivation-induced hyperalgesia via astrocytic GAT-3.

Sleep deprivation is a prevalent health problem that amplifies pain and prolongs recovery after tissue injury, yet the neural and astrocytic mechanisms underlying this relationship remain poorly defined. Here, we demonstrate that sleep deprivation activates locus coeruleus noradrenergic neurons and elevates spinal norepinephrine levels. Norepinephrine acts on spinal dorsal horn astrocytes through α1-adrenergic receptors, upregulating the astrocyte-enriched GABA transporter GAT-3. The increase in astrocytic GAT-3 was accompanied by reduced spinal GABA content and enhanced activation of spinopar

Activation of G12-type G proteins in pancreatic β cells protects against impaired glucose homeostasis.

Impaired insulin secretion represents a key feature of type 2 diabetes (T2D), a major global health burden. Insulin release from pancreatic β cells is regulated by G protein-coupled receptors linked to different functional classes of G proteins. The potential metabolic role of β cell G12/13 (G12 hereafter) signaling remains unexplored. Thus, we studied the potential metabolic outcomes of β cell G12 activation by using a combination of mouse gene knockout technology and β cell-specific chemogenetic approaches. We found that selective activation of β cell G12 signaling strongly promoted insulin

A cross-species atlas of the dorsal vagal complex reveals neural mediators of the effects of cagrilintide on energy balance.

Amylin receptor agonists such as cagrilintide represent emerging obesity therapies. To understand mediators of cagrilintide action, we generated a transcriptomics atlas of over 530,000 cells comprising 80 neuronal cell populations across rat, mouse and macaque caudal brainstem, with spatial profiling to map distribution in the rat dorsal vagal complex (DVC). Here we show that cagrilintide regulates two conserved Calcr-expressing DVC neuronal populations. While acute cagrilintide treatment alters gene expression in area postrema Calcr/Ramp3 neurons, chemogenetic activation in rats fails to affe

The mPFC-reuniens-hippocampus pathway links brain circuitry and neural plasticity in antidepressant response.

The pathophysiology of depression involves multiple biological processes, including circuit dysfunction and impaired neuroplasticity, yet an integrative view linking these processes remains elusive. Here, we identify a convergent circuit for antidepressant response and plasticity modulation. We demonstrate that chemogenetic activation of the infralimbic cortex (IL) exerts rapid antidepressant-like effects across multiple behavioral domains in a mouse model of stress-induced depression. IL stimulation exerts top-down control over the hippocampus, enhancing structural plasticity, restoring long-

The PRC2.1 subcomplex opposes G1 progression through regulation of CCND1 and CCND2.

Progression through the G1 phase of the cell cycle is the most highly regulated step in cellular division. We employed a chemogenetic approach to discover novel cellular networks that regulate cell cycle progression. This approach uncovered functional clusters of genes that altered sensitivity of cells to inhibitors of the G1/S transition. Mutation of components of the Polycomb Repressor Complex 2 rescued proliferation inhibition caused by the CDK4/6 inhibitor palbociclib, but not to inhibitors of S phase or mitosis. In addition to its core catalytic subunits, mutation of the PRC2.1 accessory
