Citrullination of tissue factor pathway inhibitor alpha by peptidylarginine deiminase 4 impairs its natural anticoagulant activity toward factors Xa and VIIa/tissue factor and reduces binding to its cofactor protein S.

BACKGROUND: Neutrophils are known to externalize their DNA and intracellular contents to neutralize invading pathogens. This process may enhance blood coagulation during inflammation. Tissue factor (TF) pathway inhibitor (TFPI) binds to extracellular DNA and may be citrullinated by peptidylarginine deiminase 4. Citrullination of TFPI reduces its anticoagulant activity toward factor (F)Xa but appears to retain its inhibition of TF-triggered thrombin generation, indicating differential regulation of TFPI functions by peptidylarginine deiminase 4. OBJECTIVES: This work aimed to study the effects…

Novel Biomarker for Rheumatoid Arthritis: Serum Peptidylarginine Deiminase 4-Potentiating Activity in an Anti-Citrullinated Protein Antibody-Positive At-Risk Population.

Enzymes like peptidylarginine deiminase 4 (PAD4) generate citrullinated epitopes that are crucial in triggering and driving the anti-citrullinated protein antibody (ACPA) response in rheumatoid arthritis. Here, we assessed whether serum PAD4-potentiating activity is associated with subsequent arthritis development among ACPA-positive individuals during the pre-clinical phase. The study included 267 ACPA-positive individuals from the Karolinska Risk-RA cohort, among whom 101 (37.8%) developed arthritis within the observational period (range 22-60 months). We measured the ability of serum to pot

Pronounced Reductions in Plasma Citrulline Indicate Severe Intestinal Mucosal Barrier Injury During Induction Therapy for Pediatric Acute Myeloid Leukemia.

OBJECTIVES: Pediatric acute myeloid leukemia (AML) chemotherapy regimens are frequently burdened by fever episodes, systemic inflammation and bloodstream infections (BSI). Mucosal barrier injury may contribute to these complications, but its role in childhood AML remains uninvestigated. This study assessed mucosal barrier injury using plasma citrulline and examined associations with inflammation, fever, and BSI. METHODS: Twenty children (1-16 years) with AML were prospectively studied during two induction courses (NOPHO-DBH AML 2012 protocol). Plasma citrulline was measured weekly from days 1

Prognostic Value of Circulating NETosis Biomarkers in ST-Elevation Myocardial Infarction: Citrullinated Histone H3 Outperforms Cell-Free DNA and Nucleosomes.

BACKGROUND: Even after successful primary percutaneous coronary intervention (PPCI), a substantial proportion of ST-elevation myocardial infarction (STEMI) patients develop major adverse cardiovascular events (MACE). Neutrophil extracellular trap formation (NETosis) contributes to atherothrombosis and coronary microvascular injury, but the prognostic value of circulating NETosis biomarkers in STEMI has not been systematically compared. METHODS: We studied 420 STEMI patients who survived index hospitalisation after PPCI. Plasma citrullinated histone H3 (H3Cit), cell-free DNA (cfDNA) and nucleo

Could serum intestinal fatty acid-binding protein and citrulline levels be predictive markers of mortality in critically ill COVID-19 patients?

INTRODUCTION: The global Coronavirus Disease 2019 (COVID-19) pandemic has been demonstrated to cause severe acute respiratory syndrome and is frequently associated with gastrointestinal (GI) manifestations. Intestinal fatty acid binding protein (IFABP) and citrulline have been identified as potential biomarkers of intestinal (dys)function. The present study was undertaken to ascertain the relationship between serum IFABP and citrulline concentrations and mortality in patients with COVID-19. METHODOLOGY: This observational study was conducted in the medical intensive care unit (ICU) and includ

L-Citrulline Alleviates Testicular Ischemia/Reperfusion Injury in Rats by Modulating eNOS/iNOS Induced Nitric Oxide Production, Inflammation, and Apoptosis.

Testicular ischemia/reperfusion injury (TI/RI) is a significant clinical contributor to subfertility and infertility resulting from testicular torsion and subsequent detortion. Insufficient nitric oxide (NO) synthesis in TI/RI can result in endothelial dysfunction, as the vascular endothelium fails to produce sufficient NO to sustain appropriate vasodilation and blood perfusion. Many studies have found that NO plays an important role in the I/RI and its increase or decrease can affect the progression and outcome of I/RI. However, the role of NO in I/RI is controversial and complicated. NO deri

Influence of body mass index on cardiovascular risk in rheumatoid arthritis varies across anti-citrullinated protein antibody status and biologic use.

OBJECTIVES: The impact of body mass index (BMI) on cardiovascular risk in rheumatoid arthritis (RA) is unclear. RA characteristics may influence the association between BMI and risk. Disease activity, which predicts cardiovascular risk, is associated with obesity only among anticitrullinated antibody (ACPA)-positive patients. Biologics alter body composition and mitigate cardiovascular risk in RA. We explored the association of BMI with cardiovascular risk and whether this varied across ACPA status and biologic use. METHODS: We evaluated 3982 patients from an international observational cohor

Diagnostic Utility of Anti-Cyclic Citrullinated Peptide Antibodies Versus Rheumatoid Factor in Libyan Patients with Rheumatoid Arthritis: A Cross-Sectional Study.

BACKGROUND: Rheumatoid arthritis (RA) is a chronic autoimmune disease characterized by synovial inflammation and progressive destruction of joint structures. Anti-cyclic citrullinated peptide (anti-CCP) antibodies have emerged as highly specific biomarkers, whereas rheumatoid factor (RF) demonstrates lower diagnostic specificity. OBJECTIVE: To assess the diagnostic utility of anti-CCP in comparison with rheumatoid factor (RF) in Libyan patients with RA and to investigate their associations with immunological markers, demographic characteristics, comorbid conditions, and clinical manifestation

Factors related to seroconversion of anti-cyclic citrullinated peptide antibody after the onset of rheumatoid arthritis: a case-control study and systematic review.

Anti-cyclic citrullinated peptide (CCP) antibodies typically present before rheumatoid arthritis (RA) but appear (seroconvert) after disease onset in some patients. This study analyzed factors related to anti-CCP seroconversion. Fifty-six consecutive patients with anti-CCP negative RA were enrolled. The first determination of anti-CCP status was defined as the baseline. Anti-CCP was then reassessed with an interval of 77 (±41) months. Demographics and baseline characteristics were compared between patients with and without anti-CCP seroconversion. Moreover, relevant studies were systematically

Impact of high-intensity interval training with or without l-citrulline on physical performance, skeletal muscle, and adipose tissue in obese older adults.

BACKGROUND: Aging is associated with a progressive decline in skeletal muscle mass and strength as well as an increase in adiposity. These changes may have devastating impact on the quality of life of older adults. Mitochondrial dysfunctions have been implicated in aging-related and obesity-related deterioration of muscle function. Impairments in mitochondrial quality control processes (biogenesis, fusion, fission, and mitophagy) may underlie this accumulation of mitochondrial dysfunction. High-intensity interval training (HIIT) was shown to improve muscle and mitochondrial function in healthy

Folic acid, either solely or combined with L-citrulline, improves NO signaling and ameliorates chronic hypoxia-induced pulmonary hypertension in newborn pigs.

Concomitant with developing pulmonary hypertension (PH), newborn piglets exposed to chronic hypoxia develop pulmonary vascular NO signaling impairments. PH is reduced and NO signaling is improved in chronically hypoxic piglets treated with the NO-arginine precursor, L-citrulline. Folic acid positively impacts NO signaling. We evaluated whether the effect on NO signaling and PH is greater using co-treatment with folic acid and L-citrulline than either alone. From day 3 to day 10 of hypoxia, piglets were treated solely with folic acid, solely with L-citrulline, or co-treated with both. Catheters

Antibodies to Cyclic Citrullinated Peptides in Patients With Juvenile Idiopathic Arthritis and Patients With Rheumatoid Arthritis: Shared Expression of the Inherently Autoreactive 9G4 Idiotype.

OBJECTIVE: Antibodies to cyclic citrullinated peptides (anti-CCP) in rheumatoid arthritis (RA) can express the inherently autoreactive gene VH 4-34, detected using the rat monoclonal antibody 9G4. Patients with the polyarticular subtype of juvenile idiopathic arthritis (JIA) share some but not all of the features of adult patients with RA. This study was undertaken to compare serologic findings for rheumatoid factor (RF), anti-CCP, and 9G4-expressing anti-CCP in a large JIA cohort with a cohort of adult RA patients. METHODS: Serum from 88 patients with polyarticular JIA, 29 patients with enth

Association of Antibodies to Citrullinated Protein Antigens with Blood Pressure in First-Degree Relatives of Rheumatoid Arthritis Patients: The Studies of the Etiology of Rheumatoid Arthritis.

BACKGROUND: Hypertension is more common in patients with rheumatoid arthritis (RA) than in the general population. It is unknown whether hypertension is due to RA-related medications or the disease itself. Therefore, we sought to investigate associations between RA-related autoantibodies, specifically antibodies to citrullinated protein antigens (ACPA) and systolic blood pressure (SBP) and diastolic blood pressure (DBP) in first-degree relatives of RA patients, who were free of RA and RA-related medications. We hypothesized that a greater number of detectable ACPA would be associated with high
