Cycling status of CD34+ cells mobilized into peripheral blood of healthy donors by recombinant human granulocyte colony-stimulating factor.

In this study, we assessed the functional and kinetic characteristics of highly purified hematopoietic CD34+ cells from the apheresis products of 16 normal donors undergoing glycosylated granulocyte colony-stimulating factor (G-CSF) treatment for peripheral blood stem cells (PBSC) mobilization and transplantation in allogeneic recipients. Mobilized CD34+ cells were evaluated for their colony-forming capacity and trilineage proliferative response to selected recombinant human (rh) CSF in vitro and the content of very primitive long-term culture initiating cells (LTC-IC). In addition, the cyclin

Abnormal in vitro differentiation of clonogenic B-cells in common acute lymphoblastic leukemia in complete remission. A marker for minimal residual disease?

An in vitro B-cell colony assay system was used to evaluate B-cell differentiation from peripheral blood precursors in common acute lymphoblastic leukemia (cALL) patients in remission as compared to normal controls. Significant differences in the morphologic and phenotypic features of pooled colony cells were found between the two groups. The morphology and surface markers of control-cultured cells were those of young plasmocytes. In contrast, patients' cells had predominantly a lymphoblastoid appearance and a mean of 18% (2-72%) of the cells expressed the cALL (CALLA) antigen. This marker, kn

Long-term marrow culture of cells from patients with acute myelogenous leukemia. Selection in favor of normal phenotypes in some but not all cases.

Long-term cultures were initiated with leukemic marrow aspirate cells from each of 13 newly diagnosed acute myelogenous leukemia (AML) patients. Initial assessment of the clonogenic potential of the marrow suggested that normal hemopoietic progenitors were reduced in most cases and progenitors of abnormal colonies and clusters were present in 10 cases. Subsequent assays of both nonadherent and adherent fractions of long-term cultures revealed two patterns of progenitor cell behavior. The most common pattern (nine cases) featured the detection after 1-4 wk of near normal numbers of typical eryt

Hormone dependency of human breast neoplasms cultured in vitro in the stem cell assay.

The stem cell soft agar culture system provides a biological tool with which to study tumor clones derived from heterogeneous tumor cell populations. For testing the feasibility of this approach in identifying hormone-responsive clones of mammary tumors, a study was done to determine whether estrogen deprivation or supplementation would alter colony formation in sequential breast neoplasms received in the Cell Kinetics Laboratory, The Pennsylvania State University. Single-cell suspensions obtained from 22 freshly excised breast neoplasms (20 malignant, 2 benign) were plated in soft agar with a

The effect of processed meat and meat starter cultures on gastrointestinal colonization and virulence of Listeria monocytogenes in mice.

Listeria monocytogenes is a foodborne pathogen of major concern to the food industry in general and the meat industry in particular. The aim of this study was firstly to identify a strain of Listeria that was virulent in SPF BALB/c mice. Secondly, to investigate if a traditional meat starter culture (FloraCarn) and nontraditional meat starter (NTMS) cultures of dairy product and human origin (Lactobacillus and bifidobacteria) inhibit this pathogen in vivo. In addition, the inhibition of Listeria was investigated in vitro. In vitro inhibition was investigated using an agar inhibition assay, whe

Nitric oxide mediates immunosuppression induced by Listeria monocytogenes infection: quantitative studies.

Our laboratory has shown that immunization of mice with an attenuated strain of Salmonella typhimuriuminduces profound suppression in the capacity of splenocytes to mount an in vitro antibody plaque-forming cell (PFC) response to sheep red blood cells (SRBC) and to proliferate in response to mitogens. In vitro addition of NG-monomethyl-L-arginine (NMMA), an inhibitor of nitric oxide (NO) synthase, to cell cultures from Salmonella-immunized mice completely blocked suppression of the PFC responses, implicating that NO is the suppressor factor. The present study quantified the role of nitric oxid

Effect of surface polishing on roughness, biofilm formation, and biocompatibility of LCD-printed denture base polymer.

This study investigated the effect of surface polishing on roughness, mechanical performance, and biological properties of a liquid crystal display (LCD)‑printed denture base polymer. Disc‑shaped samples of three-dimensional (3D) printing polymer were fabricated and allocated to polished (P) or not polished (NP) groups for comparative analysis. Polishing was performed manually in a standardized manner using sequential silicon carbide sandpapers, while the NP samples were sanded to reproduce the texture generated by the printing lines. Surface roughness was measured by contact and non‑contact p

Optimized Surveillance Testing for Toxigenic Clostridioides difficile to Scale Prevention Programs Across Clinical Settings.

Clostridioides difficile is the leading cause of healthcare acquired infections (HAIs). Healthcare systems lack scalable strategies to identify patients colonized with toxigenic strains to inform prevention programs. We developed a scalable program using rectal swabs, collected for vancomycin resistance Enterococci surveillance, with ex vivo amplification of C. difficile in Cdiff Banana Broth (BB). Optimized confirmatory testing of BB-positive tubes used the rapid C. difficile QuickCheck assay with Cepheid's Xpert® C. difficile/Epi (Xpert) to confirm toxin gene carriage for QuickCheck results

233 nm far-UV-C radiation for biofilm inactivation.

The formation of biofilms protects bacteria from antibiotic therapy or disinfection based on different mechanisms. The treatment of biofilms is furthermore impeded by their characteristic of being composed of multiple species; thus, it is necessary to identify involved species to determine an appropriate treatment. Consequently, new methods for inactivating involved bacteria in biofilms are necessary. One possibility might be the use of skin-compatible doses of 233 nm far-UV-C irradiation. In the presented study, the potential for biofilm formation of various bacteria species and strains was i

Brassicaceae-Based Biosolarization Reduces Lettuce Fusarium Wilt (FOLac) in Naturally Infested Soils.

Brassicaceae-based bio-disinfestation is a promising non-chemical option to manage lettuce Fusarium wilt caused by Fusarium oxysporum f. sp. lactucae (FOLac), but evidence integrating pathogen suppression, residue chemistry, and soil microbiome response remains limited. We conducted a greenhouse mesocosm assay with naturally infested soil to compare biofumigation (uncovered soil) and biosolarization (plastic-covered soil for 40 days) using five Brassicaceae residues (Brassica carinata, B. juncea, B. napus, Sinapis alba and Raphanus sativus) applied at two field-equivalent doses. Comparison of two models of biofilm formation on reusable stainless steel medical device material.

BACKGROUND: Reusable medical devices require reprocessing before subsequent patient use. Inadequate reprocessing can create suitable conditions for contaminating microbes to form biofilm. Despite this threat, there is a lack of guidance for medical device manufacturers to make biofilm assessment on their device labelling and instructions for use. Currently, there are no US Food and Drug Administration-recognized standardized models for biofilm formation on medical devices or device materials. AIM: To assess established standard methods for their suitability to be used as validation tools for

Comparative Evaluation of Antimicrobial Efficacy of Fluoride-Based and Self-Assembling Peptide P11-4-based Tooth Remineralization Agents on Streptococcus mutans: A Microbiological Study.

BACKGROUND: Dental caries is a biofilm-related oral disease that continues to afflict the majority of the world's population. The disease results from an interaction between specific bacteria and dietary constituents within a biofilm known as dental plaque. Among the cariogenic microorganisms, Streptococcus mutans (S. mutans) plays pivotal role in caries-inducing processes. OBJECTIVES: Evaluate and compare the antimicrobial efficacy of self-assembling peptide P11-4-based tooth remineralization agents on S. mutans. MATERIALS AND METHODS: An in vitro microbiological study. The antibacterial ef

Assessment of changes in Streptococcus pyogenes levels using N-acetylgalactosamine-6-sulfatase marker and pharyngeal airway space with appliance therapy in mouth breathers - An ELISA-based study.

BACKGROUND: The frequency of adenotonsillar hypertrophy in mouth-breathing children when compared to the average found in the general population is considered to be higher. Mouth breathing is considered as one of the causative factors for tonsillitis in children. Through continuous irritation on tonsillar wall, tonsils swell up and inflammation develops. PURPOSE: The purpose of the study is to evaluate Streptococcus pyogenes count using colony-forming units (CFUs) and N-acetylgalactosamine-6-sulfatase side chain marker on ELISA (enzyme linked immunosorbent assay) in mouth breathers and to est

Tolerance of biofilm of a carbapenem-resistant Klebsiella pneumoniae involved in a duodenoscopy-associated outbreak to the disinfectant used in reprocessing.

BACKGROUND: One possible transmission route for nosocomial pathogens is contaminated medical devices. Formation of biofilms can exacerbate the problem. We report on a carbapenemase-producing Klebsiella pneumoniae that had caused an outbreak linked to contaminated duodenoscopes. To determine whether increased tolerance to disinfectants may have contributed to the outbreak, we investigated the susceptibility of the outbreak strain to disinfectants commonly used for duodenoscope reprocessing. Disinfection efficacy was tested on planktonic bacteria and on biofilm. METHODS: Disinfectant efficacy t

Development of a large animal rabbit model for chronic periprosthetic joint infection.

AIMS: Periprosthetic joint infections (PJIs) and osteomyelitis are clinical challenges that are difficult to eradicate. Well-characterized large animal models necessary for testing and validating new treatment strategies for these conditions are lacking. The purpose of this study was to develop a rabbit model of chronic PJI in the distal femur. METHODS: Fresh suspensions of Staphylococcus aureus (ATCC 25923) were prepared in phosphate-buffered saline (PBS) (1 × 109 colony-forming units (CFUs)/ml). Periprosthetic osteomyelitis in female New Zealand white rabbits was induced by intraosseous inj

The smart activatable P2&3TT probe allows accurate, fast, and highly sensitive detection of Staphylococcus aureus in clinical blood culture samples.

Staphylococcus aureus bacteraemia (SAB) is associated with high mortality and morbidity rates. Yet, there is currently no adequate diagnostic test for early and rapid diagnosis of SAB. Therefore, this study was aimed at exploring the potential for clinical implementation of a nuclease-activatable fluorescent probe for early diagnosis of SAB. To this end, clinical blood culture samples from patients with bloodstream infections were incubated for 1 h with the "smart" activatable P2&3TT probe, the total assay time being less than 2 h. Cleavage of this probe by the secreted S. aureus enzyme microc
