We characterized protective immune response in mice infected with the attenuated Japanese encephalitis virus (JEV) strain SA(A) derived from the live JE vaccine strain SA14-14-2. In the sera of mice infected with SA(A), antibodies to JEV envelope (E) and non-structural NS1 proteins were detected by Western blot analysis and neutralization, haemagglutination-inhibition (HAI), and complement fixation (CF) antibodies were detected by serological tests. We have developed a one-point microcapsule agglutination test (MCAT) for the serodiagnosis of leptospirosis. The MCAT kit was evaluated for use in humans by six WHO Collaborating Centres for Reference and Research on Leptospirosis. The laboratories classified their serum samples on the basis of the microscopic agglutination test (MAT) and the following screening tests: enzyme-linked immunosorbent assay (ELISA), macroscopic (slide) agglutination test, or the complement fixation test. Direct examination of urethral smears made evident the presence of chlamydial infection in 162 (38.1%) of 425 male patients with nongonococcal urethritis. Serum antibodies to the group antigen could be detected by complement fixation and microagglutination reactions only in about 50% of the cases with chlamydial infection. The proportion of chlamydial strains isolated in chick embryos was rather low (14.8%) and the isolation rate appeared to correlate with the abundance of urethral secretion.

A passive microcapsule agglutination test for the diagnosis of leptospirosis was developed by utilizing chemically stable microcapsules instead of sheep erythrocytes. In the test, sonically disrupted antigens of leptospira were sensitized to microcapsules treated with glutaraldehyde. Compared with the microscopic agglutination test, the passive microcapsule agglutination test showed a relatively genus-specific tendency and a 4- to 32-fold-higher sensitivity. The detection of antibodies in subjects with visceral larva migrans syndrome caused by Toxocara canis is usually carried out with antigenic larvae, whose preparation presents some difficulties. Results obtained using extracts prepared from adult parasites show that such antigens are quite suitable for the detection of antibodies present in visceral larva migrans syndrome. This phenomenon is easily understood when one considers that there are numerous antigenic points in common between the different stages of the evolution of this parasite.
