Functional convergence amid taxonomic variability in gut microbiome-immune checkpoint inhibitor research: a bibliometric and mechanistic synthesis.

Immune checkpoint inhibitors (ICIs) have transformed cancer therapy, yet clinical responses remain highly variable, and microbiome-associated findings lack reproducibility across studies. Increasing evidence implicates the gut microbiome in modulating ICI efficacy; however, findings remain inconsistent at the taxonomic level, raising the possibility that functionally convergent immunological mechanisms may underlie this apparent variability. To address this, a critical synthesis was conducted, integrating bibliometric mapping of publications indexed in the Web of Science Core Collection (2013-

Nanomedicine-enabled in situ vaccination: Advancing a trilogy of immunogenic cell death induction, antigen presentation potentiation, and microenvironment remodeling.

In situ cancer vaccination based on immunogenic cell death (ICD) represents a paradigm-shifting strategy in cancer immunotherapy, leveraging the tumor cells itself as an endogenous source of patient-specific antigens and damage-associated molecular patterns (DAMPs). This approach circumvents the need for exogenous antigen identification while minimizing off-target toxicities. Nanomedicine serves as a pivotal enabler in this paradigm by providing sophisticated spatiotemporal control over the critical stages of the antitumor immunity cycle. Specifically, nanodelivery systems are engineered to po

Programming regulated cell death to engineer whole-tumor-cell vaccines for cancer immunotherapy.

Regulated cell death (RCD) encompasses a variety of genetically controlled cell death programs that are essential for maintaining tissue homeostasis and have emerged as promising therapeutic targets in oncology. When used to prepare whole-tumor-cell (WTC) vaccines, dying cancer cells provide broad tumor-antigen repertoires together with damage-associated molecular patterns (DAMPs) that can function as endogenous adjuvants. This review examines how distinct RCD pathways shape antitumor immunity and how they can be exploited to engineer next-generation WTC vaccines. We summarize the molecular an

Combinatorial in situ cancer vaccines: unlocking broad and enhanced antitumor responses.

In situ cancer vaccination, also termed intratumoral immunotherapy, transforms the tumor microenvironment into an endogenous vaccine platform by leveraging the tumor itself as a source of antigens. Unlike conventional tumor-associated antigen (TAA) or personalized neoantigen vaccines that require predefined targets and complex manufacturing, in situ cancer vaccination presents the tumor's full antigenic repertoire, including TAAs, neoantigens, post-translationally modified epitopes, cryptic peptides, and viral antigens within their native context. This broad antigen exposure elicits robust pol

Exposome-driven molecular sensitization signatures in pediatric rhinitis and asthma across Spanish regions.

BACKGROUND: Allergic rhinitis and asthma in childhood arise from dynamic interactions between genetic predisposition and the exposome. However, how environmental heterogeneity shapes molecular IgE sensitization profiles during early life remains insufficiently characterized. METHODS: We conducted a prospective, observational, multicentre, cross-sectional study including 354 children (0-15 years) with allergic rhinitis and/or asthma recruited across eight Spanish cities contrasting regional climatic contexts. All patients were skin prick test-positive to at least one aeroallergen and underwent

COVID-19 mRNA Vaccines as Antigen-Agnostic Immunomodulators: A Repurposing Perspective.

Since 2020, billions of doses of COVID-19 messenger RNA (mRNA) lipid nanoparticle vaccines have been administered worldwide, generating one of the largest pharmacovigilance databases for a modern therapeutic class. Intriguingly, their immunopharmacologic profile-including the engagement of innate nucleic acid sensors, systemic type I interferon signaling, dendritic cell-mediated cross-priming of CD8+ T cells, and upregulation of programmed cell death ligand 1 (PD-L1)-may extend beyond COVID-19 prophylaxis. Retrospective evidence from six independent cohorts, one of which is a multinational hea…

Effects of parental care on skin microbial community composition in poison frogs.

Parent-offspring interactions constitute the first contact of many newborns with their environment, priming community assembly of microbes through priority effects and shaping host health and disease. Microbe acquisition during parental care is well studied in humans and agriculturally relevant species but remains poorly understood in other vertebrate groups, such as amphibians. Here, we investigate vertical transmission of skin microbiota in poison frogs (Dendrobatidae), where fathers transport tadpoles piggyback-style from terrestrial clutches to aquatic nurseries. We found that substantial

Epigenetic modulation of antitumor immunity for improved cancer immunotherapy.

Epigenetic mechanisms play vital roles not only in cancer initiation and progression, but also in the activation, differentiation and effect … tumor immunogenic cell death (ICD). ICD provides a rich source of immunogens for anti-tumor T cell cross-priming and sensitizing cancer cells to interventional immunotherapy. In this way, epigenetic modulators may be envisioned as effective components in combination immunotherapy approaches capable of mediating superior therapeutic efficacy.

Early IL-17 production by intrahepatic T cells is important for adaptive immune responses in viral hepatitis.

This study was conducted to examine the interactions among the innate and adaptive immune components of the liver parenchyma during acute viral hepatitis. Mice were i.v. infected with a recombinant adenovirus, and within the first 24 h of infection, we found a transient but significant accumulation of IL-17 and IL-23 in the liver. In vivo neutralization of these interleukins alleviated the liver injury. Further investigations showed that IL-17 neutralization halted the intrahepatic accumulation of CTLs and Th1 cells. A majority of the IL-17-producing cells in the liver were γδ T cells. Additio

Vitellogenin detection in Caiman latirostris (Crocodylia: Alligatoridae): a tool to assess environmental estrogen exposure in wildlife.

Environmental pollution with endocrine disrupting compounds (EDCs) has adverse effects on the ecosystem's health. Caiman latirostris are widely distributed in South American aquatic ecosystems. Caimans have physiological and ecological characteristics that make them particularly vulnerable to EDCs exposure and suitable candidate as a sentinel species. Vitellogenin (Vtg) is a yolk pre-cursor protein synthesized by the liver of non-mammalian vertebrates and induced in response to estrogen. Purified plasma Vtg from caimans injected with estradiol-17beta (E2) was used to generate a polyclonal anti

Recognition of the major histocompatibility complex restriction element modulates CD8(+) T cell specificity and compensates for loss of T cell receptor contacts with the specific peptide.

Triggering of a T cell requires interaction between its specific receptor (TCR) and a peptide antigen presented by a self-major histocompatibility complex (MHC) molecule. TCR recognition of self-MHC by itself falls below the threshold of detection in most systems due to low affinity. To study this interaction, we have used a read-out system in which antigen-specific effector T cells are confronted with targets expressing high levels of MHC compared with the selecting and priming environment. More specifically, the system is based on CD8(+) T cells selected in an environment with subnormal leve

A highly sensitive and specific radioimmunoassay for quantitation of plasma fluphenazine.

Antisera of high sensitivity and selectivity were obtained from rabbits immunized with conjugates of hemisuccinylated fluphenazine and porcine thyroglobulin. The antiserum selected (titer 1:6000) for the development of the RIA was obtained after a priming dose and a single iv booster injection three months later. This antiserum had negligible crossreactivity with known available metabolites of fluphenazine (FPZ) and an affinity constant of 2 X 10(10) L/mol. Tritiated FPZ was further purified by HPLC and used as a ligand. The method detects as little as 20 pg/mL of plasma (4 pg/RIA tube) after

Neurovascular and synaptic milieu of brain-resident cells in cognitive dysfunction of myalgic encephalomyelitis/chronic fatigue syndrome.

BACKGROUND: Cognitive dysfunction (“brain fog”), encompassing impairments in attention, processing speed, memory, and executive function, is a prevalent and disabling feature of myalgic encephalomyelitis/chronic fatigue syndrome (ME/CFS). While systemic immune abnormalities are well documented, the central nervous system processes linking peripheral immune disturbance to cognitive impairment remain poorly defined, limiting mechanism-based stratification and therapeutic development. MAIN BODY: This review synthesizes evidence from neuroimaging, neuroimmunology, neurovascular biology, and cellul

A novel FcγRIIa-TRIM54-STAT2 axis negatively regulates type I interferon signaling and promotes viral susceptibility in human monocytes.

UNLABELLED: While adaptive humoral immunity and type I interferon (IFN-I) responses are known to cross-regulate each other during viral infection, the mechanisms by which antibodies (Abs) or immune complexes (ICs) modulate IFN-I signaling in immune cells remain unclear. Here, using plate-coated human IgG (cIgG) as an antigen-independent IC mimic, we show that FcγRIIa cross-linking in human blood monocytes induces the expression of TRIM54, an E3 ubiquitin ligase previously thought to be constitutively expressed only in muscle tissue. Mechanistically, TRIM54 binds to and promotes the ubiquitinat

Systemic and Mucosal Immune Responses Induced by Adenoviral-Vectored Consensus H5 Influenza A Vaccines in Mice and Swine.

BACKGROUND/OBJECTIVES: The continued evolution and cross-species transmission of clade 2.3.4.4b H5Nx highly pathogenic avian influenza (HPAI) viruses underscores the need for broadly protective vaccines in swine, a key intermediary host. This study aimed to evaluate systemic and mucosal immune responses elicited by adenoviral-vectored (Ad) vaccines encoding a centralized consensus hemagglutinin antigen (H5CC) in mice and swine. METHODS: We constructed H5CC-based vaccines that were delivered using replication-defective (Ad5 and Ad6) and replication-competent (Ad28 and Ad48) human adenoviral ve

Avian influenza virus cross-infections as test case for pandemic preparedness: From epidemiological hazard models to sequence-based early viral warning systems.

Pandemic preparedness starts with an early warning system of viruses with a pandemic potential. Based on information collected in a multitude of surveys, hazard models were developed identifying influenza viruses presenting a pandemic threat. Scores are attributed for 10 viral traits by expert panels which identified avian influenza viruses (AIV) belonging to subtypes H7N9 and H5N1 as representing the greatest pandemic risk. In 2013, more than 100 human cases infected with AIV H7N9 were observed in China. Case fatality rate (CFR) was high (27%), but the human-to-human transmission rate was low

Heterologous prime-boost immunisation with mRNA- and AdC68-based 2019-nCoV variant vaccines induces broad-spectrum immune responses in mice.

The ongoing evolution of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2 or 2019-nCoV) variants has been associated with the transmission and pathogenicity of COVID-19. Therefore, exploring the optimal immunisation strategy to improve the broad-spectrum cross-protection ability of COVID-19 vaccines is of great significance. Herein, we assessed different heterologous prime-boost strategies with chimpanzee adenovirus vector-based COVID-19 vaccines plus Wuhan-Hu-1 (WH-1) strain (AdW) and Beta variant (AdB) and mRNA-based COVID-19 vaccines plus WH-1 strain (ARW) and Omicron (B.1.1.529)
