Antigen-presenting T cells. II. Clonal responses of alloreactive and virus-specific self-restricted human cytotoxic T cell responses stimulated by T lymphoblasts.

The capacity of various stimulator cell types to present alloantigens or viral antigens to resting human CD8+ cytotoxic lymphocyte precursors (CLP) was analyzed in a limiting dilution culture system. Cell sorter-separated T lymphoblasts of both CD4+ and CD8+ phenotypes but not resting T cells were found to efficiently stimulate the clonal development of allogeneic CD8+ CLP. Thus, 5000 CD4+ T lymphoblasts activated as many (one out of 200 to one out of 300) allogeneic CLP as 50,000 peripheral blood mononuclear stimulator cells. This potent stimulator activity was found in CD4+ and CD8+ T lympho

Differentiation-dependent sensitivity of human B-cell-derived lines to major histocompatibility complex-restricted T-cell cytotoxicity.

Sets of Burkitt lymphoma lines and Epstein-Barr virus (EBV)-transformed lymphoblastoid cell lines (LCLs) derived from the same individuals were compared for sensitivity to cytotoxic T-lymphocyte (CTL) clones. Major histocompatibility complex class I antigen-restricted CTL clones were generated by stimulating the lymphocytes of an EBV-seropositive individual with the autologous LCL. One clone (BK-20) lysed the autologous and allogeneic HLA-A11-expressing LCLs but not mitogen-induced B lymphoblasts. Thus the clone was selectively cytotoxic for LCLs. Allospecific CTL clones directed against the H

Subacute sclerosing panencephalitis after drug-induced immunosuppression.

A girl developed subacute sclerosing panencephalitis (SSPE). Eight years earlier she had had measles infection contracted shortly after cytotoxic treatment and radiotherapy for a spinal neuroblastoma. The case illustrates that typical SSPE, like immunosuppressive measles encephalopathy, can arise after drug-induced immunosuppression, and supports the view that these diseases probably represent opposite ends of a spectrum induced by measles virus infection in an individual with some form of immunological deficiency.

[Development in the immunotherapy of malignant melanoma].

Recent progress in the investigation of malignant melanoma has encouraged the possibilities of using immunological methods in the treatment of cancer. Under investigation are especially attempts to influence both the humoral and cellular immunity with cytotoxic effect on tumor cells. In perspective of these studies schedules for greater effectiveness of immunotherapy by combination of active, passive, eliminative or adoptive methods are conceivable. They provide a rational basis particulary because essential questions of alternation between the immune reactions are not yet understood.

Impaired nonrestricted cytolytic activity in systemic lupus erythematosus: involvement of a pathway independent of Fas, tumor necrosis factor, and extracellular ATP that is associated with little detectable perforin.

OBJECTIVE: To determine the cytolytic effector pathway involved in impaired generation of nonrestricted cytolytic activity in systemic lupus erythematosus (SLE). METHODS: Peripheral blood mononuclear cells (PBMC) from normal subjects and SLE patients were stimulated in vitro with anti-CD3 monoclonal antibody (MAb) and interleukin-2 to promote the generation of nonrestricted cytolytic activity. On day 13 of culture, the PBMC were assayed for cytolytic activity against Fas-Daudi cells and Fas+ Jurkat cells. The effects on cytotoxicity of calcium chelation, antagonist anti-Fas MAb, tumor necrosi

Mineral dust exposure and systemic diseases.

Based on clinical and immunological studies, we have proposed the hypothesis that occupational dust exposure might cause not only pneumoconiosis but also autoimmune diseases and malignancies of various organs such as neoplasms of lymphatic and hematopoietic tissues and gastric cancer. Evidence from cohort studies of pneumoconiotic patients in Japan, copper miners, and stone masons support our hypothesis. The carcinogenicity and cytotoxic effect of inhaled dust on immune cells are considered to contribute to the development of these diseases.

Isolated bilateral T-cell renal lymphoblastic lymphoma.

A 3-year-old girl presented with bilateral renal masses. The histologic diagnosis was lymphoblastic lymphoma. Cell surface markers showed a cytotoxic/suppressor T cell phenotype. The patient was treated with APO therapy. She has remained continuously disease-free for 30 months and has been off all therapy for 6 months. This rare case supports the value of precise classification (by histologic and immunologic methods) as a guide to appropriate tumor therapy.

Altered immunity in migraine: a comprehensive scoping review.

BACKGROUND: The pathogenesis of migraine remains unclear; however, a large body of evidence supports the hypothesis that immunological mechanisms play a key role. Therefore, we aimed to review current studies on altered immunity in individuals with migraine during and outside attacks. METHODS: We searched the PubMed database to investigate immunological changes in patients with migraine. We then added other relevant articles on altered immunity in migraine to our search. RESULTS: Database screening identified 1,102 articles, of which 41 were selected. We added another 104 relevant articles.

Analysis of murine CAR-T cell-mediated cytotoxicity in solid tumor models.

Chimeric Antigen Receptor T-cell (CAR-T) based immunotherapy represents a ground-breaking advancement in the treatment of certain haematological malignancies, such as acute lymphoblastic leukemia (ALL) and multiple myeloma (MM), often leading to remarkable remission rates. Despite these successes, many patients relapse within the first-year post-treatment, highlighting a critical limitation of these therapies. Furthermore, translating the success of CAR-T therapies to solid tumors has been challenging, with limited clinical efficacy, highlighting the need for innovative designs and strategies

Impact of chemo-immunotherapy on tumour-immune interactions: A non-autonomous Model of Tumour Necrosis Factor and T cell dynamics.

This study explores the interaction between cancer cells, helper T cells, cytotoxic T cells, and tumour necrosis factors in chemotherapy and immunotherapy treatment microenvironment. The goal is to analyse the connection of helper and cytotoxic T-cell levels with the anti-tumour immune response and the impact of various dosing regimens when combined with immunotherapy and chemotherapy. We discussed the positivity and boundedness of the model. Further, we analyse the biologically valid equilibria and investigate their local stability properties, examining transcritical, saddle-node, Hopf, and B

Pomalidomide enhances CD8+ T and NK cell mediated killing of HIV-infected cells.

BACKGROUND: Overcoming persisting immune dysfunction in people living with HIV (PLHIV) on suppressive antiretroviral therapy (ART) is a key challenge to curing HIV. Agents that reverse immune dysfunction may enhance viral immunity to support the immunological control and clearance of persisting HIV as part of curative strategies. Here, we investigated pomalidomide, a well-tolerated immunomodulatory drug, as an agent to enhance HIV-directed immune responses. METHODS: We collected peripheral blood mononuclear cells (PBMC) from PLHIV on ART, cultured cells in the presence of HIV peptides and pom

Mechanism of tumor cells escaping from immune surveillance of NK cells.

Natural killer (NK) cells play an important role in anti-tumor and anti-infection, and perform their immune surveillance function in various ways. However, no matter what kind of cancer, the functional activity of NK cells in the tumor microenvironment (TME) is suppressed. Understanding the relationship between tumor cells and NK cells is very critical for tumor immunotherapy. This review discusses the mechanism of tumor cells escaping the immune surveillance of NK cells. These include a variety of factors that inhibit the activity of NK cells, an imbalance of activating receptors and inhibiti

Altered Cytotoxicity Profile of CD8+ T Cells in Ankylosing Spondylitis.

OBJECTIVE: Ankylosing spondylitis (AS) is an inflammatory arthritis in which men have a higher risk of developing progressive axial disease than women. Transcriptomic studies have shown reduced expression of cytotoxic cell genes in the blood of AS patients. HLA-B27 contributes the greatest risk for AS, suggesting a role for CD8+ T cells. This study was undertaken to profile AS patient cytotoxic cells with the hypothesis that an alteration in CD8+ T cells might explain the aberrant cytotoxic profile observed in patients. METHODS: Whole blood was examined for GZM and PRF1 gene expression by qua

New approaches to HIV vaccine development.

Development of a safe and effective vaccine for HIV is a major global priority. However, to date, efforts to design an HIV vaccine with methods used for development of other successful viral vaccines have not succeeded due to HIV diversity, HIV integration into the host genome, and ability of HIV to consistently evade anti-viral immune responses. Recent success in isolation of potent broadly neutralizing antibodies (bnAbs), in discovery of mechanisms of bnAb induction, and in discovery of atypical mechanisms of CD8T cell killing of HIV-infected cells, have opened new avenues for strategies for

Broad CTL Response in Early HIV Infection Drives Multiple Concurrent CTL Escapes.

Recent studies have highlighted the ability of HIV to escape from cytotoxic T lymphocyte (CTL) responses that concurrently target multiple viral epitopes. Yet, the viral dynamics involved in such escape are incompletely understood. Previous analyses have made several strong assumptions regarding HIV escape from CTL responses such as independent or non-concurrent escape from individual CTL responses. Using experimental data from evolution of HIV half genomes in four patients we observe concurrent viral escape from multiple CTL responses during early infection (first 100 days of infection), prov

Bystander immune cytolysis.

In addition to alloimmune and autoimmune cell lysis, a third category of immune destruction of blood cells should be recognized. This additional immunologic response occurs when cells or tissues are injured by immunologic reactions in which the cells act as "innocent bystanders." One mechanism by which an immune response to an exogenous antigen leads to the destruction of autologous blood cells is the temporary development of autoantibodies. This is actually an alloimmune reaction which results in a temporary state of "pseudo"-autoimmunity. Although originally described as a type of hemolysis

Irregular cytokine pattern of CD4+ T lymphocytes in response to Staphylococcus aureus in patients with Wegener's granulomatosis.

The initial stage of Wegener's Granulomatosis (WG) is often marked by symptoms of infection and it has been postulated that a bacterial infection could be the aetiologic factor of this disease. The objective of our work was to investigate T-cell-mediated immunity in WG by testing proliferative responses on bacterial antigens and particularly Staphylococcus aureus. We investigated the bulk proliferative response of peripheral blood lymphocytes (PBL) from patients with clinically active WG to gram-positive bacteria and purified proteinase 3 (PR-3), the major target antigen of c-ANCA. We generate

Interleukin-2 restores natural killer activity inhibited by sera from HIV+ hemophilic patients.

Natural killer (NK) activity is impaired in patients with positive serology for the human immunodeficiency virus (HIV). We previously found an inhibitory effect of sera from hemophilic (He) HIV+ patients on normal NK activity. In the present study, we have further characterized this effect by studying its reversibility, temperature and time incubation dependence. Since interleukin 2 (IL-2) is able to enhance NK levels, we analyzed the capacity of this lymphokine to reverse the effect of He HIV+ sera. We found that when IL-2 activation of NK activity occurred simultaneously or after HIV+ serum-

[Immunodepressive effect of measles and parotitis virus in vitro].

Mechanisms of suppressive effect of measles and parotitis virus on lymphocyte proliferation and cytolytic activity of natural killers are studied in vitro. Both viruses exert a weak mitogenic effect on human blood lymphocytes and at the same time suppress their response to phytohemagglutinin stimulation in vitro. Suppressive effect of measles virus is more expressed than of parotitis virus. Moreover, measles virus suppresses lysis of human blood killer target cells. The suppressive effect manifests as early as in 1 h and increases after 3 h of natural killer cell contact with measles virus.
