Investigation of Echinococcus multilocularis in foxes and dogs in Pakistan by detection of copro-DNA.

Alveolar echinococcosis (AE) is a zoonosis caused by Echinococcus multilocularis, a heteroxenous parasite belonging to Cestoda class. AE is currently considered an important public health issue, but epidemiological and notably molecular data from several endemic countries, including Pakistan, are sparse. Here we report the first detection of Echinococcus multilocularis in wildlife from Pakistan after real-time PCR and sequencing confirmation in the faecal samples of three foxes from northern Kaghan and Siran regions. The occurrence is estimated at 4.4% (95% CI 0.9-12.4). In order to go further

Characterization of a non-sexual population of Strongyloides stercoralis with hybrid 18S rDNA haplotypes in Guangxi, Southern China.

Strongyloidiasis is a much-neglected but sometimes fatal soil born helminthiasis. The causing agent, the small intestinal parasitic nematode Strongyloides stercoralis can reproduce sexually through the indirect/heterogonic life cycle, or asexually through the auto-infective or the direct/homogonic life cycles. Usually, among the progeny of the parasitic females both, parthenogenetic parasitic (females only) and sexual free-living (females and males) individuals, are present simultaneously. We isolated S. stercoralis from people living in a village with a high incidence of parasitic helminths,

Molecular phylogenetic position of Haplometroides intercaecalis (Digenea, Plagiorchiidae).

Three valid species of Haplometroides Odhner, 1910 parasitise snakes and amphisbaenians from South America. This study provides additional data on morphometric and molecular phylogenetic position inferred from the nuclear ribosomal gene 28S (partial). DNA sequences were isolated from Haplometroides intercaecalis Silva, Ferreira and Strüssmann, 2007 found in one specimen of Phalotris matogrossensis Lema, D'Agostini and Cappellari, 2005. Five digenean specimens were recovered from the esophagus of this snake, and four specimens were used for morphometrical studies and one specimen for molecular

A new species of Aphanoblastella Kritsky, Mendoza-Franco and Scholz, 2000 (Monogenea, Dactylogyridae) parasitic on heptapterid catfish (Siluriformes) in the Neotropical region.

Aphanoblastella magna n. sp. is described from the gills of the heptapterid catfish Pimelodella avanhandavae Eigenmann, 1917 in southeastern Brazil (Paraná River Basin). This new species most closely resembles Aphanoblastella chagresii Mendoza-Franco, Aguirre-Macedo and Vidal-Martínez, 2007 described from a congeneric fish host in Panama, but can be distinguished by the shape of the accessory piece and shape and size of the ventral and dorsal bars. From the other species of the genus, A. magna n. sp. differs mainly in the shape of the male copulatory organ (MCO), which is sinuous, versus spira

Genetic diversity and phylogeography of the elusive, but epidemiologically important Echinococcus granulosus sensu stricto genotype G3.

Cystic echinococcosis (CE) is a severe parasitic disease caused by the species complex Echinococcus granulosus sensu lato. Human infections are most commonly associated with E. granulosus sensu stricto (s.s.), comprising genotypes G1 and G3. The objective of the current study was to provide first insight into the genetic diversity and phylogeography of genotype G3. Despite the epidemiological importance of the genotype, it has remained poorly explored due to the ambiguity in the definition of the genotype. However, it was recently demonstrated that long sequences of mitochondrial DNA (mtDNA) p

The complete mitochondrial genome of Orthocoelium streptocoelium (Digenea: Paramphistomidae) for comparison with other digeneans.

Orthocoelium streptocoelium is a common paramphistome species parasitizing the rumen and/or reticulum of small ruminants, leading to significant losses. This study first determined the complete mitochondrial (mt) genome of O. streptocoelium. The complete mt genome of O. streptocoelium was amplified, sequenced, assembled, analysed and then compared with those of other digeneans. The entire mt genome of O. streptocoelium is 13,800 bp in length, which is smaller than those of other digeneans except for Opisthorchis viverrini. This mt genome contains 12 protein-coding genes, 22 transfer RNA genes,

Strong-LAMP: A LAMP Assay for Strongyloides spp. Detection in Stool and Urine Samples. Towards the Diagnosis of Human Strongyloidiasis Starting from a Rodent Model.

BACKGROUND: Strongyloides stercoralis, the chief causative agent of human strongyloidiasis, is a nematode globally distributed but mainly endemic in tropical and subtropical regions. Chronic infection is often clinically asymptomatic but it can result in severe hyperinfection syndrome or disseminated strongyloidiasis in immunocompromised patients. There is a great diversity of techniques used in diagnosing the disease, but definitive diagnosis is accomplished by parasitological examination of stool samples for morphological identification of parasite. Until now, no molecular method has been te

Geographic Range Expansion for Rat Lungworm in North America.

Using quantitative PCR analysis and DNA sequencing, we provide evidence for the presence of rat lungworm (Angiostrongylus cantonensis) in Oklahoma, USA, and identified a potentially novel rat host (Sigmodon hispidus). Our results indicate a geographic range expansion for this medically and ecologically relevant parasite in North America.

Cystic echinococcosis in a fox-hound hunt worker, UK.

A male resident in Vanuatu with prior history of employment as a hunt kennel-man in England (1980-2001) was surgically treated for the removal of a hydatid cyst subsequently confirmed as Echinococcus granulosus (G1 genotype). This is the first human molecularly identified CE case reported from the UK and a first in a fox-hound handler and indicates a general neglected occupational risk.

The pig tapeworm Taenia solium, the cause of cysticercosis: Biogeographic (temporal and spacial) origins in Madagascar.

Cysticercosis is a serious public health problem in Madagascar. The prevalence rate of active cysticercosis reached 21% in regions with a high level of livestock farming. Taenia solium of African-American and Asian genotypes are both present on the island. The times of divergence of the 13 specimens studied suggests a very ancient diversification of T. solium. These events are widely thought to be prior to the domestication of pigs, and seem to follow the expansion of Homo in Asia. Multiple human migrations and the diversity of potential intermediate hosts may have led to a complex epidemiolog

Genetic analysis of Echinococcus multilocularis originating from a patient with alveolar echinococcosis occurring in Minnesota in 1977.

To date, only a single proven case of autochthonous human alveolar echinococcosis has been recorded in Minnesota in 1977. At that time, echinococcal lesions removed from the patient were experimentally inoculated into voles, and the parasite materials obtained from the voles were preserved as histopathologic specimens for 30 years. In this study, retrospective genetic analysis of larval Echinococcus multilocularis originating in the human case was performed using the histopathologic specimens. DNA was extracted from the hematoxylin and eosin-stained specimens, and mitochondrial cytochrome c ox

Echinococcus shiquicus n. sp., a taeniid cestode from Tibetan fox and plateau pika in China.

The taeniid cestode Echinococcus shiquicus n. sp. was found from the Tibetan fox Vulpes ferrilata and the plateau pika Ochotona curzoniae in the Qinghai-Tibet plateau region of China. In the adult stage, E. shiquicus from the foxes is morphologically similar to Echinococcus multilocularis. However, the new species is differentiated by its smaller rostellar hooks, fewer segments, distinct position of genital pore in the mature segment and fewer eggs in the gravid segment. Hydatid cysts of E. shiquicus found in the livers from the pikas were essentially unilocular but an oligovesicular cyst was

DNA differential diagnosis of taeniasis and cysticercosis by multiplex PCR.

Multiplex PCR was established for differential diagnosis of taeniasis and cysticercosis, including their causative agents. For identification of the parasites, multiplex PCR with cytochrome c oxidase subunit 1 gene yielded evident differential products unique for Taenia saginata and Taenia asiatica and for American/African and Asian genotypes of Taenia solium with molecular sizes of 827, 269, 720, and 984 bp, respectively. In the PCR-based detection of tapeworm carriers using fecal samples, the diagnostic markers were detected from 7 of 14 and 4 of 9 T. solium carriers from Guatemala and Indon

A multiplex PCR for unequivocal differentiation of all encapsulated and non-encapsulated genotypes of Trichinella.

We have developed a single PCR test for the simple and unequivocal differentiation of all currently recognised genotypes of Trichilnella. Partial DNA sequence data were generated from internal transcribed spacers ITS1 and ITS2, and from the expansion segment V region of the ribosomal DNA repeat from five species of Trichinella and two additional genotypes, designated T5 and T6. Five different PCR primer sets were identified which, when used simultaneously in a multiplex PCR, produce a unique electrophoretic DNA banding pattern for each species and genotype including three distinct genotypes of

Evaluation of molecular methods for the field study of the natural history of Dicrocoelium dendriticum.

There is a need for improved methods for the study of the impacts of climatic and livestock management change on the epidemiology of production-limiting helminth parasitic diseases. In this study we report the application of molecular methods to describe the natural history of the small lancet fluke, Dicrocoelium dendriticum on Machair pastures on the Inner Hebridean Isle of Coll. Our results build upon those of the only previous historic field study of D. dendriticum in the British Isles that had been undertaken on the same study site. We demonstrate the value of combining conventional parasi

Molecular Detection of Schistosome Infections with a Disposable Microfluidic Cassette.

Parasitic helminths such as schistosomes, as well as filarial and soil-transmitted nematodes, are estimated to infect at least a billion people worldwide, with devastating impacts on human health and economic development. Diagnosis and monitoring of infection dynamics and efficacy of treatment depend almost entirely on methods that are inaccurate, labor-intensive, and unreliable. These shortcomings are amplified and take on added significance in mass drug administration programs, where measures of effectiveness depend on accurate monitoring of treatment success (or failure), changes in disease

Efficacy of mebendazole in the spontaneous NZBxNZWF1 animal model of systemic lupus erythematosus.

Systemic lupus erythematosus (SLE) is a chronic autoimmune disease with a complex etiology involving both innate and adaptive immune dysregulation. Among several perturbed signaling pathways, decreased ERK activity in CD4⁺ T-cells has been linked to DNA hypomethylation and aberrant gene expression in SLE. Mebendazole (MBZ), an anti-helminthic drug with a well-established safety profile, has shown immunomodulatory effects in preclinical studies, including activation of the MEK/ERK pathway and inhibition of MAPK14 (p38), a known driver of inflammation. To evaluate the therapeutic potential of MB

'First record of Hsiungia pekingensis (Nematoda: Strongylidae) in North America: Morphological and molecular identification of a rare equine strongyle.

Equids are infected by a diversity of gastrointestinal nematode parasites, including 64 species of equine strongyle nematodes from19 genera. Despite numerous surveys of horse strongyles worldwide, certain geographic regions and rare species remain understudied. In 1964, a new species of equine strongyle, Cylicocyclus pekingensis, was described from a donkey in China. Subsequently, this species was recorded in horses from Kazakhstan and reclassified as Hsiungia pekingensis (K'ung and Yang, 1964), the only species in this genus. Since then, H. pekingensis has not been reported elsewhere, with li

Occurrence and host associations of Dollfustrema bagarii (Digenea: Bucephalidae) in channel catfish cage culture in Northern Vietnam.

A survey was conducted across 11 provinces in Northern Vietnam to investigate the distribution of Dollfustrema bagarii metacercariae - a causative agent of significant losses in cultured channel catfish (Ictalurus punctatus) - and to identify its first intermediate host. A total of 337 fish specimens were collected from 103 farms. Metacercariae of D. bagarii were detected in 72 farms (69.9% of farms), and 44.51% of the sampled fish were positive. The prevalence in lowland provinces was 3.14 times higher than that in mountainous regions (p < 0.001), while no significant difference was observed

PHYLOGENETIC POSITION OF NEOGOGATEA (DIPLOSTOMOIDEA; CYATHOCOTYLIDAE).

Neogogatea Chandler and Rausch, 1947, is a small genus of cyathocotylid digeneans parasitic in the intestines of birds of prey. The genus contains only 3 species: 2 from the Nearctic and 1 from the Palearctic. No sequence data from mature, properly identified representatives of Neogogatea were available until now. In the present study, we collected mature adult Neogogatea kentuckiensis (Cable, 1935) Hoffman and Dunbar, 1963, from osprey in Georgia, Florida, and Delaware, and sequenced fragments of the nuclear large ribosomal subunit (28S) gene and cytochrome c oxidase subunit I (COI) mtDNA gen
