The breakage of a drug-eluting stent delivery system leading to limb amputation.

A 68-year-old female with total occlusion of the bare metal stent in the left superficial femoral artery was treated by placement of three drug-eluting stents (DESs). Acute thrombosis developed on the next day. Thrombosis in the left popliteal artery to anterior tibial artery was found. An additional stent was placed in the popliteal artery. Rethrombosis was observed. The left leg could not be salvaged. Examination of the amputated limb revealed broken DES delivery system below the knee vessel. This was an unusual case of breakage of a DES delivery system that led to major amputation.

Orally dissolving strips: A new approach to oral drug delivery system.

Recently, fast dissolving films are gaining interest as an alternative of fast dissolving tablets. The films are designed to dissolve upon contact with a wet surface, such as the tongue, within a few seconds, meaning the consumer can take the product without need for additional liquid. This convenience provides both a marketing advantage and increased patient compliance. As the drug is directly absorbed into systemic circulation, degradation in gastrointestinal tract and first pass effect can be avoided. These points make this formulation most popular and acceptable among pediatric and geriatr

Nasal drug administration: potential for targeted central nervous system delivery.

Nasal administration as a means of delivering therapeutic agents preferentially to the brain has gained significant recent interest. While some substrates appear to be delivered directly to the brain via this route, the mechanisms governing overall brain uptake and exposure remain unclear. Some substrates utilize the olfactory nerve tract and gain direct access to the brain, thus bypassing the blood-brain barrier (BBB). However, most agents of pharmacologic interest likely gain access to the brain via the olfactory epithelium, which represents a more direct route of uptake. While the tradition

Electrophysiologic characteristics of a pulsed iontophoretic drug-delivery system in coronary arteries.

This study evaluated the electrophysiologic effects of a pulsed iontophoretic drug-delivery system when used in the coronary arteries. Prevention of acute thrombosis and restenosis after intravascular procedures may be enhanced by high concentrations of therapeutic agents within the vessel wall. A new intravascular drug-delivery system uses iontophoresis to maximize local tissue concentrations of drug. However, the electrophysiologic effects of such a system in coronary arteries are unknown. An iontophoretic membrane balloon-tipped catheter was placed fluoroscopically in the mid left anterior

RBCs as a Bioinspired Drug Delivery System for Co-delivery of Irinotecan and Nanoalumina Enhances Colorectal Cancer Therapy.

Colorectal cancer (CRC) is a malignant epithelial tumor with high morbidity and mortality. In CRC treatment, irinotecan (CPT-11) as a chemotherapeutic drug is widely applied. However, its half-life is short, leading to large dosages and severe side effects. Red blood cells (RBCs) are biocompatible drug carriers with high capacity, avoiding premature drug degradation and achieving slow drug release. Nanoalumina (AN) is an emerging immune adjuvant that can enhance the immune response. Here, we used RBCs as carriers and absorbed AN to construct AN-CPT-11-RBCs. CPT-11 would induce tumor cell death

Silk sericin/fibroin composite drug delivery systems for lung cancer: mechanistic Rationale, potential functional complementarity, and future perspectives.

OBJECTIVES: Silk fibroin (SF) and silk sericin (SS) are promising biomaterials for lung cancer drug delivery because of their biocompatibility, biodegradability, and versatile processing. SF-based systems can provide structural stability and controlled or prolonged release, although release behaviour depends on drug properties, loading strategy, secondary structure, and formulation architecture. Engineered SS-containing systems may contribute hydrophilicity, formulation-dependent environmental responsiveness, and biologically relevant interactions, but SS-rich formulations can show limited str

SAHA-coupled biocompatible cellulose-based nanoparticles as HDAC-inhibitory drug delivery systems with slow and sustained release behavior.

Nanoparticles (NPs) are being increasingly investigated as drug delivery systems to overcome common challenges in drug therapy, such as rapid drug metabolism and short serum half-life. In this study, a facile method was applied to combine the incorporation of a physiologically cleavable linkage with the introduction of a strained alkyne enabling efficient attachment of SAHA, a histone deacetylase inhibitor, to various structures. SAHA was chemically attached to a cellulose backbone hydrophobized by valproate groups, allowing the formulation of NPs by the emulsion-evaporation technique, resulti…
