Personality traits and risky behavior among motorcyclists: An exploratory study.

BACKGROUND: Personality traits have been associated with a series of dysfunctional behaviors, ranging from violence to drug abuse and other risky behaviors. However, few studies have investigated motorcycle riders' personality traits, and no research using the psychobiological model of personality was found. Thus, we investigated the association between temperament and character traits and the occurrence of Motorcycle Accidents (MAs). METHODS: This cross-sectional study was conducted with a randomly selected of 153 Brazilian motorcycle riders (116 male and 37 female) with a mean age of 31.8ra

The impact of voluntary wheel-running exercise on hippocampal neurogenesis and behaviours in response to nicotine cessation in rats.

RATIONALE: The literature indicates that nicotine exposure or its discontinuation impair adult hippocampal neurogenesis in rats, though the impact of exercise on this process remains unclear. We have previously shown that disturbances in the number of doublecortin (DCX, a marker of immature neurons)-positive (DCX+) cells in the dentate gyrus (DG) of the hippocampus during nicotine deprivation may contribute to a depression-like state in rats. OBJECTIVES: This study aimed to investigate the effect of running on hippocampal neurogenesis, depression-like symptoms, and drug-seeking behaviour duri

The selective anandamide transport inhibitor VDM11 attenuates reinstatement of nicotine seeking behaviour, but does not affect nicotine intake.

BACKGROUND AND PURPOSE: The endocannabinoid system appears to play a pivotal role in mediating the rewarding and reinforcing effects of nicotine. Recent studies have shown that the inhibition of fatty acid amide hydrolase (FAAH) attenuates reinstatement of nicotine-seeking induced by nicotine priming and nicotine-associated cues. FAAH hydrolyses the endogenous endocannabinoid anandamide, as well as other non-cannabinoid ligands such as oleoylethanolamide (OEA) and palmitoylethanolamide (PEA). As OEA and PEA can attenuate both nicotine-taking and nicotine-seeking behaviour, the specific role of

Rewarding Social Interaction in Rats Increases CaMKII in the Nucleus Accumbens.

Calcium/calmodulin-dependent protein kinase II (CaMKII) is known to be involved in the sensitized locomotor responses and drug-seeking behavior to psychostimulants. However, little is known about the contribution of CaMKII signaling in the nucleus accumbens (NAc) in natural rewards such as social interaction. The present experiments explored the implication of CaMKII signaling in drug versus natural reward. In the NAc of rats expressing cocaine or social interaction conditioned place preference (CPP), αCaMKII activation was induced in those expressing social interaction but not cocaine CPP. In…

[Research advances in neuroinflammatory mechanisms of substance addiction].

Substance addiction is a chronic psychiatric disorder characterized by compulsive substance use, impaired behavioral control, and high relapse rates. Although mesolimbic dopaminergic reward system dysfunction is a key neurobiological substrate of substance addiction, it cannot fully explain persistent drug craving, addiction-related memory, or compulsive drug-seeking and drug-taking behaviors after abstinence. Repeated exposure to addictive substances can induce oxidative stress, activation of pattern-recognition receptors, increased blood-brain barrier permeability, and gut-brain axis dysregu

The role of orexin-1 receptor signaling in demand for the opioid fentanyl.

The orexin system is a potential treatment target for drug addiction. Orexin-1 receptor (OxR1) antagonism reduces demand for cocaine and remifentanil, indicating that orexin-based therapies may reduce demand for many classes of abused drugs. However, pharmacokinetics vary greatly among opioids and it is unclear if OxR1 antagonism would reduce demand for all opioids, particularly ones with high abuse liability. Here, we established a behavioral economics (BE) procedure to assess the effects of OxR1 antagonism on demand for the highly abused opioid fentanyl. We also investigated the utility of o

Finasteride withdrawal induces anxiety-like behavior and novelty avoidance in adult male rats.

Finasteride, an inhibitor of the enzyme 5alpha-reductase, prescribed for benign prostatic hyperplasia and androgenetic alopecia, induces a wide variety of side effects during treatment and upon withdrawal, like sexual dysfunction and cognitive and psychological disorders, inducing the so-called post-finasteride syndrome (PFS). Here, we explored the behavioral effects of this drug in adult male rats after subchronic finasteride treatment (20 days) and at drug discontinuation (1 month). We employed multiple behavioral paradigms, including the open field test and elevated plus maze to assess loco

Melatonin attenuates fentanyl - induced behavioral sensitization and circadian rhythm disorders in mice.

Melatonin is a neurohormone synthesized by the pineal gland to regulate the circadian rhythms and has proven to be effective in treating drug addiction and dependence. However, the effects of melatonin to modulate the drug-seeking behavior of fentanyl and its underlying molecular mechanism is elusive. This study was designed to investigate the effects of melatonin on fentanyl - induced behavioral sensitization and circadian rhythm disorders in mice. The accompanying changes in the expression of Brain and Muscle Arnt-Like (BMAL1), tyrosine hydroxylase (TH), and monoamine oxidase A (MAO-A) in re…

Cocaine-induced Changes in the Expression of NMDA Receptor Subunits.

Cocaine use disorder is manifested by repeated cycles of drug seeking and drug taking. Cocaine exposure causes synaptic transmission in the brain to exhibit persistent changes, which are poorly understood, while the pharmacotherapy of this disease has not been determined. Multiple potential mechanisms have been indicated to be involved in the etiology of cocaine use disorder. The glutamatergic system, especially N-methyl-D-aspartate (NMDA) receptors, may play a role in several physiological processes (synaptic plasticity, learning and memory) and in the pathogenesis of cocaine use disorder. Th

Sex-Specific Role for Egr3 in Nucleus Accumbens D2-Medium Spiny Neurons Following Long-Term Abstinence From Cocaine Self-administration.

BACKGROUND: We previously showed that the transcription factor Egr3 (early growth response 3) is oppositely regulated in nucleus accumbens (NAc) cell subtypes 24 hours following cocaine exposure and bidirectionally mediates cocaine-related behaviors in male rodents. Overexpressing Egr3 in D2 receptor-containing medium spiny neurons (D2-MSNs) before drug exposure reduces the rewarding and psychomotor sensitization effects of cocaine. However, it is unknown if Egr3 plays a role in long-term neuroadaptations in the NAc and relapse to cocaine seeking. METHODS: We measured EGR3 protein levels in t

Diazepam promotes choice of abstinence in cocaine self-administering rats.

When facing a choice between cocaine and a potent, albeit inessential, non-drug alternative (i.e. water sweetened with saccharin), most cocaine self-administering rats abstain from cocaine in favor of the non-drug pursuit, regardless of the dose available and even after extended drug use. Only a minority continues to take the drug despite the opportunity of making a different choice and increasing stakes. This pattern of individual variation could suggest that the majority of rats are resilient to addiction, taking cocaine by default of other options. Only a minority would be vulnerable to add

Psilocybin and Ibogaine in Cocaine-Seeking: Extinction Enhancement Without Relapse Prevention.

Psychedelics have emerged as potential therapeutics for substance use disorders, yet preclinical data validating their efficacy remain limited. Here, we investigated the effects of a clinically inspired dose-escalation protocol of psilocybin and ibogaine on extinction and cue-induced reinstatement in Wistar male rats following intravenous cocaine self-administration (IVSA). Rats were trained on a fixed ratio 1 (FR1) schedule with cocaine dose-escalation during the acquisition phase (0.25 mg/kg/infusion, followed by 0.5 mg/kg/infusion). Following acquisition, animals were randomised into treatm

Assessing vicarious trauma and attitudes toward mental health services for forensic sciences professionals.

Research indicates that forensic science professionals operate under significant pressure, the magnitude of which varies depending on their field, workload, case type, tenure, and the evidentiary significance of their testimony in court. This study conducted a needs analysis of forensic science professionals by examining their psychological well-being, coping mechanisms, and barriers to accessing mental health services. A total of 618 individuals from the AAFS community completed the anonymous online Qualtrics survey. Out of 618 participants, 601 participants responded to the PCL-5 questionnai

Changes in activity across the light-dark cycle during ethanol self-administration, extinction, and ethanol cue-induced reinstatement in rats.

Alcohol use disorder (AUD) patients have a high prevalence of sleep disturbances and report disrupted sleep as a contributor to relapse. Studies are needed to understand the development and progression of sleep problems throughout the AUD cycle. This study tracked activity across the light-dark cycle as a measure of sleep-related endpoints in rats during ethanol self-administration, extinction of self-administration, and ethanol cue-induced reinstatement. Male and female rats implanted with accelerometer-based telemetry devices were induced to drink ethanol using a sucrose-fade procedure. Then

The Role of the Posterior Paraventricular Nucleus of the Thalamus in Food Deprivation-Induced Heroin-Seeking Relapse, in Male and Female Rats.

Abstinence from drug use is often the result of the associated negative consequences. However, relapse occurs in a large proportion of abstinent users, and the underlying brain mechanisms are not clear. An arguably relevant brain area is the posterior paraventricular nucleus of the thalamus (pPVT), which plays a role in motivational processes and addiction-like behaviours. Using a punishment-imposed abstinence procedure, we assessed the effect of chemogenetic inhibition and excitation of the pPVT on food deprivation-induced relapse to heroin-seeking in male and female rats. Rats were trained t

Independent Heritability of Aversive Learning Influences on Cocaine-Seeking and Punishment Resistance in Rats.

Cocaine produces widely recognized rewarding effects but also produces aversive effects that occur several minutes after rewarding effects dissipate. Prior work shows these aversive effects are particularly strong in some animals, in whom they produce conditioned avoidance effects that reduce overall cocaine-seeking. The sources of these individual variations are largely unknown, but we found evidence for contributions from heritable influences. Using outbred male and female Sprague Dawley and heterogeneous stock rats, we found that offspring of individuals expressing higher versus lower level

Decreased incubation of fentanyl seeking in the absence of proximal drug-paired stimuli.

Treating substance use disorders is difficult as individuals often resume substance use during abstinence. One potential factor contributing to the recurrence of substance use is incubation of drug craving. Specifically, individuals report higher levels of craving when presented with drug-paired stimuli across abstinence, although this effect is largely absent in opioid-dependent individuals. In preclinical studies, rodents show increased responding on a previously reinforced manipulandum when presented with drug cues, including for opioids. When proximal cues are not presented, self-reported

mGlu5 function in the nucleus accumbens core during the incubation of methamphetamine craving.

Many studies have demonstrated that negative allosteric modulators (NAM) of metabotropic glutamate receptor 5 (mGlu5) reduce cocaine and methamphetamine seeking in extinction-reinstatement animal models of addiction. Less is known about effects of mGlu5 NAMs in abstinence models, particularly for methamphetamine. We used the incubation of drug craving model, in which cue-induced craving progressively intensifies after withdrawal from drug self-administration, to conduct the first studies of the following aspects of mGlu5 function in the rat nucleus accumbens (NAc) core during abstinence from m

The vagus nerve mediates the suppressing effects of peripherally administered oxytocin on methamphetamine self-administration and seeking in rats.

The neuropeptide oxytocin has emerged as a promising pharmacotherapy for methamphetamine (METH) addiction, and clinical trials of intranasal oxytocin are underway. However, there is debate as to how peripherally administered oxytocin alters brain signalling to modulate addiction processes. Interestingly, there is evidence for functional interactions between peripheral oxytocin administration and the vagus nerve. Therefore, this study investigated whether the effects of peripherally administered oxytocin require vagal signalling to reduce METH self-administration and reinstatement of METH-seeki

H3.3 Barcoding of Nucleus Accumbens Transcriptional Activity Identifies Novel Molecular Cascades Associated with Cocaine Self-administration in Mice.

Although numerous epigenetic modifications have been associated with addiction, little work has explored the turnover of histone variants. Uniquely, the H3.3 variant incorporates stably and preferentially into chromatin independently of DNA replication at active sites of transcription and transcription factor binding. Thus, genomic regions associated with H3.3-containing nucleosomes are particularly likely to be involved in plasticity, such as following repeated cocaine exposure. A recently developed mouse line expressing a neuron-specific hemagglutinin (HA)-tagged H3.3 protein was used to tra
