A Heteroplasmic MT-CO2 m.8024G > A Variant Is Associated with Mitochondrial Bioenergetic Deficiency and Optic Atrophy.

Leber's hereditary optic neuropathy (LHON) is a hereditary neurodegenerative disorder caused by pathogenic mitochondrial DNA (mtDNA) variants. While MT-CO2 defects are implicated in neurodegeneration, their direct association with optic atrophy has not been reported. We identify a heteroplasmic MT-CO2 variant, m.8024G > A (p.Glu147Lys), in a patient with progressive optic atrophy and explore its potential association with mitochondrial dysfunction. A 13-year-old male with progressive unilateral-then-bilateral vision loss underwent comprehensive ophthalmic/neurological evaluation, trio whole-ex

Insights into the life cycles of trematodes infecting pelagic gastropods in Japan.

Pelagic gastropods, including holoplanktonic and neustonic species, have been considered potential sources of trematodes that parasitize marine fishes. However, the taxonomy and life cycles of metacercariae detected in pelagic gastropods remain poorly understood. This study examined metacercaria infections in 10 species of pelagic gastropods (N = 415) collected from Japanese coastal waters between 2016 and 2025. Metacercariae were detected in four pelagic gastropod species and were identified as seven trematode species based on molecular analyses of the 28S ribosomal RNA and cytochrome c oxida

First report of Gyrodactylus teuchis (Monopisthocotyla: Gyrodactylidae) on farmed rainbow trout Oncorhynchus mykiss (Walbaum, 1792) in Russia.

We recorded Gyrodactylus teuchis Lautraite, Blanc, Thiery, Daniel & Vigneulle, 1999 on one-year-old farmed rainbow trout Oncorhynchus mykiss (Salmoniformes, Salmonidae) from Lake Kalmozero (White Sea drainage system, Republic of Karelia, Russia) in May and October 2023. Morphological identification of these specimens was confirmed by molecular data on the mitochondrial cytochrome c oxidase subunit 1 (cox1) gene and the ribosomal internal transcribed spacer 2 (ITS2). Analyses of the newly obtained cox1 gene sequences show they are identical to fragments of sequences of this gene for G. teuchis

A distinct mitochondrial cytochrome c oxidase subunit I lineage of the poultry red mite Dermanyssus gallinae in Tunisian egg-layer flocks.

The poultry red mite, Dermanyssus gallinae is a globally significant ectoparasite affecting layer hens, with substantial economic and animal welfare impacts. Despite its importance, the genetic diversity and population structure of Dermanyssus gallinae in North Africa remain completely uncharacterized. We conducted the first molecular phylogenetic characterization of D. gallinae in Tunisia, analyzing mitochondrial cytochrome c oxidase subunit I (COI) gene sequences from 18 specimens collected across 20 commercial layer farms in four distinct regions (Tabarka, Manouba, Nabeul, and Sfax). Phylog

DNA barcoding reveals cryptic diversity in chiggers (Trombiculidae) parasitizing small mammals across Peninsular Malaysia.

Chiggers (Acariformes: Trombiculidae) are ectoparasites of small mammals and important vectors of scrub typhus, yet their taxonomy and molecular diversity in Southeast Asia remain poorly resolved. This study represents the first DNA barcoding and phylogenetic assessment of chiggers from small mammals across Peninsular Malaysia, integrating mitochondrial (COI) and nuclear (28 S rDNA) genes. A total of 192 chiggers representing 11 morphospecies from six genera were sequenced, with tree topologies inferred using maximum likelihood and supported by multiple species delimitation methods. The COI ge

COA5 has an essential role in the early stage of mitochondrial complex IV assembly.

Pathogenic variants in cytochrome c oxidase assembly factor 5 (COA5), a proposed complex IV (CIV) assembly factor, have been shown to cause clinical mitochondrial disease with two siblings affected by neonatal hypertrophic cardiomyopathy manifesting a rare, homozygous COA5 missense variant (NM_001008215.3: c.157G>C, p.Ala53Pro). The most striking observation in the affected individuals was an isolated impairment in the early stage of mitochondrial CIV assembly. In this study, we report an unrelated family in whom we have identified the same COA5 variant with patient-derived fibroblasts and ske

Use of Deep Amplicon Sequencing Based on the Cytochrome Oxidase I Gene to Quantify the Relative Percentages of Eimeria spp. Oocysts in Poultry Litter.

The purpose of this study was to evaluate a deep amplicon sequencing approach for estimating the relative abundances of different Eimeria spp. oocysts in litter from commercial broiler farms that may or may not be experiencing necrotic enteritis (NE) infections. Oligonucleotide primers directed to the mitochondrial cytochrome oxidase I (COI) gene, a sequence that is conserved among all chicken Eimeria spp., were first used to PCR amplify Eimeria acervulina, Eimeria maxima, and Eimeria tenella oocyst DNA. COI amplification was applied to samples containing either a single Eimeria species or an

Conservation hints for Pinna nobilis from a century-old genetic time capsule.

The noble pen shell, Pinna nobilis, is an iconic marine bivalve endemic to the Mediterranean Sea, playing a key role as an ecosystem engineer. Over the past century, it has faced severe threats from overharvesting, pollution, and catastrophic mass mortality events. This study analysed 119 mitochondrial COI gene sequences from historical (1700s, 1920s, 1970s, 1990s) and modern (2000s) samples, including survivors of recent mass mortality crises. We standardised a protocol to extract DNA from ancient byssus samples over a century old and dated the emergence of the mitochondrial lineages of Pinna

Novel cytochrome c-oxidase deficiency and mitochondrial myopathy in non-syndromic cleft lip with or without palate.

The role of mitochondria in regulating cells during the embryonic phase is crucial, yet their function in causing deformation to orbicularis oris muscle of cleft lip remains controversial. The study was performed to explore morphological abnormalities of orbicularis oris muscle and mitochondria in patients with cleft lip with or without palate. Seventeen cleft lip tissue samples were obtained from consented patients who underwent cleft lip repair. All tissue samples were processed according to the respective analyses: modified Gomori trichrome, cytochrome c-oxidase (COX), adenosine triphosphat

Histopathological and molecular evaluation of hydatid cysts according to necropsy findings of 52 equids in Türkiye.

Echinococcus equinus is a parasitic cestode primarily maintained within an equine-canine life cycle, with horses, donkeys, mules, and other ungulates serving as intermediate hosts. Although E. equinus has historically been considered non-zoonotic, recent molecular studies suggest that this assumption may need to be reevaluated. This study aimed to investigate the presence and molecular characterization of E. equinus in equids from Türkiye. A retrospective analysis of 52 equine necropsies performed between 2020 and 2025 identified hydatid cysts in one Arabian horse and two donkeys. Gross and hi

Hidden in plain sight: discovery of sand flies in Singapore and description of four species new to science.

BACKGROUND: Phlebotomine sand flies (Diptera: Psychodidae) are tiny, blood-sucking insects that are of significant public and veterinary health importance for their role in the transmission of Leishmania parasites, bacteria, and arboviruses. Although sand flies have been documented in most Southeast Asian countries, there are no published records confirming their presence in Singapore. Here, we present this fauna with descriptions of new species. METHODS: Sand fly species identification was confirmed using an integrative taxonomic approach that combines morphological analysis with DNA barcodi

Molecular characterization of Rhipicephalus microplus and Haemaphysalis bispinosa ticks from cattle across Thailand: Regional identification and evidence of different genetic sub-structures between mainland and peninsular populations.

Phylogenetic and population genetic analyses were conducted on tick specimens collected from cattle in northern, northeastern, central, and southern regions of Thailand. Morphological identification indicated these ticks consisted of three species, Rhipicephalus microplus from all four regions, R. sanguineus from the northern and northeastern regions, and a Haemaphysalis species only collected from the northeastern region. Analysis of cytochrome c oxidase subunit I gene (COI) sequences identified R. microplus clades A and C, while clade B was not detected in this study. The same analysis indic

From host to host, and continent to continent: Two phoresy-enabled Guimaraesiella hitchhiker louse species revealed by integrative taxonomy (Phthiraptera: Ischnocera).

The 'core Guimaraesiella' comprise a morphologically rather homogeneous group of avian chewing lice (Phthiraptera), most of which remain undescribed. Based on an integrative approach combining morphological characters and analyses of COI barcoding sequences, we here describe two new species within this group: Guimaraesiella impiger new species and Guimaraesiella stellana new species. Both species were collected from hippoboscid flies in Singapore, suggesting that they are capable of moving phoretically between hosts. In at least G. impiger, this was confirmed as louse specimens from another 30

Chalkophore-mediated respiratory oxidase flexibility controls M. tuberculosis virulence.

Oxidative phosphorylation has emerged as a critical therapeutic vulnerability of M. tuberculosis (Mtb). However, it is unknown how intracellular bacterial pathogens such as Mtb maintain respiration during infection despite the chemical effectors of host immunity. Mtb synthesizes diisonitrile lipopeptides that tightly chelate copper, but the role of these chalkophores in host-pathogen interactions is also unknown. We demonstrate that M. tuberculosis chalkophores maintain the function of the heme-copper bcc:aa3 respiratory supercomplex under copper limitation. Chalkophore deficiency impairs Mtb

Inferotemporal face patches are histo-architectonically distinct.

An interconnected group of cortical regions distributed across the primate inferotemporal cortex forms a network critical for face perception. Understanding the microarchitecture of this face network can refine mechanistic accounts of how individual areas function and interact to support visual perception. To address this, we acquire a unique dataset in macaque monkeys combining fMRI to localize face patches in vivo and then ex vivo histology to resolve their histo-architecture across cortical depths in the same individuals. Our findings reveal that face patches differ based on cytochrome oxid
