Immune escape pathways from the HBV core18-27 CD8 T cell response are driven by individual HLA class I alleles.

BACKGROUND AND AIMS: There is growing interest in T cell-based immune therapies for a functional cure of chronic HBV infection including check-point inhibition, T cell-targeted vaccines or TCR-grafted effector cells. All these approaches depend on recognition of HLA class I-presented viral peptides. The HBV core region 18-27 is an immunodominant target of CD8+ T cells and represents the prime target for T cell-based therapies. Here, a high-resolution analysis of the core18-27 specific CD8+ T cell and the selected escape pathways was performed. METHODS: HLA class I typing and viral sequence an

Design and development of a novel recombinant multi-epitope antigen: A promising candidate for melioidosis disease diagnosis.

Melioidosis, a deadly seasonal disease prevalent in the tropics and subtropics, is caused by exposure to the opportunistic pathogen Burkholderia pseudomallei. Diagnosing the disease is challenging due to its wide range of clinical symptoms and reliance on slow bacterial culture. Although molecular assays like real-time PCR offers high sensitivity and specificity, they are not suitable for field use and require trained personnel. Immunological tests often suffer from reduced accuracy in endemic regions due to the background seropositivity and the application of crude antigen preparations. To ad

B-Cell Epitope Mapping of TprC and TprD Variants of Treponema pallidum Subspecies Informs Vaccine Development for Human Treponematoses.

Several recent studies have focused on the identification, functional analysis, and structural characterization of outer membrane proteins (OMPs) of Treponema pallidum (Tp). The Tp species encompasses the highly related pallidum, pertenue, and endemicum subspecies of this pathogen, known to be the causative agents of syphilis, yaws, and bejel, respectively. These studies highlighted the importance of identifying surface-exposed OMP regions and the identification of B-cell epitopes that could be protective and used in vaccine development efforts. We previously reported that the TprC and TprD OM

B-Cell Epitope Mapping of the Vibrio cholera Toxins A, B, and P and an ELISA Assay.

Oral immunization with the choleric toxin (CT) elicits a high level of protection against its enterotoxin activities and can control cholera in endemic settings. However, the complete B-cell epitope map of the CT that is responsible for protection remains to be clarified. A library of one-hundred, twenty-two 15-mer peptides covering the entire sequence of the three chains of the CT protein (CTP) was prepared by SPOT synthesis. The immunoreactivity of membrane-bound peptides with sera from mice vaccinated with an oral inactivated vaccine (Schankol™) allowed the mapping of continuous B-cell epit

Prediction of linear B-cell epitopes of hepatitis C virus for vaccine development.

BACKGROUND: High genetic heterogeneity in the hepatitis C virus (HCV) is the major challenge of the development of an effective vaccine. Existing studies for developing HCV vaccines have mainly focused on T-cell immune response. However, identification of linear B-cell epitopes that can stimulate B-cell response is one of the major tasks of peptide-based vaccine development. Owing to the variability in B-cell epitope length, the prediction of B-cell epitopes is much more complex than that of T-cell epitopes. Furthermore, the motifs of linear B-cell epitopes in different pathogens are quite dif

Novel B cell epitopes mapping in pD205R protein of African swine fever virus using monoclonal antibodies.

BACKGROUND: African swine fever virus (ASFV) is a highly significant pathogen that causes hemorrhagic fever in domestic pigs and wild boars, resulting in substantial economic losses to the swine industry worldwide. The D205R protein (pD205R), which exhibits high similarity to the eukaryotic subunit 5 of RNA polymerase II (RPB5), has been identified as a component of the ASFV virion. However, the biological functions of pD205R remain largely unexplored. RESULTS: In this study, the recombinant pD205R (rpD205R) protein was expressed using a prokaryotic expression system, and five monoclonal anti

B-cell epitope mapping of Orthoflavivirus zikaense in mother-newborn immune interaction reveals postnatal immune signatures.

In 2015, the Orthoflavivirus zikaense (ZIKV) outbreak in Brazil caught the attention of the global community as a public health emergency. ZIKV, an Orthoflavivirus transmitted by Aedes mosquitoes, initially identified in 1947, has evolved from a historically inconspicuous pathogen to a significant public health concern. Beyond mild symptoms resembling Dengue fever, severe conditions such as microcephaly and Guillain-Barré syndrome have been linked to the infection caused by ZIKV. In regions already burdened by arboviruses such as Orthoflavivirus denguei (DENV), the co-circulation of these path

Concentration-Dependent Reactivity Profiles of a Human Monoclonal Antibody Recognising HLA-DQB1*04/05/06 and at Reduced Intensities to DQB1*03 Antigens.

Identifying HLA amino acid residues that account for an observed specificity (reactivity profile) is critical for serological categorisation at the B-cell epitope level. Human monoclonal antibody, LB_DQB0402_A, produced by an individual typed DQA1*03:03, DQA1*05:05/*05:11, DQB1*03:01, DQB1*03:01 against an immuniser typed DQA1*01:02, DQA1*05:05, DQB1*03:01, DQB1*06:02, reacts strongly to DQB1*04/05/06 single antigen beads (SABs), for which 55R, present in DQB1*04/05/06 alleles, has been previously proposed to be the target eplet. In this study, the antibody was also observed to react with DQB1

Design of a novel multiepitope vaccine against Chlamydia pneumoniae using the extracellular protein as a target.

Chlamydia pneumoniae (C. pneumoniae) infection in humans is universal and causes various respiratory infectious diseases, making a safe and effective preventive vaccine essential. In this study, a multi-epitope vaccine with CTLA-4 extracellular structure was constructed by an immunoinformatics approach. Since MOMP protein is the major extracellular protein in C. pneumoniae and has good immunogenicity and high conservation, we selected the MOMP protein of C. pneumoniae as the antigen target, predicted the T and B cell epitopes of the MOMP protein and then connected the CTLA-4 extracellular stru

Linear and conformational determinants of visceral leishmaniasis diagnostic antigens rK28 and rK39.

BACKGROUND: Recombinant antigens rK39 (based on kinesin sequence) and rK28 (comprising kinesin and HASPB sequences) are a mainstay of serological diagnosis for visceral leishmaniasis (VL). However, their key epitopes and the significance of their structural conformation are not clearly defined, particularly in relation to reported cross-reactivity with sera from patients with malaria, schistosomiasis, and tuberculosis. METHODS: To assess the effect of conformation on antigenicity with Sudanese VL sera, antigens rK39 and rK28 were heat-denatured at 95 °C for 10 min and then assayed by enzyme-l

A novel strategy for developing a tetravalent vaccine (dvac) against dengue utilizing conserved regions from all DENV proteins.

Dengue fever is a global health issue which is infecting millions of people each year and number of reported infections are constantly increasing. Though the only commercialized vaccine i.e. dengvaxia has banned in several countries due to its potential health risk, overall vaccine holds promising potential against viruses. In this study, we have developed a novel formulation of multi-epitope peptide vaccine (dvac), which utilizes peptides from each dengue protein with >80% sequence conservancy within each serotype and their respective genotypes. Simultaneous utilization of all dengue proteins

Variable and conserved B cell epitopes of GII.4 human noroviruses.

Norovirus is a leading cause of acute gastroenteritis worldwide, imposing a major burden on public health and healthcare systems. Despite its significant medical impact, no licensed vaccines or specific antiviral therapies are currently available. Norovirus vaccine development is complicated by several factors, including the extreme genetic and antigenic diversity. In particular, the predominant genotype GII.4 exhibits a continuous emergence of novel variants that can evade immune responses acquired from previous infections. In this manuscript, we will summarize the characteristics and current

Improved detection sensitivity of anti-PRV variant antibodies through preparation of anti-gB and anti-gE monoclonal antibodies and development of blocking ELISAs.

Since 2011, PRV has resurged in China and is characterized by a mutated strain with significant alterations in antigenicity and virulence. Therefore, we hypothesized that antibody detection kits based on classic PRV strains may have limitations in detecting PRV variants. For more sensitive antibody detection of PRV variants, two MABs targeting the gB and gE proteins were developed. IFA revealed that these MABs exhibited strong reactivity toward both classic and variant PRV strains. MAB-gE recognizes a novel conserved linear B-cell epitope (41PSAEVWD47), while MAB-gB recognizes a conformational

Effects of an epitope-specific CD8+ T-cell response on murine coronavirus central nervous system disease: protection from virus replication and antigen spread and selection of epitope escape mutants.

Both CD4(+) and CD8(+) T cells are required for clearance of the murine coronavirus mouse hepatitis virus (MHV) during acute infection. We investigated the effects of an epitope-specific CD8(+) T-cell response on acute infection of MHV, strain A59, in the murine CNS. Mice with CD8(+) T cells specific for gp33-41 (an H-2D(b)-restricted CD8(+) T-cell epitope derived from lymphocytic choriomeningitis glycoprotein) were infected with a recombinant MHV-A59, also expressing gp33-41, as a fusion protein with enhanced green fluorescent protein (EGFP). By 5 days postinfection, these mice showed signifi

Designing a novel multiepitope vaccine candidate against Treponema pallidum via adhesins using reverse vaccinology.

Syphilis, caused by Treponema pallidum (T. pallidum), represents a significant worldwide public health threat. This spiral-shaped, Gram-negative pathogen is a strict human-specific obligate parasite primarily transmitted through sexual contact. This pathogen induces a multistage and multisystem progressive disease, against which no effective prophylactic vaccine currently exists. This study focuses on syphilis prevention and control by employing a reverse vaccinology approach to investigate the immunogenic properties of T. pallidum adhesin proteins. Fifteen T-cell epitopes and seven B-cell epi

Designing a multi-epitope universal vaccine for concurrent infections of SARS-CoV-2 and influenza viruses using an immunoinformatics approach.

BACKGROUND: Severe acute respiratory syndrome coronavirus 2 (SARS‑CoV‑2) and influenza viruses share several conserved epitopes that can be utilized for the development of universal vaccines. Our previous research demonstrated that recombinant M2e-HA2 (Matrix-2 ectodomain-Hemagglutinin subunit 2) proteins derived from influenza elicited an immune response against the virus, suggesting their potential use in universal influenza vaccine formulations. Given the lack of a specific vaccine to address SARS‑CoV‑2 and influenza co-infections, this study aimed to design a universal vaccine using immuno

A Stable mRNA-Based Novel Multi-Epitope Vaccine Designs Against Infectious Heartland Virus by Integrated Immunoinformatics and Reverse Vaccinology Approaches.

The Heartland virus (HRTV) is a tick-borne human pathogenic phlebovirus that primarily causes leukopenia and thrombocytopenia. It is transmitted by Amblyomma americanum type of tick, that is, notable for their aggressive biting behavior, affinity for human hosts, and high prevalence. Developing vaccines or immunizations against HRTV is gaining importance as a public-health preventive strategy. The current study was planned to prioritize a multi-epitope stable mRNA vaccine model against HRTV from lead B-cell and T-cell epitopes (with IC50 < 100 nM) of HRTV proteome following advanced immunoinfo

Rational design of B-cell and T-cell multi epitope-based vaccine against Zika virus, an in silico study.

The Zika virus (ZKV) is a single-stranded positive-sense, enveloped RNA virus. Zika infection during pregnancy can cause congenital microcephaly, Guillain-Barré syndrome, miscarriage, and other CNS abnormalities. The world needs safe and effective vaccinations to fight against ZIKV infection since vaccination is generally regarded as one of the most effective ways to prevent infectious diseases. In the present work, we used immunoinformatics and docking studies to construct a vaccine containing multi-epitopes using the structural and non-structural proteins of ZKV. The structural models of ZKV
