Parsing the hypophagic and anxiogenic effects of cocaine in male and female rats.

Cocaine produces effects including euphoria, local anesthesia, hypophagia/anorexia, and anxiogenesis. The novelty-suppressed feeding (NSF) task is often used as a measure of anxiety-like behavior, except that this task is sensitive to changes in hunger state. First, we determined whether cocaine impacts behavior in the NSF task in male and female Long-Evans rats. Then, to determine whether cocaine-induced alterations in NSF behaviors are due to changes in motivated feeding, we measured the effects of cocaine on operant responding maintained by sucrose. Our results indicate that cocaine adminis

Lateral hypothalamic circuits for feeding and reward.

In experiments conducted over 60 years ago, the lateral hypothalamic area (LHA) was identified as a critical neuroanatomical substrate for motivated behavior. Electrical stimulation of the LHA induces voracious feeding even in well-fed animals. In the absence of food, animals will work tirelessly, often lever-pressing thousands of times per hour, for electrical stimulation at the same site that provokes feeding, drinking and other species-typical motivated behaviors. Here we review the classic findings from electrical stimulation studies and integrate them with more recent work that has used c

Variations in the 24 h temporal patterns and time budgets of grazing, rumination, and idling behaviors in grazing dairy cows in a New Zealand system.

This study investigated the variations in the temporal distributions and the lengths of times utilized for grazing, ruminating, and idling behaviors by grazing dairy cows over 24 h. Spring-calved lactating dairy cows (N = 54) from three breeds, Holstein-Friesian (HFR), Jersey (JE), and KiwiCross (KC) in different lactations (1st, 2nd, 3rd) and with different breeding worth index values (103 < BW > 151) were selected. The cows were managed through a rotational grazing scheme and milked once a day at 0500 hours. The cows grazed mainly pasture and consumed additional feeds (maize silage and turni

Seasonal variations in botanical and nutritive composition, forage availability and ingestive behavior of grazing goats in the semiarid region.

The aim of this study was to assess how the different seasons of the year impact the botanical composition, availability, and quality of forage in the caatinga, as well as the frequency of ingestion and ingestive behavior of goats in a semi-arid region. The experiment used four crossbred castrated males, with an average weight of 30.2 ± 4.76 kg and an average age of 20 months. The experimental period lasted for 12 months, encompassing two rainy seasons and one dry season within this timeframe, with three replications (data collection) per season. All collected forage was categorized based on i

Food makes you a target: disentangling genetic, physiological, and behavioral effects determining susceptibility to infection.

Genetics, physiology, and behavior are all expected to influence the susceptibility of hosts to parasites. Furthermore, interactions between genetic and other factors are suggested to contribute to the maintenance of genetic polymorphism in resistance when the relative susceptibility of host genotypes is context dependent. We used a maternal sibship design and long- and short-term food deprivation treatments to test the role of family-level genetic variation, body condition, physiological state, and foraging behavior on the susceptibility of Lymnaea stagnalis snails to infection by a trematode

Parallel strain-dependent susceptibility to environmentally-induced stereotypies and stress-induced behavioral sensitization in mice.

Food restriction (9 days) promoted stereotyped behavior in drug-free mice of the DBA/2 (DBA), but not in those of the C57BL/6 (C57), inbred strain. Indeed, behavior presented by food-restricted mice of the DBA strain within the home cage was characterised by a very high response rate within a single response: cage cover climbing. Moreover, enhanced climbing in food-restricted mice of the DBA strain was also observed in a test designed to detect stereotypic effects of drugs in mice. Stereotypic behavior in DBA mice did not depend on nutritional status because: 1. No stereotypies were observed i

Testing the Impact of Prey Presentation Method on the Feeding Kinematics of Terrestrial and Aquatic Ambystomatidae.

In animal behavior, standardizing experimental protocols ensures a rigorous interpretation of the results. However, when working with multisource data, homogeneous standardization is often difficult to attain. This is the case for many studies on feeding kinematics where experimental protocols are carried out using similar but not identical methods. In particular, the impact of the way in which prey is presented remains poorly tested and understood. The aim of this study is to assess whether prey position and the way prey is presented have an impact on the kinematics of prey capture. To do so,

Acute stress suppresses hunger-driven food seeking through PVN activation: Reversal by anxiolytic drug and ghrelin receptor agonist, with anxiolytic-like effects of refeeding.

Feeding behavior is influenced by both metabolic drive and emotional context, yet how acute stress interferes with hunger-driven motivation remains poorly understood. Using a conflict-based open-field feeding paradigm, we examined how 30-min restraint stress alters food-seeking behavior in fasted mice and how pharmacological or physiological interventions modulate this effect. Acute restraint abolished fasting-induced increases in food seeking and intake, an effect that was reversed by intraperitoneal diazepam, an anxiolytic, and by MK-677, a ghrelin receptor agonist that enhances appetite. To…

Weight Cycling Deregulates Eating Behavior in Mice via the Induction of Durable Gut Dysbiosis.

BACKGROUND & AIMS: Alternating periods of excessive and restrained eating results in weight cycling ('yo-yo' effect), a suspected risk factor for eating behavior dysregulation such as binge eating. The hypothesis that recurrent diet alternation alters hedonic feeding regulation by changing either or both intestinal microbiota and brain neuronal and glial regulation in mouse is tested. METHODS: C57BL/6 mice undergo 3 cycles of 1 week of western diet (WD) separated by 2 weeks of chow diet (CYCL group) or remain under chow diet (CTRL group). RESULTS: CYCL mice exhibit weight cycling, with enhan

scd knockout activates β-oxidation of fatty acids via accumulating stearic acid (18:0) and induces anorexia in zebrafish.

Stearoyl-CoA desaturase (scd) is the rate-limiting enzyme for the biosynthesis of monounsaturated fatty acids (MUFA), and it plays a critica … ed food intake and reduced body weight. Further analysis confirmed that scd knockout suppressed the feeding behavior by decreasing expression of orexigenic peptide genes and increasing expression of anorexigenic peptide genes. The high-level stearic acid (18:0) feeding experiment results showed that the accumulation of 18:0 inhibited feeding behavior, reduced food …

2-Ethylhexyl Diphenyl Phosphate Causes Obesity in Zebrafish by Stimulating Overeating via Inhibition of Dopamine Receptor D2.

Obesity is a popular public health problem worldwide and is mainly caused by overeating, but little is known about the impacts of synthetic chemicals on obesity. Herein, we evaluated the obesogenic effect caused by 2-ethylhexyl diphenyl phosphate (EHDPHP) on zebrafish. Adult zebrafish were exposed to 5, 35, and 245 μg/L of EHDPHP for 21 days. Results showed that EHDPHP exposure significantly promoted the feeding behavior of zebrafish, as evidenced by shorter reaction time, increased average food intake, feeding rate, and intake frequency (p < 0.05). Transcriptomic, real-time quantitative PCR, …

Overexpression of the Gene Encoding Neurosecretory Protein GL Precursor Prevents Excessive Fat Accumulation in the Adipose Tissue of Mice Fed a Long-Term High-Fat Diet.

We previously identified a novel small hypothalamic protein, neurosecretory protein GL (NPGL), which induces feeding behavior and fat accumulation in rodents depending on their diet. In the present study, we explored the effects of NPGL on feeding behavior and energy metabolism in mice placed on a long-term high-fat diet with 60% calories from fat (HFD 60). Overexpression of the NPGL precursor gene (Npgl) over 18 weeks increased food intake and weight. The weekly weight gain of Npgl-overexpressing mice was higher than that of controls until 7 weeks from induction of overexpression, after which…

Pharmacological characterization of the forced swim test in Drosophila melanogaster.

The forced swim test is commonly used as a preclinical screen of antidepressant medication efficacy in rats and mice. Neckameyer and Nieto-Romero (Stress 18:254-66, 2015) adopted the forced swim test for use with the fruit fly Drosophila melanogaster and showed that behavior in this test is sensitive to several physiologically relevant stressors. However, whether this test might be sensitive to the effects of antidepressant medications or other compounds is unknown. In the current studies, we fed drugs to male and female flies that we expected to either decrease or increase the duration of imm

Comparison of the capture efficiency, prey processing, and nutrient extraction in a generalist and a specialist spider predator.

Predators are traditionally classified as generalists and specialists based on the presence of adaptations that increase efficiency of prey capture and consumption and selection of particular prey types. Nevertheless, empirical evidence comparing foraging efficiency between generalist and specialist carnivores is scarce. We compared the prey-capture and feeding efficiency in a generalist and a specialist (araneophagous) spider predator. By using two related species, the generalist Harpactea rubicunda (Dysderidae) and the specialist Nops cf. variabilis (Caponiidae), we evaluated their fundament

Effects of central infusion of ghrelin on food intake and plasma levels of growth hormone, luteinizing hormone, prolactin, and cortisol secretion in sheep.

Ghrelin is an endogenous ligand for the GH secretagogue/ghrelin receptor (GHS-R) and stimulates feeding behavior and GH levels in rodents and humans. A preprandial increase in plasma ghrelin levels is seen in sheep on programmed feeding, followed by a postprandial rise in plasma GH levels, but effects on food intake and endocrine function are not defined in this ruminant species. We administered ghrelin to female sheep in various modes and measured effects on voluntary food intake (VFI) and plasma levels of GH, LH, prolactin, and cortisol. Whether administered intracerebroventricularly or iv, …

Plant-Derived BtHCYP Promotes Phloem Feeding and Fecundity in Bemisia tabaci.

Bemisia tabaci is a destructive agricultural pest with a remarkable capacity to exploit diverse host plants. Horizontally transferred genes (HTGs) have recently been implicated in this adaptive success, yet the functions of most HTGs in the whitefly remain unclear. Feeding behavior is crucial for nutrient acquisition and reproduction in piercing-sucking insects, but whether HTGs contribute to host adaptation by regulating feeding remains largely unknown. Here, we identified BtHCYP, a plant-derived HGT gene in B. tabaci encoding a cysteine protease. Biochemical and in vivo assays confirmed that…

The effect of repeat feeding on dengue virus transmission potential in Wolbachia-infected Aedes aegypti following extended egg quiescence.

As Wolbachia pipientis is more widely being released into field populations of Aedes aegypti for disease control, the ability to select the appropriate strain for differing environments is increasingly important. A previous study revealed that longer-term quiescence in the egg phase reduced the fertility of mosquitoes, especially those harboring the wAlbB Wolbachia strain. This infertility was also associated with a greater biting rate. Here, we attempt to quantify the effect of this heightened biting behavior on the transmission potential of the dengue virus using a combination of assays for

Optogenetic Analysis of Behavior in the Mosquito Aedes aegypti.

The mosquito Aedes aegypti is an important vector of viral pathogens and serves as a model for other vector species. Pathogens are transmitted when a mosquito bites a host animal, but the neural circuits that control seeking and biting behavior remain unknown. This paper details methods for manipulating neural activity in the mosquito using optogenetics, a key technique for determining the causal relationship between neural circuits and behavior. These methods include rearing mosquitoes for optogenetics and three assays designed to measure different steps in the sequence of arousal, attraction
