High-density lipoprotein, beta cells, and diabetes .

High-density lipoproteins (HDLs) exert a series of potentially beneficial effects on many cell types including anti-atherogenic actions on the endothelium and macrophage foam cells. HDLs may also exert anti-diabetogenic functions on the beta cells of the endocrine pancreas, notably by potently inhibiting stress-induced cell death and enhancing glucose-stimulated insulin secretion. HDLs have also been found to stimulate insulin-dependent and insulin-independent glucose uptake into skeletal muscle, adipose tissue, and liver. These experimental findings and the inverse association of HDL-choleste…

Lysosomal dysfunction in vascular smooth muscle cells: Mechanisms and therapeutic targets in vascular disease.

Vascular smooth muscle cell (VSMC) dysfunction is a common feature of atherosclerosis, aortic aneurysms, vascular calcification, and other vascular diseases. In VSMCs, lysosomes play an essential role in autophagic degradation, endocytic cargo processing, intracellular recycling, and cellular stress responses. Under pathological conditions, lysosomal dysfunction promotes foam cell formation, osteogenic differentiation, and extracellular matrix remodeling in VSMCs, thereby contributing to vascular diseases. This review summarizes current evidence on the role of lysosomal dysfunction in VSMC bio

[New findings on the pathogenesis of atherosclerosis].

The initial step in the process of atherosclerosis is supposed to be an increased penetration of lipoprotein particles (especially LDL or Lp … hagocytosis of the lipoprotein complexes with antibody or proteoglycans, macrophages transform into foam cells which are typical for atherosclerotic lesions. In addition, activated macrophages secern various growth factors and cytokins stimulating vascular smooth muscle cells to migrate, proliferate and form extracellular matrix.

SENP3 Deficiency Inhibits Atherosclerosis by Regulating TLR4/NF-κB and SOAT2.

SUMOylation plays critical roles in both initiation and development of atherosclerosis. SUMO-specific protease 3 (SENP3), a SUMO-specific protease that targets SUMO2/3 for deSUMOylation, is involved in vascular remodeling and the modulation of macrophage functions. Here, we probed for the role of SENP3 in macrophages in the development of atherosclerosis. Stable and unstable plaques were collected from patients with atherosclerosis. A macrophage-specific SENP3 knockout mouse (Senp3Mac-KO) was generated and performed for a murine atherosclerosis model. Transcriptional sequencing was performed t

Engineering ROS-responsive size-transformable nanoassemblies to reprogram plaque microenvironment and activate autophagy for atherosclerosis therapy.

Oxidative stress and macrophage autophagy dysfunction jointly drive plaque vulnerability and rupture, ultimately leading to acute atherothrombotic cardiovascular events. However, current monotherapies based solely on antioxidant intervention or autophagy induction remain insufficient for effective atherosclerosis management. Here, we reported the development of a cationic antioxidant dextran derivative (DKT) with intrinsic reactive oxygen species (ROS)-scavenging capacity, designed to co-deliver rapamycin-loaded reconstituted high-density lipoprotein (RC-rHDL) for synergistic atherosclerosis t

SMTP-44D Inhibits Atherosclerotic Plaque Formation in Apolipoprotein-E Null Mice Partly by Suppressing the AGEs-RAGE Axis.

SMTP-44D has been reported to have anti-oxidative and anti-inflammatory reactions, including reduced expression of receptor for advanced glycation end products (RAGE) in experimental diabetic neuropathy. Although activation of RAGE with its ligands, and advanced glycation end products (AGEs), play a crucial role in atherosclerotic cardiovascular disease, a leading cause of death in diabetic patients, it remains unclear whether SMTP-44D could inhibit experimental atherosclerosis by suppressing the AGEs-RAGE axis. In this study, we investigated the effects of SMTP-44D on atherosclerotic plaque f

Trigger Digits in Mucopolysaccharidosis Type I: Clinical Characteristics, Surgical Outcomes, and Histopathological Findings.

PURPOSE: Mucopolysaccharidosis type I (MPS-I), a genetic lysosomal storage disorder, often leads to musculoskeletal deformities such as trigger digits (TDs). Despite systemic therapies, connective tissues such as bone, muscle, and tendons remain difficult for the enzyme to reach, and TDs develop. We performed a retrospective study to assess the prevalence of TDs, evaluate surgical outcomes and recurrence, and explore accumulation of foam cell macrophages contributing to TD development in patients with MPS-I. METHODS: Data were obtained retrospectively from a cohort of 33 children born between

Tolerance driven lightweight design and interface robustness of multi material aircraft horizontal tail structures.

This study addresses the challenge of balancing weight reduction with stiffness in aircraft horizontal tails by proposing a multi-material design strategy combining carbon fiber reinforced polymer (CFRP) spars, closed-cell foam cores, and aluminum alloy joints. A three-dimensional nonlinear finite element model was developed to quantitatively assess how manufacturing tolerances—specifically variations in adhesive layer thickness and foam core density—affect interfacial mechanical performance. The co-optimized structure achieved a single-wing mass of 17.8 kg, representing a 32% reduction compar

A simplified method for monitoring cytokines in wound fluid.

Cytokines in wound fluid are used as surrogates for wound healing in clinical research. The current methods used to collect and process wound fluid are noninvasive but not optimal. The aim of this prospective study was to evaluate a method (NovaSwab) by which wound fluid is collected by a surface swab and eluted in a physiological buffer for subsequent cytokine analysis. Wound fluid from 12 patients with leg ulcers was assessed by NovaSwab at the start (Day 0) and at the end of a 23-h collection period of wound fluid retained by foam oblates beneath an occlusive film dressing (Day 1). New dressing combination for the treatment of partial thickness burn injuries in children.

Burns are among the most common causes of injury in children. Several wound dressings are available to conservatively treat second-degree burns. Seventy-three children were treated with Aquacel Ag foam and Zn-hyaluronon gel to determine their effectiveness on partial thickness burns. We applied silver nitrate solution on 25% of patients for 24 hrs, then checked burn depth. If the burn was superficial second degree, we applied the dressing under study. All dressings were removed 6-7 days later. In the remaining children we used Aquacel Ag foam dressing with Zn-hyaluronon gel at first interventi

Anorectal balloon cell melanoma: a rare variant.

Balloon cell melanoma is a rare presentation of malignant melanoma, usually on the skin, with less than 100 cases reported. Mucosal BCM is even rarer, with only one case of anorectal BCM reported in English literature. The diagnosis is based on the histopathologic findings of a tumor composed of large, foamy melanocytes, with or without pigmentation, and confirmed by immunohistochemical studies showing expression for melanocytic markers. The foam cell appearance of the tumor cells and the lack of melanin pigment lead to a diagnostic dilemma, mostly when presented at an unusual location. Herein

Bifurcation and dynamics in a mathematical model of early atherosclerosis: How acute inflammation drives lesion development.

We present here a mathematical model describing the primary mechanisms that drive the early stages of atherosclerosis. This involves the interactions between modified low density lipoprotein (LDL), monocytes/macrophages, cytokines and foam cells. This model suggests that there is an initial inflammatory phase associated with atherosclerotic lesion development and a longer, quasi-static process of plaque development inside the arterial wall that follows the initial transient. We will show results that show how different LDL concentrations in the blood stream and different immune responses can a…

Congenital nephrotic syndrome in Sudan: histological and immunological features.

We report a 5-month-old Sudanese boy with a probable diagnosis of Congenital Nephrotic Syndrome. Our case had features which are neither typical of the Finnish type nor of other hereditary renal diseases. Histologically, the most striking changes were in the glomerular basement membranes which show patchy thinning, thick segments (with reduplication) and occasional low spikes. Tubules are well preserved, and no foam cells were seen. Electron microscopy showed extensive fusion of foot processes with podocyte microvilli. In parts the glomerular basement membrane shows irregular thickening and sp…

Exploring the Impact of NRF2 Signaling on Atherosclerosis: Insights Into Hydrogen Sulfide, NLRP3 Inflammasome, microRNAs, and HO-1.

Atherosclerosis (AS) is a chronic cardiovascular disorder characterized by lipid accumulation and the formation of atherosclerotic plaques on the arterial walls, leading to arterial stenosis and serving as a principal pathological feature. This condition significantly contributes to elevated mortality and disability rates globally. The NRF2 pathway plays a pivotal role in the pathogenesis of AS. It governs a variety of physiological and pathophysiological processes including lipid homeostasis, foam cell formation, macrophage polarization, redox balance and inflammation, which are integral to t

Hepatic granulomas in leprosy. Their relation to bacteremia.

A clinicopathologic study of liver disease was conducted on 28 patients with leprosy who lived in Taiwan. None of the patients exhibited symptoms or signs of liver disease. Hepatic granulomas were found in 21 patients. Histologically, the infiltrates were epithelioid, foam cell, and histiocytic in type. Hepatic dysfunction was absent, except for mild sulfobromophthalein elevations in the severely infected cases. Hepatic granulomas correlated with the cutaneous reactions in lepromatous leprosy, but the association was poor for other stages of disease. Hepatic involvement varied with the severit

Integrated histopathology and single-cell transcriptomics reveal distinct pathological features and foamy macrophage characteristics in spinal tuberculosis and brucellar spondylitis.

BACKGROUND: Spinal tuberculosis (STB) and brucellar spondylitis (BS) show substantial overlap in clinical and imaging manifestations, but their local immunopathological programs and corresponding cellular states have not been systematically compared. This study integrated histopathology and single-cell transcriptomics to compare lesion-level differences between STB and BS and to characterize macrophage states associated with the foamy cell reaction. METHODS: This retrospective case-control study included 80 STB cases and 80 BS cases treated between January 2020 and December 2024. H&E-stained
