Minimolide, a sesquiterpene lactone isolated from Mikania minima, has displayed promising activity against Trypanosoma cruzi. The biocatalytic derivatization of minimolide and its analogs via selective acetylation and deacetylation is reported here using commercial lipases under mild and sustainable conditions. Reaction optimization identified Rhizomucor miehei lipase (RMIM) and Candida antarctica lipase B (CAL B) as the most efficient catalysts for acetylation and regioselective deacetylation, respectively. Structural transformations allow the synthesis of heliangolide and elemanolide analogs

Diversity-oriented synthesis of derivatives of natural products is an important approach for the discovery of novel drugs. In this paper, a series of novel 3,4-diaryl-1H-pyrazoles and 3,5-diaryl-1H-pyrazoles derivatives were synthesized through the one-pot reaction of flavones and isoflavones with the hydrazine hydrate and substituted hydrazine hydrate. Some of these novel compounds exhibited antifungal effects against Candida albicans SC5314, and displayed more potent inhibitory activities against the efflux-pump-deficient strain DSY654. In addition, compounds 25, 28 and 32a displayed outstan

The incidence of invasive fungal infections is rising due to the increase in susceptible populations. Current clinically available drugs have therapeutic limitations due to toxicity, a narrow spectrum of activity, and, more importantly, the consistent rise of fungal species that are intrinsically resistant or that develop resistance due to prolonged therapy. Thus, there is an urgent need for new broad-spectrum antifungal agents with low toxicity and a novel mechanism of action. We previously reported a new class of potent antifungal compounds, acylhydrazones, that target the fungal sphingolipi

Fungal resistance remains a significant threat and a leading cause of death worldwide. Thus, overcoming microbial infections have again become a serious clinical problem. Although acridine derivatives are widely analyzed as anticancer agents, only a few reports have demonstrated their antifungal activity. In an effort to develop biologically active antifungals, twelve novel C-857 (9-(2'-hydroxyethylamino)-1-nitroacridine) and C-1748 (9-(2'-hydroxyethylamino)-4-methyl-1-nitroacridine) derivatives were synthesized. The evaluation of biological properties suggests that starting compounds: C-1748,

An uncommon opportunistic fungal infection known as mucormycosis is caused by a class of molds called mucoromycetes. Currently, antifungal therapy and surgical debridement are the primary treatment options for mucormycosis. Despite the importance of comprehensive knowledge on mucormycosis, there is a lack of well-annotated databases that provide all relevant information. In this study, we have gathered and organized all available information related to mucormycosis that include disease's genome, proteins, diagnostic methods. Furthermore, using the AlphaFold2.0 prediction tool, we have predicte

Human polymorphonuclear leukocytes (PMNLs, neutrophils) play a major role in the immune response to bacterial and fungal infections and eliminate pathogens through phagocytosis. During phagocytosis of microorganisms, the 5-lipoxygenase (5-LOX) pathway is activated resulting in generation of leukotrienes, which mediate host defense. In this study, a library of oligodeoxyribonucleotides (ODNs) with varying numbers of human telomeric repeats (d(TTAGGG)n) and their analogues with phosphorothioate internucleotide linkages and single-nucleotide substitutions was designed. These ODNs with the potenti

Twelve drugs from four chemical classes are currently available for treatment of systemic fungal infections in humans. By contrast, more than 100 structurally distinct compounds from over 30 chemical classes have been developed as agricultural fungicides, and these fungicides target many modes of action not represented among human antifungal drugs. In this article we introduce the diverse aspects of agricultural fungicides and compare them with human antifungal drugs. We propose that the information gained from the development of agricultural fungicides can be applied to the discovery of new m

Mycoviruses, or fungal viruses, are widespread throughout the fungal kingdom. This study aimed to identify environmental factors associated with mycoviruses, explore their transmission mechanisms, including vector-mediated transmission, and assess their potential practical applications. A systematic, retrospective, and cross-sectional bibliographic review was conducted. These viruses are closely linked to their fungal hosts, thus developing a symbiotic relationship. Among environmental variables, temperature appears to play a more significant role in limiting fungal virulence than other factor

Benzimidazole and its derivatives have been extensively investigated for their antifungal activity, playing a crucial role in drug development. Initially, known for their agricultural applications, these compounds have evolved into significant therapeutic agents in medical mycology. The antifungal activity of benzimidazole was first identified in fungicides used in agriculture, especially for the management of fungal diseases in crops. Over time, its mode of action, mainly acting on fungal microtubules, led to its application in veterinary and human medicine. Early preclinical studies demonstr

Recent work demonstrates that habitat conditions exert striking effects on symbiont performance by mediating trade-offs in plants, AM fungi and environmental interactions. However, how local temperature conditions influence the functional diversity of mycorrhizal symbioses and the genetics of coexisting AM fungi at the local scale remain unclear. In the present study, we conducted a reciprocal inoculation experiment to explore the performance of sympatric associations against allopatric associations under contrasting temperatures and the AM fungal community in colonized roots. No local adaptat

To characterize the skin-associated fungal microbiota (mycobiota) in dogs, and to evaluate the influence of body site, individual dog or health status on the distribution of fungi, next-generation sequencing was performed targeting the internal transcribed spacer region. A total of 10 dogs with no history of skin disease were sampled at 10 distinct body sites consisting of haired and mucosal skin, and 8 dogs with diagnosed skin allergies were sampled at six body sites commonly affected by allergic disease. Analysis of similarities revealed that body site was not an influencing factor on member

The oral microbiome functions as an intricate and coordinated microbial network, residing throughout the oral cavity in both health and disease. Although most oral microbiome research has focused on bacteria, there is a growing interest in oral fungal communities, known as the oral mycobiome. The oral cavity hosts a complex and diverse mycobiome comprising of an estimated 100 fungal species; however, the roles of these fungi have been largely overlooked and remain insufficiently characterized, particularly in children. This represents a critical gap, as early life is a key window for establish

Phialophora verrucosa (P. verrucosa) is a pathogen that can cause chromoblastomycosis and phaeohyphomycosis. Recent evidence suggests that neutrophils can produce neutrophil extracellular traps (NETs) that can protect against invasive pathogens. As such, we herein explored the in vitro functional importance of P. verrucosa-induced NET formation. By assessing the co-localization of neutrophil elastase and DNA, we were able to confirm the formation of classical NETs entrapping P. verrucosa specimens. Sytox Green was then used to stain these NETs following neutrophil infection with P. verrucosa i

Ustilago maydis, the causal agent of corn smut disease, acquires and transports ferric ion by producing the extracellular, cyclic peptide, hydroxamate siderophores ferrichrome and ferrichrome A. Ferrichrome biosynthesis likely proceeds by hydroxylation and acetylation of L-ornithine, and later steps likely involve covalently bound thioester intermediates on a multimodular, nonribosomal peptide synthetase. sid1 encodes L-ornithine N(5)-oxygenase, which catalyzes hydroxylation of L-ornithine, the first committed step of ferrichrome and ferrichrome A biosynthesis in U. maydis. BACKGROUND: Forest nurseries are critical for producing resilient nursery stock for reforestation and planting of ornamental trees, yet the microbial communities associated with nursery grown plants remain poorly characterized. Quercus robur L. seedlings from seven Czech forest nurseries were analyzed to assess microbial diversity, co-occurrence patterns, and environmental drivers.
RESULTS: Microbial communities were characterized using fungal ITS2 and bacterial 16S rRNA gene amplicon sequencing, fungal isolation, and comprehensive soil chemistry. HTAS revealed broader taxonomic and functiona

Plant-associated fungi can host unique bacterial microbiota to provide multiple benefits to their fungal hosts. Here it was characterised the bacterial microbiota associated with an Epichloë fungal endophyte (strain AR135) isolated from perennial ryegrass (Lolium perenne) via both 16S rRNA gene sequencing and direct microbial isolation and investigated the microbe-microbe interactions between these bacteria and the fungus. The bacterial microbiota of AR135 was dominated by members within the genus Paenibacillus, with 99% of abundance (on average); although bacteria within genera Delftia and Br

Diagnosis of superficial/cutaneous fungal infections from skin, hair and nail samples is generally achieved using microscopy and culture in a microbiology laboratory, however, any presentation that is unusual or subcutaneous is sampled by taking a biopsy. Using histological techniques a tissue biopsy enables a pathologist to perform a full examination of the skin structure, detect any inflammatory processes or the presence of an infectious agent or foreign body. Histopathological examination can give a presumptive diagnosis while a culture result is pending, and may provide valuable diagnostic

The need for new therapies to treat systemic fungal infections continues to rise. Naturally occurring hexapeptide echinocandin B (1) has shown potent antifungal activity via its inhibition of the synthesis of beta-1,3 glucan, a key fungal cell wall component. Although this series of agents has been limited thus far based on their physicochemical characteristics, we have found that the synthesis of analogues bearing an aminoproline residue in the 'northwest' position imparts greatly improved water solubility (> 5 mg/mL). The synthesis and structure-activity relationships (SAR) based on whole ce

Invasive fungal infections have increased remarkably, which have become unprecedented concern to human health. However, the effectiveness of current antifungal drugs is limited due to drug resistance and toxic side-effects. It is urgently required to establish the effective biosynthetic strategy for developing novel and safe antifungal molecules economically. Echinocandins become a promising option as a mainstay family of antifungals, due to specifically targeting the fungal specific cell wall. To date, three kinds of echinocandins for caspofungin, anidulafungin, and micafungin, which derived
