Immunohistochemical detection of galectin-1 in renal biopsy specimens of children and its possible role in proteinuric glomerulopathies.

AIMS: Galectin-1 is an endogenous lectin that specifically binds to beta-galactoside structures. It has been associated with developmental mechanisms ranging from differentiation to apoptosis and exerts immunoregulatory functions in autoimmune diseases. The aim was to determine the immunohistochemical expression of galectin-1 in renal biopsy specimens of children with primary idiopathic proteinuric glomerulopathies. METHODS AND RESULTS: We examined 18 children with minimal change disease (MCD), 30 with diffuse mesangial proliferation (DMP) and 11 with focal segmental glomerulosclerosis (FSGS)

Effect of 5-azacytidine and galectin-1 on growth and differentiation of the human b lymphoma cell line bl36.

BACKGROUND: 5-AzaCytidine (AzaC) is a DNA demethylating drugs that has been shown to inhibit cell growth and to induce apoptosis in certain cancer cells. Induced expression of the galectin1 (Gal1) protein, a galactoside-binding protein distributed widely in immune cells, has been described in cultured hepatoma-derived cells treated with AzaC and this event may have a role in the effect of the drug. According to this hypothesis, we investigated the effect of AzaC and Gal1 on human lymphoid B cells phenotype. METHODS: The effect of AzaC and Gal1 on cell growth and phenotype was determined on the

Evaluation of serum human galectin 3 as a marker of activity in inflammatory bowel disease in comparison to fecal calprotectin.

Inflammatory Bowel Disease (IBD) is a chronic and recurring inflammatory condition of the gastrointestinal system that has an unknown cause and is marked by periods of relapse and remission. Endoscopy remains the gold standard for assessing disease activity; however, noninvasive biomarkers are needed to reduce the burden of repeated invasive procedures. This study aimed to evaluate the role of serum galectin 3 as a biomarker of disease activity in IBD and compare its performance with fecal calprotectin. The study included 40 individuals who were diagnosed with active IBD ulcerative colitis (UC

γ-Tocotrienol inhibition of galectin-3 expression, distribution and oligomerization in highly metastatic breast cancer cells.

Experimental studies were conducted to compare the relative effects of γ-tocotrienol, a natural isoform within the vitamin E family of compounds, with established galectin-3 inhibitor β-lactose, on galectin-3 levels and distribution in highly metastatic human MDA-MB-231, and mouse + SA and TS/A breast cancer cell lines. In contrast to β-lactose, γ-tocotrienol displays a dual-mode binding profile with the carbohydrate recognition domain and hydrophobic pocket located on galectin-3, as determined using Schrödinger molecular modeling. However, treatment with either γ-tocotrienol or β-lactose indu

Galectin-4 contributes to the maintenance of expression and activation of multiple receptor-type kinases involved in peritoneal metastasis.

Owing to the lack of effective therapeutic strategies, peritoneal metastasis of metastatic and recurrent gastric cancers is associated with poor prognosis. Our previous study showed that galectin-4 suppression attenuated the peritoneal metastasis of malignant gastric cancer cells in a mouse model; however, the underlying mechanisms are unclear. Therefore, we aimed to elucidate the role of galectin-4 in signal transduction by identifying interacting molecules and the relationship to glycans. Antibody array analysis and proximity ligation assays revealed a close association between galectin-4 an

Disrupting galectin-3 and NET-mediated microglia-neutrophil cross-talk suppresses pathological retinal angiogenesis.

Current research on pathological retinal neovascularization primarily focuses on growth factors, inflammation, and endothelial signaling pathways. However, increasing attention is being directed toward disruptions in the retinal immune microenvironment. Therefore, deciphering the immune-angiogenic interplay could uncover therapeutic avenues for neovascular disorders. Through integrative analyses of single-cell RNA sequencing from human fibrovascular membranes and multicohort clinical datasets, we identified neutrophil infiltration as an independent risk factor for diabetic retinopathy progress

Galectin-10 and Charcot-Leyden Crystals in Chronic Rhinosinusitis with Nasal Polyps: Pathogenetic Insights and Clinical Implications.

Chronic rhinosinusitis with nasal polyps (CRSwNP) is a heterogeneous inflammatory disease in which type 2 inflammation predominates in most patients in Western populations. Galectin-10 (Gal-10) and Charcot-Leyden crystals (CLCs), linked to eosinophil activation, cytolysis and eosinophil extracellular trap cell death (EETosis), have emerged as potential biomarkers and mediators of eosinophilic inflammation. This narrative review summarises current evidence on their biological role and clinical relevance in CRSwNP. The literature was identified through searches of PubMed/MEDLINE, Web of Science

Galectin-3 Release in the Bone Marrow Microenvironment Promotes Drug Resistance and Relapse in Acute Myeloid Leukemia.

Reciprocal signaling between acute myeloid leukemia (AML) cells and the surrounding bone-marrow microenvironment (BMME) promotes AML progression through several mechanisms. One of the most important mechanisms is the induction of Galectin-3 (Gal-3) expression by AML cells and bone marrow mesenchymal stromal cells (BM-MSCs). Emerging evidence indicates that Gal-3 upregulation in the BMME promotes AML cell adhesion and survival, leading to the development of chemotherapy resistance, relapse, and poor prognosis. Identifying the biological function and critical signaling pathways of Gal-3 may cont

Inhibitory Effect of Fucoidan Analogs on Highly Metastatic Gastric Cancer Cells via Galectin-4 Inhibition.

In malignant-type gastric cancer, peritoneal dissemination is the most frequent metastatic process and is an inoperable condition for which effective treatment is lacking. Our research has revealed that galectin-4 plays an important role in the peritoneal metastasis of gastric cancer cells. Based on this, we hypothesized that inhibiting galectin-4 could suppress peritoneal metastasis. The inhibitory activity towards galectin-4 binding was evaluated using an enzyme-linked immunosorbent assay, while the suppressive effect on gastric cancer cell proliferation was assessed using an adenosine triph

Galectin-3 in endometrial small extracellular vesicles promotes cytotrophoblast cell fusion during the implantation phase.

BACKGROUND: Small extracellular vesicles (sEVs) mediate intercellular communication by transporting microRNAs and proteins. Endometrial sEVs have been implicated in embryo implantation due to their content of implantation-related molecules; however, their precise functions and underlying mechanisms remain unclear. This study aimed to identify proteins in endometrial sEVs that contribute to embryo implantation and to clarify their functional significance. METHODS: Uterine fluid (UF) samples were collected from five women in each of the ovulatory and implantation phases. UF-derived sEVs (UF-sEV

Carotid femoral pulse wave velocity and serum galectin-3 level significantly increases in patients with acromegaly.

Insulin-like growth factor-1 (IGF-1) and growth hormone release is elevated in acromegaly, a chronic condition. Our objective was to study the correlation between carotid femoral pulse wave velocity (CF-PWV) and galectin-3 level in acromegaly patients. This protein is believed to alter in response to early inflammation and atherosclerosis. Our study was planned as a prospective, single-center cross-sectional study. Our study comprised 40 healthy controls and 40 newly diagnosed acromegaly patients ranging in age from 18 to 65 years old at Adana City Training and Research Hospital. The patients'

Cross-Sectional Study of Serum Galectin-3 Levels in Patients with Type 2 Diabetes and Colorectal Polyps.

Galectin-3 (Gal-3) secreted by activated macrophages is involved in inflammation, fibrosis, and tumorigenesis. It is considered a potential biomarker and therapeutic target. This study assessed the association between serum Gal-3, type 2 diabetes (T2D), and colorectal polyps (CRPs). In this cross-sectional study, 80 non-cancer patients undergoing colonoscopy were divided into four subgroups based on T2D and CRP status. Serum Gal-3 and metabolic parameters were measured. All patients' mean serum Gal-3 level was 13.63 ng/mL. Gal-3 levels were significantly higher in T2D+ than in the T2D- group (

Galectin-3/CD146 interaction promotes renal damage and systemic inflammation after acute kidney injury.

Galectin-3 (Gal-3) expression is abnormally increased after renal ischemic injury, in various studies, further promoting tissue damage. Gal-3 may interact with the endothelial layer to induce inflammation during renal injury, linking the systemic and tissue compartments. However, this mechanism has not been investigated after acute kidney injury (AKI). To this end, wild-type (WT) and Gal-3-/- mice underwent renal ischemia followed by reperfusion (rIR). Bone marrow transplantation was used to differentiate Gal-3 expression in renal tissue from systemic bone marrow cells. WT mice were injected w

Galectin-4 potentiates CD8+ T cell immunity by enhancing MHC-I expression on dendritic cells.

Galectin-4 (Gal-4), a member of the β-galactoside-binding galectin family, plays a role in various physiological processes, including tumor progression and intestinal disorders. However, its contribution to adaptive immunity remains poorly understood. In this study, Gal-4 is identified as a critical factor for effective generation of CD8+ T cell responses against tumors and viral infections. Gal-4-deficient mice exhibit significantly enhanced tumor growth in syngeneic mouse cancer models, attributed to impaired CD8+ T cell responses. Similarly, antiviral CD8+ T cell responses against lymphocyt

Redefining Immune Dynamics in Acute Pancreatitis: The Protective Role of Galectin-3 Deletion and Treg Cell Enhancement.

Acute pancreatitis (AP) is a complex inflammatory condition that can lead to systemic inflammatory responses and multiple organ dysfunction. This study investigates the role of Galectin-3 (Gal-3), a β-galactoside-binding lectin, in modulating acquired immune responses in AP. Acute pancreatitis was induced by ligation of the bile-pancreatic duct in wild-type and Galectin-3-deficient C57BL/6 mice. We determined the phenotypic and molecular features of inflammatory cells, serum concentrations of amylase, pancreatic trypsin activity, and pancreatic and lung pathology. Galectin-3 deficiency decreas

Vdelta1 T cells are more resistant than Vdelta2 T cells to the immunosuppressive properties of galectin-3.

Ovarian carcinomas have the highest lethality amongst gynecological tumors. A problem after primary resection is the recurrence of epithelial ovarian carcinomas which is often associated with chemotherapy resistance. To improve the clinical outcome, it is of high interest to consider alternative therapy strategies. Due to their pronounced plasticity, γδ T cells are attractive for T-cell-based immunotherapy. However, tumors might escape by the release of lectin galectin-3, which impairs γδ T-cell function. Hence, we tested the effect of galectin-3 on the different γδ T-cell subsets. After cocul
