Identification of Immune&Driver Molecular Subtypes Optimizes Immunotherapy Strategies for Gastric Cancer.

Immunotherapy has become a promising treatment for gastric cancer. However, its effectiveness varies significantly across subtypes because of heterogeneous immune microenvironments and genomic alterations. Here, we established Immune&Driver molecular subtypes CS1 and CS2 by systematically integrating multi-omics data for immune-related and driver genes. CS1 was linked to a better prognosis, while CS2 represented a poorer prognostic phenotype. CS1 displayed enhanced genomic instability, marked by higher mutation frequency and chromosomal alterations. In contrast, CS2 exhibited higher immune act

Integrative Analysis of Glycosylation-Related Genes Reveals Prognostic Subtypes, Immune Evasion, and Therapeutic Vulnerabilities in Lung Adenocarcinoma.

Background: Lung adenocarcinoma (LUAD) is the most common subtype of non-small cell lung cancer (NSCLC) and remains a leading cause of cancer-related mortality worldwide. Aberrant glycosylation contributes to tumor progression by regulating receptor signalling, immune evasion, and metastatic. However, the prognostic and therapeutic relevance of glycosylation-related genes (GRGs) in LUAD has not been comprehensively defined. Therefore, this study aimed to comprehensively evaluate GRG-associated molecular subtypes and their clinical and therapeutic relevance in LUAD. Methods: GRGs were curated f

Circadian clock control of breast cancer hallmarks: molecular mechanisms and therapeutic implications.

Circadian rhythms regulate physiological functions and critical biological processes in almost all living forms. The circadian system enables organisms to anticipate and adapt to environmental fluctuations, thereby optimizing physiological responses. Proper synchronization between the master pacemaker in the brain and peripheral clocks throughout the organism is essential for maintaining homeostasis and promoting health. Conversely, circadian misalignment is increasingly recognized as a contributing factor in the pathogenesis of various diseases. Emerging evidence implicates circadian disrupti

Prognostic impact of spatial niches in prostate cancer.

The formation of intratumoral spatial niches has been reported for many human malignancies. However, the translational potential of such spatial niches is understudied. Herein, we utilize digital spatial profiling (DSP) to explore the prognostic relevance of spatially defined protein expression in high-risk prostate cancer. A total of 49 patient samples were analyzed for the expression of 46 proteins in 463 regions of interest (ROIs) from the tumor center (n = 198) and the tumor periphery (n = 265) resulting in 21,298 primary data points (mean per patient n = 9.4). Expression data from either

Screening Biomarkers Related to Circadian Rhythm of Gastric Cancer Through Bioinformatics.

Circadian rhythm (CR) dysregulation is increasingly recognized as a contributor to gastric cancer (GC) development, yet the biological and immunological consequences of CR-related alterations remain unclear. In this study, integrative transcriptomic analyses identified 147 CR-related differentially expressed genes in GC. Functional enrichment revealed significant involvement in circadian processes, cytokine activity, and immune-related signaling pathways. By applying three complementary machine learning algorithms, five robust hub genes (INHBA, DEPTOR, PDCD4, SELENBP1, and SIRT4) were identifi

Targeting Spatiotemporal Heterogeneity of oncomiRNAs: A New Frontier in Cancer Therapy.

BACKGROUND: miRNAs are short RNA transcripts that modulate gene expression after transcription and have emerged as pivotal regulators of cancer biology. A subset, termed oncomiRNAs, functions as oncogenes or tumor suppressors, influencing key cellular events such as cell growth, programmed cell death, neovascularization, tissue invasion, and metastatic spread. Dysregulation of these miRNAs drives tumor initiation and progression, underscoring their role in cancer evolution. Traditionally, studies have relied on bulk tissue analyses, overlooking the profound spatiotemporal heterogeneity of onco

Construction of a prognostic model using lactylation-related genes for predicting the prognosis of glioma patients.

Gliomas are the most lethal malignant tumors of the central nervous system, and their treatment continues to face serious challenges. Increasing evidence suggests that lactylation is strongly associated with tumorigenesis and progression. However, studies of lactylation in gliomas are rare. In this study, we screened for lactylation-related genes in glioblastoma affecting patient prognosis based on TCGA and GEO databases and constructed a prediction model for lactylation-related genes using various machine learning methods. In addition, the researchers have also performed tumor somatic mutatio

Epigenetic implications in the pathogenesis of corticotroph tumors.

Non-mutational epigenetic reprogramming is considered an important enabling characteristic of neoplasia. Corticotroph tumors and other subtypes of pituitary tumors are characterized by distinct epigenetic profiles. The DNA methylation profile is consistent with disease-specific gene expression, which highlights the importance of epigenetic changes in tumor formation and progression. Elucidating the epigenetic changes underlying tumorigenesis plays an important role in understanding the molecular pathogenesis of corticotroph tumors and may ultimately contribute to improving tumor-specific treat

The Clinical Role of miRNAs in the Development and Treatment of Glioblastoma.

Glioblastoma multiforme (GBM) is the most common brain tumor and one of the most aggressive, with a median overall survival (OS) of only 15-18 months. These characteristics make it necessary to identify new targets for the improvement of prognosis and better prediction of response to therapies currently available for GBM patients. One possible candidate target could be the evaluation of miRNAs. miRNAs are small non-coding RNAs that play important roles in post-transcriptional gene regulation. Due to their functions, miRNAs also control biological processes underlying the development of GBM and

Functional plasticity of RNA-binding proteins in cancer: both friend and foe.

RNA-binding proteins (RBPs) govern RNA-based post-transcriptional processes that generate the abundance and diversity of the proteome. RBPs have recently emerged as crucial cancer regulators that can influence multiple cancer hallmarks. However, many RBPs display remarkable variations across different tumor types and can exert both tumor-promoting and tumor-suppressive effects. These opposing roles are often attributed to context-dependency, but there is a distinct lack of clarity regarding what aspects of cellular context define the differences in the roles of RBPs. Given the recent developme

Multiple roles of circular RNAs in prostate cancer: from the biological basis to potential clinical applications.

Prostate cancer is an important health concern affecting men. Circular RNAs (circRNAs) play an important molecular biological role in regulating gene expression due to their unique structure. Studies have revealed the involvement of circRNAs in many human diseases. In prostate cancer, circRNAs can act as oncogenes or tumour suppressor genes and affect cancer cell proliferation, invasion, resistance to chemotherapy and, consequently, disease progression. Accordingly, prostate cancer-related circRNAs are expected to serve as new targets in early clinical diagnosis and targeted therapy, but the v

[DDAH1 protein: biological functions, role in carcinogenesis processes].

Dimethylarginine Dimethylaminohydrolase 1 (DDAH1) is an essential enzyme capable of degrading asymmetric dimethylarginine, which is an endogenous inhibitor of nitric oxide synthase. Increased expression of DDAH1 and subsequent increased NO production are associated with carcinogenesis. In particular, DDAH1 is involved in the creation of a vascular network by tumor cells, vasculogenic mimicry, which is closely associated with tumor progression and poor patient prognosis. This is the reason why DDAH1 may be a potential therapeutic target for the treatment of cancer.

Evolving roles for the androgen receptor and its protein interactome in castration-resistant prostate cancer.

The androgen receptor (AR) is a ligand-activated transcription factor that is a major driver of lethal prostate cancer (CaP) progression. Androgen deprivation therapy (ADT) that prevents the binding of androgens to AR has been the mainstay for the treatment of non-organ-confined CaP for more than 8 decades. Although ADT initially induces remissions, eventually resistance occurs while the majority of castration-resistant CaPs (CRPCs) continue to rely on AR's action for growth. Sustained AR-dependence of CaP that recurs under ADT has historically been linked to AR's transcriptional activity that

Integrating machine learning models with multi-omics analysis to decipher the prognostic significance of mitotic catastrophe heterogeneity in bladder cancer.

BACKGROUND: Mitotic catastrophe is well-known as a major pathway of endogenous tumor death, but the prognostic significance of its heterogeneity regarding bladder cancer (BLCA) remains unclear. METHODS: Our study focused on digging deeper into the TCGA and GEO databases. Through differential expression analysis as well as Weighted Gene Co-expression Network Analysis (WGCNA), we identified dysregulated mitotic catastrophe-associated genes, followed by univariate cox regression as well as ten machine learning algorithms to construct robust prognostic models. Based on prognostic stratification,
