No causal effect of genetically determined circulating homocysteine levels on psoriasis in the European population: evidence from a Mendelian randomization study.

BACKGROUND: Although numerous studies demonstrated a link between plasma homocysteine (Hcy) levels and psoriasis, there still exists a certain level of controversy. Therefore, we conducted a Mendelian randomization study to investigate whether homocysteine plays a causative role in the development or exacerbation of psoriasis. METHODS: A two-sample Mendelian randomization (MR) analysis was conducted. Summary-level data for psoriasis were acquired from the latest R9 release results from the FinnGen consortium (9,267 cases and 364,071 controls). Single nucleotide polymorphisms (SNPs) robustly l

Acne and risk of mental disorders: A two-sample Mendelian randomization study based on large genome-wide association data.

BACKGROUND: Despite a growing body of evidence that acne impacts mental disorders, the actual causality has not been established for the possible presence of recall bias and confounders in observational studies. METHODS: We performed a two-sample Mendelian randomization (MR) analysis to evaluate the effect of acne on the risk of six common mental disorders, i.e., depression, anxiety, schizophrenia, obsessive-compulsive disorder (OCD), bipolar disorder, and post-traumatic stress disorder (PTSD). We acquired genetic instruments for assessing acne from the largest genome-wide association study (

Investigating the Pleiotropic Role of KIF21B in Schizophrenia and Multiple Sclerosis: A Bioinformatics Analysis.

OBJECTIVE: Genome-wide association studies have identified shared genetic risk loci between schizophrenia (SCZ) and multiple sclerosis (MS), suggesting overlapping biological mechanisms despite distinct clinical presentations. Here, we aimed to characterize the biological plausibility of KIF21B as a shared genetic risk gene for SCZ and MS, two disorders with overlapping genetic risk loci despite distinct clinical presentations. METHOD: We performed bioinformatics analyses of KIF21B using publicly available data from GTEx, STRING, and the GWAS Catalog, including tissue-specific expression prof

Unraveling the causal link between immune cells and temporomandibular disorders: A Mendelian randomization analysis.

The host's immune inflammatory response plays a crucial role in the pathogenesis of temporomandibular disorders (TMD), involving numerous innate immune effector cells to initiate the initial inflammatory response. However, the regulation of immune function and the composition of immune cells in relation to the occurrence and development of TMD remain incompletely elucidated. The genetic causality of immune cells on TMD was studied using Mendelian randomization in a two-sample study. With the help of publicly available genetic data, we examined the causal associations between 731 immune cell si

According to the sensitivity analysis, the presence of heterogeneity and genetic pleiotropy was not detected. Our findings, derived from a genetic perspective, provide robust evidence of a causal connection between BMR and SSc. To verify these results and clarify the potential mechanisms, further research is warranted.

Outbreeding alleviates senescence in hermaphroditic snails as expected from the mutation-accumulation theory.

Senescence, the decline in fitness components of an organism with age [1], is a nearly universal characteristic of living beings [2-6]. This ubiquity is challenging because natural selection does not favor the evolution of traits decreasing fitness [1, 7, 8]. Senescence may result from two nonexclusive mechanisms: the accumulation of deleterious mutations acting late in life, when the strength of natural selection against them declines [9-11] (mutation accumulation or MA hypothesis [12]) and the delayed cost of genes having beneficial effects early in life (antagonistic pleiotropy or AP hypoth

SNP-based selection strategies reveal antagonistic and favorable genetic architectures between milk production and fertility traits in dairy cattle: A simulation study.

This study evaluated genetic responses to selection based on single-nucleotide polymorphism (SNP) subsets with different effect directions on milk production and fertility in dairy cattle. A population of 1000 individuals was simulated using 50,000 SNP markers sampled from the Illumina BovineSNP50 BeadChip panel to represent a realistic genomic architecture. SNP effects were derived from previously published genome-wide association study (GWAS) results, in which the direction of SNP effects was inferred from reported test statistics (t-values). Five selection scenarios were evaluated over 50 g

Phenome-wide genetic-correlation analysis and genetically informed causal inference of amyotrophic lateral sclerosis.

Leveraging genome-wide association statistics generated from a large study of amyotrophic lateral sclerosis (ALS; 29,612 cases and 122,656 controls) and UK Biobank (UKB; 4,024 phenotypes, up to 361,194 participants), we conducted a phenome-wide analysis of ALS genetic liability and identified 46 genetically correlated traits, such as fluid intelligence score (rg = - 0.21, p = 1.74 × 10-6), "spending time in pub or social club" (rg = 0.24, p = 2.77 × 10-6), non-work related walking (rg = - 0.25, p = 1.95 × 10-6), college education (rg = - 0.15, p = 7.08 × 10-5), "ever diagnosed with panic attac

Disentangling selection on genetically correlated polygenic traits via whole-genome genealogies.

We present a full-likelihood method to infer polygenic adaptation from DNA sequence variation and GWAS summary statistics to quantify recent transient directional selection acting on a complex trait. Through simulations of polygenic trait architecture evolution and GWASs, we show the method substantially improves power over current methods. We examine the robustness of the method under stratification, uncertainty and bias in marginal effects, uncertainty in the causal SNPs, allelic heterogeneity, negative selection, and low GWAS sample size. The method can quantify selection acting on correlat

No genome-wide correlations and little shared genetic architecture between reproductive life-history traits and estrogen receptor-positive breast cancer risk.

Estrogen receptor-positive breast cancer (ER + BC) is one of the most prevalent cancers, but the evolutionary processes shaping genetic variation in ER + BC risk are poorly understood. Both evolutionary life-history theory and evidence from studies of individual ER + BC risk variants suggest that increased genetic ER + BC risk is associated with faster maturation, earlier reproduction, and/or increased reproductive success (i.e. there is a trade-off), but it is unclear how well this pattern is replicated when considering the polygenic architecture of these traits after controlling for potentia

Shared genetic architecture and cellular convergence between female reproductive disorders and pulmonary function: a genome-wide cross-trait analysis.

Female reproductive disorders (FRDs), including polycystic ovary syndrome, endometriosis, uterine leiomyomata, and infertility, have been epidemiologically associated with impaired pulmonary function. However, it remains unclear whether this cross-organ link reflects shared genetic etiology and, if so, which cellular mechanisms mediate it. We performed a systematic genome-wide cross-trait analysis of three FRDs and lung function traits (FEV₁, FVC, FEV₁/FVC) using GWAS summary statistics from individuals of European ancestry, integrating genetic correlation, bidirectional causal inference, plei

Causal effects of various particulate matter on inflammatory bowel disease and its subtypes: insights from Mendelian randomization.

The role of particulate matter (PM) on genetic susceptibility to inflammatory bowel disease (IBD) is a controversial topic. The study aims to evaluate the causal effect of PM on IBD using Mendelian randomization (MR). PM and IBD datasets were screened for common single nucleotide polymorphisms (SNPs) based on the MR basic assumptions. Subsequently, the effect of these SNPs on genetic susceptibility to IBD was analyzed using inverse variance weighted. Finally, the heterogeneity and robustness of the results were assessed using Cochran's Q and leave-one-out sensitivity analysis, respectively. MR

Cross-trait genomic and sequential analyses of multiple omics datasets identified shared genetic components for the gut-eye axis.

BACKGROUND: The gut–eye axis (GEA) has been proposed as a framework for understanding comorbidity between gastrointestinal and ocular diseases. This study aimed to investigate their shared genetic architecture, pleiotropy, and putative biological pathways potentially influenced by environmental exposures and gut microbiota. METHODS: This study integrated large-scale genome-wide association study summary data on five gastrointestinal and eight ocular diseases to assess genetic correlations and genetic overlap. Pleiotropic variants were identified, followed by functional and tissue-specific anal

Mapping the genetic landscape across 14 psychiatric disorders.

Psychiatric disorders display high levels of comorbidity and genetic overlap1,2, challenging current diagnostic boundaries. For disorders for which diagnostic separation has been most debated, such as schizophrenia and bipolar disorder3, genomic methods have revealed that the majority of genetic signal is shared4. While over a hundred pleiotropic loci have been identified by recent cross-disorder analyses5, the full scope of shared and disorder-specific genetic influences remains poorly defined. Here we addressed this gap by triangulating across a suite of cutting-edge statistical and function

KIAA0319 and ROBO1: evidence on association with reading and pleiotropic effects on language and mathematics abilities in developmental dyslexia.

Substantial heritability has been reported for developmental dyslexia (DD), and KIAA0319 and ROBO1 appear as more than plausible candidate susceptibility genes for this developmental disorder. Converging evidence indicates that developmental difficulties in oral language and mathematics can predate or co-occur with DD, and substantial genetic correlations have been found between these abilities and reading traits. In this study, we explored the role of eight single-nucleotide polymorphisms spanning within KIAA0319 and ROBO1 genes, and DD as a dichotomic trait, related neuropsychological phenot

Investigating the shared genetic architecture between primary sclerosing cholangitis and inflammatory bowel diseases: a Mendelian randomization study.

BACKGROUND: Several studies have found that primary sclerosing cholangitis (PSC) and inflammatory bowel disease (IBD) are closely associated. However, the direction and causality of their interactions remain unclear. Thus, this study employs Mendelian Randomization to explore whether there are causal associations of genetically predicted PSC with IBD. METHODS: Genetic variants associated with the genome-wide association study (GWAS) of PSC were used as instrumental variables. The statistics for IBD, including ulcerative colitis (UC), and Crohn's disease (CD) were derived from GWAS. Then, five

Identification of novel variants associated with osteoporosis, type 2 diabetes and potentially pleiotropic loci using pleiotropic cFDR method.

AIMS: Clinical and epidemiological findings point to an association between type 2 diabetes (T2D) and osteoporosis. Genome-wide association studies (GWASs) have been fruitful in identifying some loci potentially associated with osteoporosis and T2D respectively. However, the total genetic variance for each of these two diseases and the shared genetic determination between them are largely unknown. The aim of this study was to identify novel genetic variants for osteoporosis and/or T2D. METHODS: First, using a pleiotropic conditional false discovery rate (cFDR) method, we analyzed two GWAS sum
