The role of functional polymorphisms in immune response genes as biomarkers of bacille Calmette-Guérin (BCG) immunotherapy outcome in bladder cancer: establishment of a predictive profile in a Southern Europe population.

OBJECTIVE: To evaluate the predictive value of genetic polymorphisms in the context of bacille Calmette-Guérin (BCG) immunotherapy outcome and create a predictive profile that may allow discrimination of the risk of recurrence. PATIENTS AND METHODS: In a dataset of 204 patients treated with BCG, we evaluated 42 genetic polymorphisms in 38 genes involved in the BCG mechanism of action, using Sequenom MassARRAY(®) technology. Stepwise multivariate Cox regression was used for data mining. RESULTS: In agreement with previous studies we found that gender, age, tumour multiplicity and treatment sc

Bayesian approach to assessing population differences in genetic risk of disease with application to prostate cancer.

Population differences in risk of disease are common, but the potential genetic basis for these differences is not well understood. A standard approach is to compare genetic risk across populations by testing for mean differences in polygenic scores, but existing studies that use this approach do not account for statistical noise in effect estimates (i.e., the GWAS betas) that arise due to the finite sample size of GWAS training data. Here, we show using Bayesian polygenic score methods that the level of uncertainty in estimates of genetic risk differences across populations is highly dependen

The relationship in women between genetic liability and the risk of onset of alcohol use disorder while pregnant or rearing an infant, toddler, or preschool child.

While pregnancy clearly reduces the risk for Alcohol Use Disorder (AUD) onset, we know less about the impact on AUD risk of having young children and how these effects vary across maternal age and level of maternal AUD genetic risk. Therefore, in 1.2 million parous Swedish women born 1960-1995, we examined those with a first registration for AUD between ages 15-40 while first pregnant, or while raising their first infant (aged 0-12 months), toddler (13-36 months) or preschooler (37-60 months). Genetic risk for AUD was assessed by their family genetic risk score. Pregnancy and having an infant …

The cumulative effects of known susceptibility variants to predict primary biliary cirrhosis risk.

Multiple genetic variants influence the risk for development of primary biliary cirrhosis (PBC). To explore the cumulative effects of known susceptibility loci on risk, we utilized a weighted genetic risk score (wGRS) to evaluate whether genetic information can predict susceptibility. The wGRS was created using 26 known susceptibility loci and investigated in 1840 UK PBC and 5164 controls. Our data indicate that the wGRS was significantly different between PBC and controls (P=1.61E-142). Moreover, we assessed predictive performance of wGRS on disease status by calculating the area under the re…

Common Single Nucleotide Polymorphisms in Clinical Cardiology and Dietary Intervention: A Narrative Review.

Genomic testing for rare, highly penetrant cardiovascular pathogenic mutations is well established, but its scarcity limits broader clinical utility. As cardiogenomics matures, common single nucleotide polymorphisms (SNPs) associated with cardiovascular disease (CVD) risk are increasingly accessible to clinicians and patients through both clinician-ordered and direct-to-consumer (DTC) platforms. This narrative review synthesizes evidence for six common loci-APOA1, APOE, LIPC, LPL, ANGPTL3, and FADS1/2, here termed "candidate-actionable" in the limited sense that genotype-by-diet associations h

Loneliness as a risk factor for bipolar disorder and its related brain structure alterations.

BACKGROUND: Loneliness has been linked to increased risk of mood disorders, but its relationship with bipolar disorder (BD) and underlying neurobiological mechanisms remain unclear. METHODS: This prospective cohort study utilized data from the UK Biobank, involving 491,472 individuals aged 38-73 years without prior BD diagnosis at baseline (2006-2010). Participants reported loneliness and provided data on demographic, socioeconomic, lifestyle factors, and polygenic risk scores (PRS) for BD. Follow-up extended until 2022, tracking incident BD cases. Neuroimaging analyses examined gray matter v

TGF-β Pathway-Based Polygenic Risk Score Modifies the Association between Red Meat Intake and Colorectal Cancer Risk: Application of a Novel Pathway-Based PRS Method.

BACKGROUND: Red meat and/or processed meat are established colorectal cancer risk factors. Genome-wide association studies (GWAS) have reported more than 200 variants associated with colorectal cancer risk. We used functional annotation data to identify subsets of variants within known pathways to construct pathway-based polygenic risk scores (pPRS) to assess interactions with meat intake. METHODS: A pooled sample of 30,812 cases and 40,504 colorectal cancer controls from 27 studies was analyzed. Quantiles for red and processed meat intake were constructed. A total of 204 GWAS variants were a

Neonatal vitamin D status and asthma risk after age 5 years: A Danish population-based cohort study.

BACKGROUND: Vitamin D may play a role in early lung development, yet epidemiologic evidence on its association with later asthma risk is mixed. We aimed to investigate the associations of neonatal 25-hydroxyvitamin D (25(OH)D) and vitamin D-binding protein (DBP) and their corresponding genetic predictors with asthma risk. METHODS: We conducted a population-based cohort study of a random sample of individuals born in Denmark during 1991-2005 from the iPSYCH2012 study. Neonatal concentrations of 25(OH)D and DBP were measured via dried blood spots, and asthma cases were identified through diagno

Population-specific polygenic risk scores for people of Han Chinese ancestry.

Predicting complex disease risks on the basis of individual genomic profiles is an advancing field in human genetics1,2. However, most genetic studies have focused on populations of European ancestry, creating a global imbalance in precision medicine and underscoring the need for genomic research in non-European groups3,4. The Taiwan Precision Medicine Initiative recruited more than half a million Taiwanese residents, providing a large dataset of genetic profiles and electronic medical record data for people with Han Chinese ancestry. Using extensive phenotypic data, we conducted comprehensive

Nonlinearity and sex differences in the performance of a polygenic risk score for juvenile idiopathic arthritis.

BACKGROUND: Juvenile idiopathic arthritis (JIA) is an immune-mediated pediatric disease believed to result from a complex interplay of genetic and environmental factors. Genome-wide association studies have enabled calculation of polygenic risk scores (PRS) for JIA. Understanding how the PRS associates with JIA and whether it performs similarly across sexes is essential for its utility in future studies. METHODS: We studied the relationship between a PRS developed from a previously published genome-wide association study of JIA and JIA in children from the Norwegian Mother, Father and Child C
