PURPOSE: We analyzed the levels of glutamate and other amino acids in aqueous humor of patients with retinal artery occlusion (RAO) to determine whether glutamate is associated with retinal ischemia in RAO.
METHODS: Aqueous humor samples were obtained from nine RAO patients by paracentesis performed as an emergent therapeutic intervention and from nine cataract patients without other ocular disease who served as controls. Thiamine-deficient encephalopathy in the rat is characterized by ataxic gait, loss of righting reflex and curvature of the spine. Neurochemical changes include a diminished activity of cerebral pyruvate decarboxylase leading to abnormal pyruvate oxidation. The enantiomers of beta,gamma-dimethyl- and beta-methyl-alpha-methylene-gamma-butyrolactones have been synthesized stereospecifically from glutamic acid and beta-hydroxy isobutyric acid, respectively. Guinea pigs have been sensitized (Freund complete adjuvant technique) and tested to them. Both enantiomers of beta-methyl lactone as well as (+)-beta,gamma-dimethyl lactone induced enantiospecific allergic contact dermatitis (ACD); in turn, (-)-beta,gamma-dimethyl lactone showed no specificity. The albumin-like fraction of psoriatic scales was isolated either directly by slab gel electroporesis or after previous treatment with polyethylene glycol6000. After both isolation procedures the amino acid composition was determined. Both proteins were rich of glutamic acid, glycine, and alanine. On the other hand both contained very small amounts of cysteine and tyrosine. Only after the direct isolation procedure the amino-sugar galactosamine could be detected.

Poly-alpha-cyanoglycine, a homopolymer synthesized from the N-carboxyanhydride of alpha-cyanoglycine, is converted by cumulative reaction of hydrogen cyanide to heteropolypeptides that can be hydrolyzed to protein amino acids, including glycine, alanine, valine, aspartic acid, and glutamic acid. These results are consistent with the hypothesis that the original heteropolypeptides on the earth arose spontaneously from hydrogen cyanide and water without the intervening formation of alpha-amino acids.
