Growth impairment is a clinical manifestation frequently observed in pediatric patients with cardiomyopathy associated with various inherited disorders, including RASopathies, lysosomal storage diseases, neuromuscular disorders, and metabolic conditions. In this narrative review, we explored the genetic and pathophysiological mechanisms underlying the development of both growth and myocardial impairment in Noonan syndrome (NS)-the most common RASopathy-Duchenne and Becker muscular dystrophies, Pompe disease, mucopolysaccharidoses, and mitochondrial diseases. For each condition, we described th

Abstract Of diverse growth factors that contribute to normal lung development, vascular endothelial growth factor (VEGF) plays an especially prominent role in the normal growth and development of the pulmonary circulation in the fetus and newborn. Strong experimental and clinical data support the role of impaired VEGF signaling in the pathogenesis of two major clinical disorders of the developing lung circulation: persistent pulmonary hypertension of the newborn (PPHN) and bronchopulmonary dysplasia (BPD). These disorders are each characterized by impaired vascular growth, structure and reacti

OBJECTIVES: The efficacy of electroconvulsive therapy (ECT) in treating mood disorders (MDs) is hypothesized to be mediated by the induction of neurotrophic factors (denoted "angioneurins") that trigger neuronal plasticity. This study aimed to assess the effects of ECT on serum angioneurin levels in patients with MD.
METHODS: A total of 110 patients with MDs including 30 with unipolar depression, 25 with bipolar depression (BD), 55 with bipolar mania (BM), and 50 healthy controls were included in the study. Patients were subdivided into two groups: those who received ECT + medication (12 ECT

Longitudinal growth is primarily influenced by the GH-IGF-I axis, which is a mixed endocrine-paracrine-autocrine system. Further, classical hormones such as thyroxine, glucocorticosteroids and sex steroids play a role, as well as primarily paracrine systems. In the GH-IGF-I axis, seven disorders can be differentiated: (1) GH deficiency; (2) GHR defects; (3) defects in the GH signal transduction pathway; (4) IGF1 defects; (5) IGFALS defects; (6) IGF1R defects, and (7) IGF2 defects. Children with one of the first 3 disorders have near-normal prenatal growth, while children with defects of IGF1,

PURPOSE OF REVIEW: Bone elongation is a complex process driven by multiple intrinsic (hormones, growth factors) and extrinsic (nutrition, environment) variables. Bones grow in length by endochondral ossification in cartilaginous growth plates at ends of developing long bones. This review provides an updated overview of the important factors that influence this process.
RECENT FINDINGS: Insulin-like growth factor-1 (IGF-1) is the major hormone required for growth and a drug for treating pediatric skeletal disorders. Temperature is an underrecognized environmental variable that also impacts lin

Tall stature and/or accelerated growth (TS/AG) in a child can be the result of a primary or secondary growth disorder, but more frequently no cause can be found (idiopathic TS). The conditions with the most important therapeutic implications are Klinefelter syndrome, Marfan syndrome and secondary growth disorders such as precocious puberty, hyperthyroidism and growth hormone excess. We propose a diagnostic flow chart offering a systematic approach to evaluate children referred for TS/AG to the general paediatrician. Based on the incidence, prevalence and clinical features of medical conditions…

Otoneurological examination with the use of modern methods (electronystagmography, electrogustometry, supraliminal cupulometry, audiometry) showed that some type of disorders are encountered in 100% of cases of tumors of the hypophysis with marked extrasellar growth (vestibular disorders in 94% of cases, disturbed sensitivity in the nasal cavity in 55%, disorders of hearing and the sense of taste in 18%, disorders of the sense of smell in 17%). The otoneurological symptoms yield additional information on the direction of the tumor growth and, what is most important, on the degree of its effect

BACKGROUND: Sleep disorders are common in childhood and are recognized as a significant public health concern, particularly due to their association with childhood obesity. However, their impact on growth disturbances has been only minimally explored. In the present study, we investigate the association between sleep disorders and linear growth and excess weight in children.
STUDY DESIGN: Data were collected retrospectively (January 2012-December 2022) from computerized medical records of 3,210 patients (newborn-18 years) diagnosed with sleep disorders who underwent sleep analysis at the Slee

OBJECTIVES: Research suggests that individuals treated for substance use disorders are not routinely assessed for post-traumatic stress disorder (PTSD) symptomatology despite high rates of comorbidity. One area of research that has been overlooked in theory, research, and practice on the comorbidity of substance use disorders and PTSD is the study of post-traumatic growth. The purpose of the current study is to explore the relevance of post-traumatic growth for the treatment of comorbid substance use disorders among individuals suffering from traumatic stress or PTSD.
METHOD: A sample of 256

The development of novel pharmaceutical treatments for disorders of the cerebral vasculature is a serious unmet medical need. These vascular disorders are typified by a disruption in the delicate Rho signaling equilibrium within the blood vessel wall. In particular, Rho kinase overactivation in the smooth muscle and endothelial layers of the vessel wall results in cytoskeletal modifications that lead to reduced vascular integrity and abnormal vascular growth. Rho kinase is thus a promising target for the treatment of cerebral vascular disorders. Indeed, preclinical studies indicate that Rho ki

Patients with juvenile idiopathic arthritis (JIA) can have alterations in bone metabolism and skeletal growth, as well as damage to the temporomandibular joint (TMJ), which can generate extra and/or intraoral alterations, resulting in craniofacial disorders. Our goal is to carry out a review of the literature on orofacial alterations in patients with JIA. Among the orofacial disorders in patients with JIA, alterations in mandibular growth, caused by dysfunctions in the TMJ region, seem highly prevalent in these patients. The most often found alterations are: retrognathia, micrognathia, anterio

BACKGROUND: Infantile anorexia nervosa (AN) is a specific eating disorder of prepubertal children. Poor data are available on growth hormone (GH)-insulin-like growth factor 1 (IGF-1) axis in this disorder.
PATIENT REPORT: We report on a boy (4.5 years) with progressive growth impairment. At psychiatric assessment (DC: 0-3 R, AXIS I), he fulfilled all required criteria for diagnosis of infantile AN. Endocrine evaluation suggested impaired peripheral response to GH (high GH and low IGF-1 levels), likely related to energy deficiency.
METHODS: Auxological evaluation was shown as raw data and SDS

After a decade of experience with recombinant human growth hormone (rhGH) in children with chronic renal failure (CRF), the long-term efficacy and safety of the drug is now established. In prepubertal children, partial catch-up growth is achieved during the first three treatment years, followed by sustained percentile-parallel growth. Discontinuation of rhGH treatment results in catch-down growth in 75% of patients. Treatment efficacy is inversely correlated with age and baseline height velocity, and positively influenced by genetic target height and residual renal function. Skeletal maturatio

The greater availability of growth hormone (GH) and the increasing knowledge of growth hormone physiology have led to a wider application of growth hormone treatment. GH is approved for use in the treatment of pituitary deficiency syndromes and, when properly administered at an early stage, is capable of inducing normal skeletal and muscular development. It has also been successfully used to augment growth in patients with Turner syndrome and the effect on the abnormal craniofacial skeleton of these patients is favourable. The not yet generally approved indications for GH therapy are small chi
