Bipolar mood state reflected in cortico-amygdala resting state connectivity: A cohort and longitudinal study.

BACKGROUND: Using resting-state functional magnetic resonance imaging (rsfMRI), we previously compared cohorts of bipolar I subjects in a manic state to those in a euthymic state to identify mood state-specific patterns of cortico-amygdala connectivity. Our results suggested that mania is reflected in the disruption of emotion regulation circuits. We sought to replicate this finding in a group of subjects with bipolar disorder imaged longitudinally across states of mania and euthymia METHODS: We divided our subjects into three groups: 26 subjects imaged in a manic state, 21 subjects imaged in

The cortisol awakening response and anterior cingulate cortex function in maltreated depressed versus non-maltreated depressed youth.

Symptomatology of depression among children who have (vs. have not) experienced maltreatment is greater in severity, more resistant to conventional treatment, and associated with elevated risk for suicide. Recent evidence implicates perturbations in stress regulatory systems and heightened negative self-appraisals as factors that increase the severity of psychopathology experienced by depressed maltreated (vs. non-maltreated) youth. Likely explanatory mechanisms for these differences are disturbances in the function of the hypothalamic-pituitary axis (HPA) and persistent negative self-referent

Childhood maltreatment moderates the effect of combat exposure on cingulum structural integrity.

Limbic white matter pathways link emotion, cognition, and behavior and are potentially malleable to the influences of traumatic events throughout development. However, the impact of interactions between childhood and later life trauma on limbic white matter pathways has yet to be examined. Here, we examined whether childhood maltreatment moderated the effect of combat exposure on diffusion tensor imaging measures within a sample of military veterans (N = 28). We examined five limbic tracts of interest: two components of the cingulum (cingulum, cingulate gyrus, and cingulum hippocampus [CGH]),

Monitoring glutamate levels in the posterior cingulate cortex of thyroid dysfunction patients with TE-averaged PRESS at 3T.

BACKGROUNDS AND OBJECTIVES: Patients with thyroid dysfunction frequently have neuropsychiatric complaints such as lack of concentration, poor memory, depression, anxiety and mania, which suggest brain dysfunction. However, the underlying process of this dysfunction remains unclear. Recent studies of the glutamatergic system have offered important insight into the neuropsychiatric process. Thus, this study investigates changes in glutamate concentration in patients with thyroid dysfunction. It also clarifies whether Glu levels are related to thyroid hormones via proton magnetic resonance spectr

Effects of acute tryptophan depletion on raphé functional connectivity in depression.

Depression remains a great societal burden and a major treatment challenge. Most antidepressant medications target serotonergic raphé nuclei. Acute tryptophan depletion (ATD) modulates serotonin function. To better understand the raphé's role in mood networks, we studied raphé functional connectivity in depression. Fifteen depressed patients were treated with sertraline for 12 weeks and scanned during ATD and sham conditions. Based on our previous findings in a separate cohort, resting state MRI functional connectivity between raphé and other depression-related regions (ROIs) was analyzed in n

Greater functional connectivity between reading and error-detection regions following training with the reading acceleration program in children with reading difficulties.

The Reading Acceleration Program is a computerized program that improves reading and the activation of the error-detection mechanism in individuals with reading difficulty (RD) and typical readers (TRs). The current study aims to find the neural correlates for this effect in English-speaking 8-12-year-old children with RD and TRs using a functional connectivity analysis. Functional magnetic resonance imaging data were collected during a lexical decision task before and after 4 weeks of training with the program, together with reading and executive functions measures. Results indicated improvem

A structural MRI study of cortical thickness in depersonalisation disorder.

Depersonalisation disorder (DPD) is characterised by a sense of unreality about the self and the world. Research suggests altered autonomic responsivity and dysfunction in prefrontal and temporal lobe areas in this condition. We report the first structural magnetic resonance imaging study of 20 patients with DPD and 21 controls using the FreeSurfer analysis tool employing both region-of-interest and vertex-based methods. DPD patients showed significantly lower cortical thickness in the right middle temporal region according to both methods of analysis. The vertex-based method revealed addition

Tics are caused by alterations in prefrontal areas, thalamus and putamen, while changes in the cingulate gyrus reflect secondary compensatory mechanisms.

BACKGROUND: Despite strong evidence that the pathophysiology of Tourette syndrome (TS) involves structural and functional disturbances of the basal ganglia and cortical frontal areas, findings from in vivo imaging studies have provided conflicting results. In this study we used whole brain diffusion tensor imaging (DTI) to investigate the microstructural integrity of white matter pathways and brain tissue in 19 unmedicated, adult, male patients with TS "only" (without comorbid psychiatric disorders) and 20 age- and sex-matched control subjects. RESULTS: Compared to normal controls, TS patient

Individual differences in brain structure and resting-state functional connectivity associated with type A behavior pattern.

Type A behavior pattern (TABP) is characterized by competitiveness and hostility, time urgency and impatience. These traits can have a significant impact on physical and mental health. We have not found studies focusing on brain structure or functional connectivity correlates associated with individual differences in TABP. The present study used voxel-based morphometry (VBM) and resting-state functional connectivity (rsFC) analysis to examine the neural correlates of TABP. The results showed that TABP was positively correlated with regional gray matter volume (rGMV) in the left subgenual anter

Thalamic abnormalities are a cardinal feature of alcohol-related brain dysfunction.

Two brain networks are particularly affected by the harmful effect of chronic and excessive alcohol consumption: the circuit of Papez and the frontocerebellar circuit, in both of which the thalamus plays a key role. Shrinkage of the thalamus is more severe in alcoholics with Korsakoff's syndrome (KS) than in those without neurological complication (AL). In accordance with the gradient effect of thalamic abnormalities between AL and KS, the pattern of brain dysfunction in the Papez's circuit results in anterograde amnesia in KS and only mild-to-moderate episodic memory disorders in AL. On the o

Assembling old tricks for new tasks: a neural model of instructional learning and control.

We can learn from the wisdom of others to maximize success. However, it is unclear how humans take advice to flexibly adapt behavior. On the basis of data from neuroanatomy, neurophysiology, and neuroimaging, a biologically plausible model is developed to illustrate the neural mechanisms of learning from instructions. The model consists of two complementary learning pathways. The slow-learning parietal pathway carries out simple or habitual stimulus-response (S-R) mappings, whereas the fast-learning hippocampal pathway implements novel S-R rules. Specifically, the hippocampus can rapidly encod

Abnormal functional connectivity of the default mode network in remitted late-onset depression.

BACKGROUND: The functional neural network model has been a major method used to investigate mechanisms of neuropsychopathy. There is considerable evidence that late-onset depression (LOD) is the prodrome, or the early clinical manifestation, of Alzheimer's disease (AD). The default mode network (DMN) is one of the neural networks that can be used to explore the complex relationships between depressive symptoms, episodic memory deficits and other cognitive impairments. To date, no study has directly linked the DMN to LOD while focusing on episodic memory and the influence of apolipoprotein E4 (

Frontocingular dysfunction in bulimia nervosa when confronted with disease-specific stimuli.

OBJECTIVE: Bulimia nervosa (BN) is characterized by dysregulation of impulse control, in other words, uncontrolled eating. Functional neuroimaging studies have been sparse and have used variable methodologies. METHOD: Thirteen medication-free female BN patients and 13 female healthy controls were investigated by functional magnetic resonance imaging using a disease-specific food paradigm. Stimuli were rated after the scanning procedure. RESULTS: Bulimia nervosa patients showed increased fear ratings and a trend for increased disgust. Magnetic resonance imaging data of 10 BN patients could be

Cerebral phaeohyphomycosis due to Cladophialophora bantiana in a Huacaya alpaca (Vicugna pacos).

Cerebral phaeohyphomycosis, caused by Cladophialophora bantiana, was diagnosed in a Huacaya alpaca (Vicugna pacos). An 8-year-old, intact male Huacaya alpaca from a farm in Indiana was found dead at pasture and submitted to the Animal Disease Diagnostic Laboratory at Purdue University for necropsy examination. Gross lesions were limited to the cerebrum, which had a 2×2.5×5 cm, well-demarcated, firm, mottled, pale grey to brown-red mass with a granular texture, centered on the left and right cingulate gyri. Microscopically, granulomatous and necrotizing meningoencephalitis with intralesional pi

Quantitative electroencephalography for tinnitus--a means for data collection, analysis, and translation.

Quantitative electroencephalography (QEEG) is the technique whereby brain electrical activity in individuals is recorded as they sit quietly with their eyes closed. The electrical activity is quantified with a variety of statistical measures to characterize the huge variation in combinations of emissions from the brain. Neuroscience research has demonstrated that such resting brain activity measures may be consistently altered in conditions such as depression or dementia. A wide variety of ongoing efforts are attempting to find characterizations that reliably denote other neurological conditio

Coping with emotions past: the neural bases of regulating affect associated with negative autobiographical memories.

BACKGROUND: Although the ability to adaptively reflect on negative autobiographical experiences without ruminating is critical to mental health, to our knowledge no research has directly examined the neural systems underlying this process. METHODS: Sixteen participants were scanned using functional magnetic resonance imaging (fMRI) as they focused on negative autobiographical memories using cognitive strategies designed to facilitate (feel strategy) versus undermine (analyze and accept strategies) rumination. RESULTS: Two key findings were obtained. First, consistent with prior emotion regul

Blocking central opiate function modulates hedonic impact and anterior cingulate response to rewards and losses.

Reward processing is linked to specific neuromodulatory systems with a dopaminergic contribution to reward learning and motivational drive being well established. Neuromodulatory influences on hedonic responses to actual receipt of reward, or punishment, referred to as experienced utility are less well characterized, although a link to the endogenous opioid system is suggested. Here, in a combined functional magnetic resonance imaging-psychopharmacological investigation, we used naloxone to block central opioid function while subjects performed a gambling task associated with rewards and losse

Stress-induced synaptic changes in the rat anterior cingulate cortex are dependent on endocrine developmental time windows.

Fetal and neonatal brain development is characterized by developmental time windows during which brain regions or neuron types are specifically sensitive to environmental influences. Previous studies on cortical development have revealed evidence for the hypothesis that the extent and the direction of experience-induced neuronal and synaptic changes correlate with time windows of endocrine development. To further test this hypothesis we exposed rats to neonatal separation stress during different phases of endocrine maturation, i.e. prior, during and after the stress hyporesponsive period (SHRP
