Multiple representations of the body and input-output relationships in the agranular and granular cortex of the chronic awake guinea pig.

The organization of somatosensory input and the input-output relationships in regions of the agranular frontal cortex (AGr) and granular parietal cortex (Gr) were examined in the chronic awake guinea pig, using the combined technique of single-unit recording and intracortical microstimulation (ICMS). AGr, which was cytoarchitectonically subdivided into medial (AGrm) and lateral (AGrl) parts, also can be characterized on a functional basis. AGrl contains the head, forelimb, and most hindlimb representations; only a small number of hindlimb neurons are confined in AGrm. Different distributions o…

Liposome-iRGD loaded with astragaloside IV alleviates the functional damage of endothelial progenitor cells via the Hippo-YAP/TAZ signaling pathway.

The characteristics of diabetic vascular complications are impaired angiogenesis, which leads to hindlimb ischemia. Although astragaloside IV (AS-IV) can promote angiogenesis, its poor targeting to endothelial progenitor cells (EPCs) limits its therapeutic effect. Here, we developed liposomes modified with iRGD to load AS-IV (Lp-iRGD@AS-IV) to enhance its delivery and explored its mechanism. Lp-iRGD@AS-IV and fluorescently labeled liposomes were prepared, and their phenotypic characteristics were detected. The study results showed that the uptake efficiency of Lp-iRGD@AS-IV by EPCs was higher …

Effect of endotoxin on systemic and skeletal muscle O2 extraction.

Patients with the adult respiratory distress syndrome (ARDS) show a pathological dependence of O2 consumption (VO2) on O2 delivery (QO2, blood flow X arterial O2 content). In these patients, a defect in tissues' ability to extract O2 from blood can leave tissue O2 needs unmet, even at a normal QO2. Endotoxin administration produces a similar state in dogs, and we used this model to study mechanisms that may contribute to human pathology. We measured systemic and hindlimb VO2 and QO2 while reducing cardiac output by blood withdrawal. At the onset of supply dependence, the systemic QO2 was 11.4 …

Effects of ageing and chronic dietary restriction on the morphology of fast and slow muscles of the rat.

The soleus and extensor digitorum longus muscles of the hindlimb and the flexor digitorum profundus muscle of the forelimb were studied in ad libitum-fed control and age-matched diet-restricted male rats at various ages from weaning to senescence. Growth of individual muscles was accomplished by fibre hypertrophy and not hyperplasia. Between weaning and one year, fibre numbers remained constant in the soleus but fell by 50% in the extensor digitorum longus. Both muscles displayed increasingly oxidative fibre type profiles with advancing age, irrespective of dietary status. This was particularl…

Mustn1 is a smooth muscle cell-secreted microprotein that modulates skeletal muscle extracellular matrix composition.

OBJECTIVE: Skeletal muscle plasticity and remodeling are critical for adapting tissue function to use, disuse, and regeneration. The aim of this study was to identify genes and molecular pathways that regulate the transition from atrophy to compensatory hypertrophy or recovery from injury. Here, we have used a mouse model of hindlimb unloading and reloading, which causes skeletal muscle atrophy, and compensatory regeneration and hypertrophy, respectively. METHODS: We analyzed mouse skeletal muscle at the transition from hindlimb unloading to reloading for changes in transcriptome and extracel…

[Closed-loop regulation system for paralyzed lower limb joint movement based on electrical stimulation of spinal central pattern generator].

Intraspinal microstimulation (ISMS) is a rehabilitation technology that activates muscle movement by electrically stimulating the spinal cord, thereby restoring the function of paralyzed limbs. In this study, a fuzzy logic-controlled self-tuning proportional-integral-derivative (PID) algorithm was adopted. By simultaneously adjusting three key electrical stimulation parameters-amplitude, pulse width, and frequency of the pulse signal-the distal locomotor central pattern generator (CPG) in rats with spinal cord injury (SCI) was activated, realizing real-time control of hindlimb ankle joint move…

Combination of kinematic analyses and diffusion tensor tractrography to evaluate the residual motor functions in spinal cord-hemisected monkeys.

BACKGROUND: Spinal cord injury (SCI) causes loss of locomotor functions. Nowadays, the relationship between the residual locomotion after SCI and the diffusion tensor tractography (DTT) results still remains unclear. METHODS: Four rhesus monkeys were suffered thoracic cord hemisection. Kinematic evaluation and DTT were performed prior- and post-SCI (6 and 12 weeks). The longitudinal changes of gait parameters and the DTT parameters were analyzed for the injury-contralateral hindlimb. The correlations between gaits and DTT parameters were also investigated. RESULTS: Almost gait parameters sig…

Acute and delayed transplantation of olfactory ensheathing cells promote partial recovery after complete transection of the spinal cord.

The present study was undertaken to determine whether olfactory ensheathing cells (OECs) from the olfactory bulb were capable to promote axonal regeneration and functional recovery when transplanted either acutely or 1 week delayed into the T8 transected rat spinal cord. OEC transplants increased recovery of functional outcomes, as shown electrophysiologically by return of motor evoked potentials and by reduction of hindlimb hyperreflexia, and behaviorally by recovery of movements of hindlimb joints. Axonal regeneration was proven histologically by demonstrating long axonal outgrowth of raphes…

Suspected botulism in dairy cows and its implications for the safety of human food.

A large outbreak of suspected botulism occurred on a dairy farm. The affected animals were listless and showed signs ranging from hindlimb unsteadiness to lateral recumbency, although the most common presentation was sternal recumbency with an apparent hindlimb weakness when stimulated to rise. Postmortem examinations revealed no conclusive gross pathology or histopathology. The affected cattle were found to have neutrophilia and hyperglycaemia with no other consistent haematological or biochemical abnormalities. The combination of clinical signs, disease epidemiology and the ruling out of oth…

Pharmacological inhibition of TLR4 ameliorates muscle and liver ceramide content after disuse in previously physically active mice.

Toll-like receptor 4 (TLR4) is a proposed mediator of ceramide accumulation, muscle atrophy, and insulin resistance in skeletal muscle. It is currently unknown whether pharmacological inhibition of TLR4, using the TLR4-specific inhibitor TAK-242 during muscle disuse, is able to prevent changes in intracellular ceramide species and consequently preserve muscle size and insulin sensitivity in physically active mice. To address this question, we subjected running wheel-conditioned C57BL/6 male mice (13 wk old; ∼10/group) to 7 days of hindlimb suspension (HS), 7 days of continued wheel running (WR…
